Description
Method of preparing tissue for microscopic examination the The invention relates to a method for preparing histological tissue for microscopic Examinations The tissue preparation for the purpose of microscopic examination requires a series of treatments of the tissue before cutting the slices from the piece of tissue for staining and for) wrestling the sections on the microscope supports. In particular, when preparing tissue, it is necessary to remove the tissue pieces to submerge one after the other in a series of liquid funds for certain periods of time, namely to fix the tissue. to wash out the fixative. for draining of the tissue, which is usually obtained by immersing it one after the other in alcohols or other dehydrating agents then used to treat the fabric with a Clarifying agent and then finally to incorporate an embedding agent in the trade, such as B. paraffin or collodion. After the fabric is treated like that it is cut into slices of the desired thickness.
The paraffin or another embedding agent is then made from these Slices or cuts are removed in the usual way by a solvent, after which the sections are then colored and placed on carriers.
As a result of the numerous treatments, which as indicated above. the tissue is subjected, and due to the relatively long time which. is required for each treatment with the various means, or in accordance with the so far from the pathologists and other professionals which deal with microscopic examination of tissue, related methods requires the preparation of tissue, especially up to and during the stage in which paraffin or another corresponding agent is incorporated into the tissue, a great span of time.
For example, the time required for the fixing treatment has been heretofore alone, if Zenker's fluid should be used as a fixative, more than 2 days.
A method for tissue treatment has now been found in which the time required to prepare the tissue for microscopic examination can be reduced considerably. In more detail it was found that, when the tissue is immersed in the liquid agent and slowly while in the same of immersion is used to treat the tissue in each stage in the course the preparation time for microscopic examination is considerably reduced and that better cuts are obtained.
It becomes natural for pathologists and other professionals in the field it will be readily apparent that the reduction in tissue dissection time required is a very significant advantage because, inter alia, possible is to prepare sections for use in a relatively short time in cases where immediate, tissue examination is an important factor is, and continues, because more tissue pieces in a given period of time with the same Devices and apparatus can be prepared than was previously possible.
The last mentioned advantage is especially for clinics or others Noteworthy laboratories in which a large number of tissue parts are dissected and needs to be investigated.
In particular, it is necessary to carry out the method of the present invention Note the following: The tissue which is prepared for microscopic examination shall be. may have one of the common forms, e.g. B. such forms, which are customary when performing an autopsy, curettage or biopsy. That Tissue becomes of the desired shape when it is held with a liquid for fixation is treated, in a suitable liquid agent, according to the preferred Working method of the pathologist, quickly immersed, e.g. B. in formalin or fixative Handlebars, Bouin, Halle and Vande-Grift.
The tissue is held in Be weighed in the fixing liquid, and preferably by gently rotating the piece of tissue in this liquid. The tissue is placed in the liquid so that it is completely immersed therein and that its area is of greatest extent in the vertical position or approximately is in a vertical position in the container with the liquid. The container or Holder in which the piece of tissue is guided in this vertical position rotates around a vertical axis within the liquid agent in its vessel, and at a low speed, preferably one of two revolutions per minute comes into consideration.
The speed of rotation can vary from this preferred speed differ. should however be low enough to have a substantial loading. movement of the Body of liquid while the tissue in it rotates. In general the speed is in the appropriate range of one to five revolutions per minute, although the present method is not accurate at this speed Area is limited.
However, the rotation speed should be used for the best results of the tissue in the liquid are kept sufficiently low. an essential one Movement of the liquid. in which the tissue is introduced to avoid.
The above procedure described in relation to the fixation is repeated in each of the following stages of tissue preparation, i.e. H. at the Washing the tissue after fixation, when dehydrating the tissue by immersion the same one after the other in alcohol, dioxane, acetone and other dehydrating agents, when the tissue is healed by immersion in chloroform, gasoline, xylene, cedarwood oil, Aniline and the like, and also when embedding the drained tissue by immersion in liquid paraffin or collodion.
The length of time during which the tissue is in the various liquid Treatment agent immersed can vary and is usually dependent of the particular methods used by the various pathologists become, different. It is well known that pathologists follow those specially trained professionals are their own individual ways of preparing tissue with the rivet testicles.
In general, it can be stated that the immersion time if the tissue in the liquid as described above, in accordance with the present Invention is moved to at least 500 / o can be reduced and that the Fixation when using Zenker's liquid, which is usually over Requires 2 days of time, according to the present invention in a period of 15 Minutes up to 4 hours depending on the type of (esxebes the shape of the pieces and the special working method of the pathologist, under whose direction the tissue preparation is carried out, can be done.
If thus the time required for tissue preparation after the present l.rlmdullg is much shorter than the one required earlier. so it follows on the other hand the exact length of time the piece of tissue is immersed in the various liquids Means depending on the wishes of the respective pathologist or specialist, under whose direction the preparation takes place.
After the tissue. how obell is described, prepared, is cut it, as before, into slices, whereupon the cuts for the purpose of distance the paraffin and can be treated for coloring. The cuts can be made if so desired is, in the paraffin removal liquid and in the staining solution essentially in the same way as is the case with the treatment of the tissue up to the level of embedding is described, rotated.
EXAMPLE 1 1. There are pieces of human tissue in a Thickness of about 2 to 3 mm prepared, 2, The pieces of tissue are in Zenkersche Liquid (potassium dichromate 2.5 g, Suhlimate 8 g, distilled water 100 ccm) brought and fixed for 5 hours. Small pieces, such as the endrometrium and skin biopsies, are fixed in even shorter times.
The tissue pieces are removed from the Zenker's fluid and washed in water for 2 hours in a container from which the water is continuously withdrawn and replaced with fresh water. One Siphon-like washing device is used for this purpose.
