Verfahren zur Herstellung von Diäthylstilböstrolen Das durch bemerkenswerte
physiologische Wirksamkeit bekanntgewordene Diäthylstilböstrol wird nach D o d d
s und Mitarbeiter (vgl. »Nature«, London, Bd. 42, S.34, 1938) auf folgendem Wege
hergestellt: Man setzt Desoxyanisoin mit Jodäthyl und Natriumäthylat zum Äthyldesoxyanisoin
um. Dieses wird mittels Äthylmagnesiumbromid in das 3, 4-Dianisylhexanol-3 übergeführt.
Durch Wasserabspaltung aus diesem tertiären Carbinol durch Behandlung z. B. mit
Phosphortribromid mit nachfolgender Erhitzung mit Dimethylanilin erhält man den
Dimethyläther des Diäthylstilböstrols; dieser wird mit alkoholischer Kalilauge durch
Erhitzen im Autoklaven auf 2oo° zum freien Diäthylstilböstrol vom F. i68° gespalton.Process for the preparation of diethylstilbestroles that by noteworthy

Bei der obenerwähnten Wasserabspaltung aus dem tertiären Carbinol
erhält man aber den gewünschten Dimethyläther des. Diäthylstilböstrols nur in geringer
Ausbeute, der Hauptanteil des Reaktionsproduktes fällt als nichtkristallisierendes
öl an. Unterwirft man nun dieses Öl der Ätherspaltung mit alkoholischer Kalilauge
durch Erhitzen im Autoklaven auf Temperaturen unterhalb 2oo", so erhält
man
im wesentlichen einen dem Diäthylstilböstrol isomeren Stoff, ein Isodiäthylstilböstrol
vom F. I.4:4 bis 4.5'. Die physiologische Wirksamkeit dieses Stoffes Beträgt nur
etwa 1/25 des Diäthylstilböstrols. Es wurde nun gefunden, daß sich das Isodiäthylstilböstrol
leicht in das physiologisch viel wertvollere Diäthylstilböstrol umlagern läßt. Die
Umlagerung kann z. B. durch folgende Formel veranschaulicht werden.
Diese Umlagerung läßt sich auf verschiedene Weise nach an sich für Umlagerungen
von cis- in trans-Isomeren, z. B. von Maleinsäure in Fumarsäure, bekannten Methoden
bewerkstelligen, wie sie z. B. in H o u b e n -W e y l , »Methoden der organ. Chemie<.
3. Aufl. 1925, Band z, S. 1288, oder in B e r n t h s e n , >;Kurzes Lehrbuch der
organ. Chemie:<, 13. Aufl. 1918, S.285, angeführt sind. So kann man durch Erhitzen
von Isoverbindungen mit alkoholischer Kalilauge im Autoklaven auf Temperaturen 'vorzugsweise
von über 2oo°, ferner durch Behandeln der gelösten Ausgangsstoffe mit Metallkatalysatoren,
z. B. Edelmetallkatalysatoren usw., insbesondere schon bei gewöhnlichen Temperaturen
oder durch Erhitzen mit einer Spur Jod auf höhere Temperaturen, zweckmäßig von etwa
18o' usw., die Umlagerung bewirken. Beispiel i i o g Isodiäthylstilböstrol werden
mit z o g Ätzkali und 4o g Alkohol 16 Stunden im Autoklaven auf a25' erhitzt. Nach
dem Erkalten wird der Autoklaveninhalt in Wasser aufgenommen und unter Eiskühlung
mit Salzsäure angesäuert. Die Hauptmenge des Diäthylstilböstrols fällt kristallin
aus und wird abgesaugt. Den Rest gewinnt man durch Ausschütteln der Mutterlauge
mit Äther. B. noble metal catalysts, etc., especially already at ordinary temperatures or by heating with a trace of iodine to higher temperatures, expediently from about 18o ', etc., cause the rearrangement. und mit Ig Palladiumkatalysator 24 Stunden bei Zimmertemperatur geschüttelt. Man
trennt hierauf vom Katalysator ab und verdampft den Äther zur Trockne. Der Rückstand,
aus verdünntem Methanol umkristallisiert, liefert 8 g Diäthylstilböstrol vom F.
