Description

Process for the extraction of antibiotic substances from algae Algae occur on trees, especially beeches, in fresh water as a greenish slimy coating and in sea water. It was previously known that the algae contain slimy substances which are used to obtain the well-known agar-agar. It was not known that they have other ingredients with a particular physiological effect. According to the investigations by Pratt (Chem. Zbl. 1947 11, 815), artificially grown algae have ingredients that have an antibiotic effect. According to the investigations of Klosa (Ztschrft. F. Naturforsch. [4b] 187 [1949,]) green algae that occur on beeches and freshwater algae have an antibiotic effect if the algae are subjected to a weakly alkaline pretreatment and then extracted with alcohol . The antibiotics obtained in this way can be used therapeutically.

Beispiele r. Examples r. Algae are collected on beech trunks and are common sufficiently dry; if not, they will be placed on glass plates dried. The antibiotic content is increased when drying takes place in sunlight. Then the algae are powdered well in the mortar and with extracted hot alcohol for 5 hours. The alcohol is evaporated, the residue with Water is added, an oily emulsion sometimes settling out. One acidifies and extracted with ether. The ether is evaporated and the residue with Amyl acetate added. An amorphous precipitate separates out, which is filtered off will. The filtrate is evaporated and the residue is taken up with ether. Of the Ether, in which the residue is dissolved, is now mixed with 2N soda solution, Separated the layers in a separating funnel and acidified the soda solution with hydrochloric acid. A partly crystalline, partly amorphous powder precipitates out and is filtered off and re-extracted with ether. After evaporation of the ether one gets one Body that melts at 145 to 150 ° C and in alcoholic solution with ferric chloride shows a wine-red color. The substance is soluble in alkalis and in a dilution from i: i ooo ooo effective against streptococci and staphylococci.

Production of a crude extract. 20 g of algae (dried) Extracted for 5 hours with 500 cc alcohol, the alcohol evaporated, the residue poured into water, acidified and extracted with ether. After evaporation of the ether an amorphous powder is obtained that has a lower antibiotic effect than the substance according to example i.

Man extrahiert mit Äther und erhält nach Verdampfen des Äthers ein amorphes Pulver, welches nach dem Verfahren des Beispiels i gereinigt werden kann.3. 20 g of algae (dried) are triturated with 10 g of soda and extracted with 300 cc of alcohol for 3 hours, the alcohol is removed in vacuo, the residue is mixed with water, filtered off, chlorophyll usually separating out, and the filtrate is acidified with hydrochloric acid .. It is extracted with ether and, after evaporation of the ether, an amorphous powder is obtained which can be purified according to the procedure of Example i.

The antibiotics obtained from green algae according to the above process are amorphous to crystalline bodies with a melting point of 14o to i48 ° C. The antibiotics obtained from freshwater algae are oily to crystalline with a melting point range of 30 to 70 ° C.
