Process for the preparation of new therapeutically active salts In the German patent 875 202 diphenylsulfone derivatives of the formula are in soft Hal is a halogen and R is hydrogen, an alkyl or aryl radical, described as intermediates.
In the further processing of this group of substances it was found that - they have a very good bacteriostatic effect on the body surface; in the body itself, i.e. applied orally or parenterally, they are little or not at all active. They are therefore particularly applicable for topical use, for example in the case of otitis and infectious diseases of the eye, and here also achieve the well-known 4-amino-q'-carboxymethylaminodiphenylsulfone in terms of activity.
bedeuten. .The alkali, alkaline earth and pyridine salts of q.-halo-q.-carboxy-alkylaminodiphenylsulfones described in the German patent document $ 75 2o2 are in practice because of their strong strengths. Alkalinity or high toxicity (pyridine) cannot be used chemotherapeutically. The main patent 951 985 relates to a process for the preparation of new chemotherapeutically effective salts of the formula in which R is a straight or branched-chain alkylene radical, R is hydrogen, an alkyl or alkanol radical, R2 or R3 is hydrogen or R2 and R3 are low molecular weight alkanol radicals or alkylene radicals linked to form a ring by means of an oxygen atom. .
The salts described there show opposite those in the German
The salts described in patent specification 875 2o2 have the advantage that they are particularly light
crystallize and have a good cleaning effect for the corresponding, underlying them
have lying acids.
bedeutet, ebenfalls wertvolle Eigenschaften zeigen.
In the further pursuit of this invention, the observation has now been made that salts of the formula in which R, R1, R2 and R3 have the meaning given above and X is a halogen atom, also show valuable properties. For example, they are readily soluble in water with a practically neutral reaction and are capable of precipitating the acids on which they are based from organic solvents in one operation. A comparison of the cleaning effects of sodium, calcium and morpholine salt precipitation of 4-chloro-4'-carboxy-methylaminodiphenyl sulfone has shown that the morpholine salt is obtained in a significantly higher yield and the acid released from it is purer and better tolerated than that via the sodium - or calcium salts isolated acid.
Ohres, z. B. bei eitrigen Otitiden, verwendet werden. Thanks to their neutral reaction, the new salts can also be used for medicinal purposes
Purposes, e.g. B. with infections of the eye, in particular - but with infections of the
Ear, e.g. B. in purulent otitis, can be used.
The new salts are obtained in a manner known per se by reacting an acid of the formula with a base of the formula in which X, R1, R2 and R3 have the meaning given above, in the presence or absence of a solvent or diluent. The salts obtained can be isolated and, if necessary, purified by recrystallization or the solutions obtained by reacting the two components in water can be used directly. Example 32.5 g of 4-chloro-4'-carboxymethylaminodiphenylsulfone are slurried in 100 cc of a mixture of equal parts of pure methanol and acetone, and 9 g of morpholine are added. Boil briefly, filter with a little charcoal and allow the filtrate to cool. 32 g of the morpholine salt melting at 189 to igi ° are obtained. This can be recrystallized from methanol.
