Description
Process for the production of sodium-calcium-copper keratin compounds To 4 German Patent 710 308 are sodium-calcium-gold keratin compounds obtained by using aqueous solutions of the calcium compounds of keratin breakdown products gold halide or gold halide hydrochloric acid added in a known manner and now the acidic reaction mixtures are neutralized or weakly with sodium hydroxide solution makes alkaline and then from the reaction mixture the gold compounds with organic Solvents such as B. alcohol fails.
It has now been found that sodium-calcium-copper keratin compounds can also be used can win if you use the aqueous solution of the keratin breakdown products at the same time neutralized with caustic soda and calcium hydroxide in a certain proportion Makes it slightly alkaline and then in a known manner by adding formaldehyde sulfoxylic acid Sodium or sodium hydrosulfite which reduces keratin breakdown products, copper salts, such as B. copper sulfate, until the sulfhydryl groups are saturated and added off the sodium-calcium-copper keratin compounds in the reaction mixture at weak alkaline reaction by stirring multiple amounts of the resulting solution Alcohol fails.
Surprisingly, sodium-calcium-copper keratin compounds are always used here of even 13 working. Come to use in well-known Way by acid hydrolysis until the more or less complete solution and subsequent Dialysis produced, even higher molecular, sulfur-rich keratin hydrolysates. The reduction of the keratin hydrolyzate is as described in patent specification 662 648 and as already mentioned, with sodium formaldehyde sulfoxylic acid or sodium hydrosulfite carried out. The copper compound mentioned there is a sodium-copper keratin compound. In the patent specification 662 647 a sodium copper keratin compound is described, Assumed a non-reduced keratin hydrolyzate for each production will. Also in the case of the copper compound in patent specification 694 1.00 e £ vtäbnten non-reduced keratin breakdown products implemented with copper salts. The received Copper compounds are calcium copper keratin compounds.
Compared to the sodium copper keratin compounds, those according to the present Invention obtained sodium-calcium-copper keratin compounds the advantage that they are obtained immediately in solid form when precipitated from the aqueous solution with alcohol and do not tend to smear, furthermore that less alcohol is required for their precipitation is. Compared to the calcium copper keratin compounds, they have the advantage of that they are not so bulky and therefore separate significantly better and leave to wash out as these, so that they can be used immediately for therapeutic purposes are usable and a reprecipitation, which always entails loss of substance, is not is required. The obtained sodium-calcium-copper keratin compounds contain 4.0 to 11.0 per cent. Copper and is very much in water with an almost neutral reaction easily soluble. They are well tolerated by intramuscular and subcutaneous injection, also do not cause any local tissue damage and show in particular at rheumatic diseases such as B. Monarthritilden and osteoarthritis, an excellent one therapeutic effect.
Example I 500 g of keratin hydrolyzate produced by hydrolysis of hair with four times the amount of 20 to 25% hydrochloric acid until just about the solution, then Dialysis and evaporation on the water bath until dryness has been obtained dissolved in 2.51 of water and mixed with 35 ccm of 3o0 / o sodium hydroxide solution and about 30 to 35 g Ca (OH) 2 added to a weakly alkaline reaction. The solution becomes 120 g sodium formaldehyde sulfoxylic acid and placed in a well-closed vessel Allowed to stand for some time at room temperature until the maximum reduction in SS groups is achieved what by titration with silver nitrate until it disappears the sodium nitroprusside reaction can be determined using the spot test. An aqueous solution of copper sulfate is then added in portions until it disappears stirred into the sodium nitroprusside reaction. There are about 65 g of copper sulfate consumed. If the solution obtained in this way reacts slightly acidic, it is treated with Io ° / oilger Sodium hydroxide solution made weakly alkaline, filtered and the clear filtrate in the multiple Amount of alcohol stirred in. The sodium-calcium-copper keratin connection falls in the process in the form of flakes that settle immediately. The yellowish product thus obtained - Is sucked off, washed thoroughly with alcohol and dried in a vacuum.
Example 2 For 500 g of keratin hydrolyzate obtained according to Example I. and was reduced, only that instead of 120 g of sodium formaldehyde sulfoxylate 100 g of sodium hydrosulphite were used, a suspension of freshly felled Cuprooxide is added in small portions while stirring vigorously until the nitroprusside / sodium reaction has just become negative and then proceed as in Example 1.
