Verfahren zur Herstellung von am Stickstoff substituierten Thiosäureamiden
bzw. von Thiazolen In manchen heterocyclischen Verbindungen besitzen Methylgruppen
eine große Reaktionsfähigkeit. Sie lassen sich unter anderem sehr leicht mit elementarem
Schwefel und aromatischen primären oder sekundären Aminen zu am Stickstoff substituierten
Thiosäureamiden umsetzen, die sich bei höherer Temperatur zu Thiazolen cycldsiere.n

bis z13).Process for the preparation of thioamides or thiazoles substituted on the nitrogen. In some heterocyclic compounds, methyl groups are highly reactive. Es wurde nun gefunden, daß es vorteilhaft ist, bei dieser Umsetzung
die Amine in Form ihrer Salze, insbesondere der Salze mit Mineralsäuren, zu verwenden.
Dies ist deshalb wichtig, weil es ermöglicht, auch niedere, leicht flüchtige aliphatische
Amine in Form ihrer Salze ohne Verwendung von Druckgefäßen in einfacher Reaktion
mit heterocyclischen Methylverbind'ungen und Schwefel zu substituierten Thiosäureamiden
umzusetzen. So gelingt es z. B., aus Methylaminhydrochlorid, Schwefel und a-Picolin
durch mehrstündiges Erhitzen auf etwa 1q.0° das Thiopicol.insäuremethylamid herzustellen.
Verbindungen dieser Art waren nach dem obengenannten bekannten Verfahren nicht zugänglich.
Die Reaktionstemperaturen liegen zwischen
ioo und 2oo°. Die Reaktion
ist beendet, wenn sich kein Schwefelwasserstoff mehr abspaltet. Auch aromatische
Amine können in Form ihrer Salze mit Schwefel und. reaktionsfähigen heterocyclischen
Methylverbindungen umgesetzt werden.It has now been found to be advantageous in this implementation

Bei Verwendung von Salzen aromatischer Amine, z. B. Anilinhydrochlorid,
erhält man (im Gegensatz zu dem Verfahren von E m me r t und Groll, Chem. Ber.,
Bd.86, 1953, S.2o8ff.) fast ausschließlich die Thiazole. Zum 'Beispiel erhält man
aus y-Picolin, Schwefel und Anilinhydrochlorid nur das 2-(y-Pyri:dyl)-benzothiazol.
Beispiel i 186 .g (2 Mol) a-Picolin, 192 g (6, Mol) Schwefel und 134 g (2 Mol) Methylaminhydrochlorid
werden etwa 5 Stunden im Ölbad auf 17o° unter Rühren erhitzt, wobei heftig Schwefelwasserstoff
entwickelt wird. Danach. kühlt man auf Zimmertemperatur, versetzt zweimal mit i5%-iger
Salzsäure, schüttelt gut durch und filtriert vom restlichen Schwefel ab. Die saure
Lösung des Produktes wird nun mit Natronlauge neutralisiert, wobei sich das Thiopi,col:insäuremethylamid
im Gemisch mit a-Picolin als Öl abscheidet; dieses wird im Scheidetrichter abgetrennt.
Die wäßrige Schicht wird -mit Chloroform ausgeschüttelt und die Chloroformlösung
mit dem abgetrennten Öl vereinigt. Die Lösung wird nun mit Natriumswlfat getrocknet,
dann .das Chloroform abdestilliert und anschließend das a-Picolin. Das zurückbleibende
Öl wird im Vakuum destilliert und erstarrt in der Vorlage zu gelben Kristallen.
Nach Umkristallisation aus Methanol Fp.75 bis 78°. Die Ausbeute beträgt 52g (= 170/0),
bezogen auf a-Pricolin. Beispiel 2 186 g (2 Mol) a-Picolin, 192 g (6 Mol) Schwefel
und 162 g (2 Mol) Dimethylaminhydrochlorid werden unter Rühren etwa 5 Stunden in
einem Ölhai auf etwa 17o° erhitzt, wobei sehr stark Schwefelwasserstoff entwickelt
wird. Das Reaktionsgemisch wird aufgearbeitet wie im Beispiel i. Das zurückbleibende
Öl, Thiopicol:insäuredimethylamid, destilliert bei i5-mm Druck und 184 bis 188°.
Es kristallisiert nicht. Die Ausbeute beträgt 89 g (= 29%), bezogen auf das a-Picolin.
Beispiel 3 186 g (2 Mol) y-Picolin, i92 g (6 Mol) Schwefel und 1349 (2 Mol) Methylaminhydrochlorid
werden im Ölbad bei 17o° unter Rühren 7 Stunden erhitzt, und das entstandene Reaktionsgemiisch
wird wie im Beispiel i aufgearbeitet. Das zurückbleibende Öl wird mehrmals im Hochvakuum
destilliert. Kp.i,l 16o bis 164°. Das gelbe Thioisonicotinsäuremethylami,d erstarrt
beim Abkühlen. Nach Umkristallisation ,aus Methanol Fp. 94 bis 97°. Die Ausbeute
beträgt 4g g (= 15%), bezogen auf y-Picolin.When using salts of aromatic amines, e.g. B. aniline hydrochloride,

und 162 g (2 Mol) Ddmethylaminhydrochlorid werden unter Rühren im Ölbad von 17o°
6. Stunden lang erhitzt, und das Reaktionsgemisch wird wie im Beispiel i aufgearbeitet.
Das zurückbleibende Öl, Thiois.onicotinsäuredimethyl,amid, wird mehrmals im Hochvakuum
destilliert. Kp.0,$ 135 bis 137°. Es kristallisiert beim Abkühlen. Fp. 64 bis 65°.
Die Ausbeute beträgt 96 g (= 29°/o), bezogen auf y-Picolin. B.eiispiel 5 i8,8
g Anilinhydrochlorid, Zoo g y-Picolin und i4og Schwefel- werden im Ölbad
unter Rühren 24 Stunden auf 16o° erhitzt. Danach wird gekühlt, mit Natronlauge schwach
alkalisch gemacht und das restliche Picolin und Anilin mit Wasserdampf abdestilliert.
Das hinterbleibende Öl wird zweimal mit 3 n-Natronlauge ausgeschüttelt. Aus dem
alkalischen Filtrat fällt beim Neutralisieren kein Thioisonicotinsäureanili:d aus.
Der in Alkali unlösliche Rückstand, welcher -das 2-(y-Pyridyl)-benzothiazol enthält,
wird mehrmals aus Alkohol nach Anwendung von Kohle umkristallisiert. Farblose Blättchen
vom Fp. 122 bis 125'; löslich in Salzsäure, unlöslich in Natronlauge, nicht oxvdierbar.
Die Ausbeute beträgt 85 g (= 270/0), bezogen auf y-Picolin. B,ei spiel 6 149 g (i
Mol) 2-Methylbenzthiazol, 129 g (i Mol) -Anilinhydrochlorid und 96 g (3 Mol) Schwefel
werden im Ölbad auf 2oo° erhitzt und 36 Stunden auf dieser Temperatur gehalten.
Danach wird mit 3 n-Natronlauge zweimal ausgeschüttelt. Beim Neutralisieren fällt
aus der alkalischen Lösung kein Benzthiazol-.2-thiocarbonsäureanilid aus. Der alkaliunlösliche
Rückstand, Dibenzoth.iazolyl, wird zweimal aus Dimethylformamid nach Anwendung von
Kohl- umkristallisiert. Farblose Blättchen vom Fp. 312°; schwer löslich in Salzsäure,
unlöslich in Natronlauge, nicht oxydierbar. Die.Ausbeute beträgt 25 g (= io%), bezogen
