Verfahren zur Herstellung von Salicyl-2-glucopyranose Beobachtungen
von Fermi (Arch. Hyg. 63, 315 [=9o7]), Marie und Urbain (C. R. Seances @Soc. Biol.
Filial. Assoc. 104, 1207 [=93o1) veranlaßten, die Salicylsäure zur Bekämpfung der
Lyssa heranzuziehen. Da Salicylsäure wegen ihrer Reiz- und Ätzwirkung nicht injiziert
werden kann (Hiller, Dtsch. Arch. klin. Med. 16, 62o [I875]) und ihr Natriumsalz
in hohen Dosen, die unverträglich sind (Hanzlik, de Eds, Tainter, Arch. internal
Med. 36, 5oo [I925]; Heymann, Therapie d.Gegenw. 66,66 [I9251_; McGuigan,
Higgins, Arch. int. Pharmacodyn. Therap. 51, 398 [I935]) , infundiert werden müßte,
zielt das den Gegenstand vorliegender Erfindung bildende Verfahren darauf ab, Salicylsäure
mit d-Glucose zu lyssicider, cardicalund coronargefäßerweiterndwirkender Salicyl-2-glucopyranose
zu verbinden. Die an die Salicylsäure gekoppelte d-Glucose spielt für den in Rede
stehenden therapeutischen Effekt die Rolle einer Gleitschiene, welche die lipoidlösliche
Salicyl-2-glucopyranose den Vorderhörnern und dem Hirn als dem Sitz des Lyssavirus
zuführt. Die hohe lyssicide Aktivität der Salicyl-2-glucopyranose wird dadurch gekennzeichnet,
daß durch 4,995 mg/6o kg Mensch, i. m. injiziert, das excitative Krankheitsbild
der Lyssa abklingt und frei von Paranoien, Paraphrenien, Fieber und Lähmungen flüssige
und breiige Nahrung wieder aufgenommen werden kann und sich die Lebensdauer um 552

Wie Versuche gezeigt haben, läßt sich Salicylsäure mit d-Glucose derart
umsetzen, daß nicht ihre Carboxylgruppe, sondern ihr orthoständiges Hydroxyl mit
dem alphaständigen Hydroxyl der d-Glucose unter Wasseraustritt reagiert. Zu diesem
Zweck schmilzt man äquimolare Mengen d-Glucose und Salicylsäure zusammen, kocht
die in der Kälte erstarrte Schmelze mit Wasser bis zur klaren Lösung aus und fällt
das Produkt durch Erkaltenlassen. Ausführungsbeispiel 18 g d-Glucose und 13,8 g
Sahcylsäure, trocken und innig miteinander vermischt, erwärmt man in einem ioo ccm
fassenden Rundkolben vorsichtig unter Vermeidung von Karamelisierung bis zur deutlich
klaren Schmelze. Da die d-Glucose bei i46° und die Salicylsäure bei 155 bis i56°
schmilzt, kommt es dabei zuerst zum Schmelzen der d-Glucose, die dann in ihrem geschmolzenen
Zustand der Salicylsäure gegenüber als Lösungsmittel wirkt. Während dieses Prozesses
ist der Rundkolben entsprechend so verschlössen, daß dem gelegentlich zunehmenden
Innendruck noch Austritt gewährt bleibt. Dann läßt man die Schmelze etwa 15 Minuten
lang etwas abkühlen, wobei dieselbe zu einer weißen amorphen Masse erstarrt. Diese
erstarrte Masse kocht man dreimal mit je ioo ccm Wasser aus, und zwar so lange,
bis sich die ganze Masse im kochenden Wasser klar gelöst hat. Die insgesamt
300 ccm heiße Lösung gießt man in eine etwa 375 ccm fassende Porzellankasserolle
und läßt durch Einstellen in ein kaltes Wasserbad erkalten. Dabei entsteht eine

dem selbständigen Erkalten, wobei die Salicyl-2-glucopyranose sich in Gestalt fahlgelblichweiß
glitzernder, kurz ausgebildeter, würzig süß und zugleich anästhesierend schmeckender
Kristallnädelchen schön abscheidet, welche unter Zersetzungserscheinungen ohne Toleranz

das heiß gemacht wird, mit 25 mg Salicyl'2-glucopyranose versetzt wird, tritt eine
klare Lösung ein, die bestehenbleibt, wenn man sie selbständig langsam erkalten
läßt.
