Description
- Gebiet der Erfindung
- Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines cis-1-Amino-2-indanols, welches als Zwischenprodukt für Arzneimittel und Agrochemikalien brauchbar ist, eine neue cis-Oxazolidinonverbindung, welche ein Vorprodukt dieses cis-1-Amino-2-indanols ist, und ein Verfahren zur Herstellung der Verbindung.
- Stand der Technik
- cis-1-Amino-2-indanol ist wichtig als Zwischenprodukt für Arzneimittel.
- Beispielsweise offenbaren J. Med. Chem. 35, 2525 (1992), J. Med. Chem. 35, 1702 (1992) und J. Med. Chem. 35, 1685 (1992), dass diese Verbindung ein Zwischenprodukt von Anti-HIV-Arzneimitteln ist.
- Es gibt mehrere konventionelle Verfahren zur Herstellung von cis-1-Amino-2-indanol.
- Beispielsweise wird in J. Am. Chem Soc. 73, 1639 (1951) die trans-Phenyloxazolinverbindung aus Bromindanol über trans-1-Amino-2-indanol gebildet, und die Verbindung wird in ihre cis-Form invertiert, wobei cis-1-Amino-2-indanol erhalten wird.
- In J. Org. Chem. 32, 540 (1967), wird Jodisocyanat zu Inden zugegebn, um die trans-Oxazolidinverbindung über trans-Iodcarbamat herzustellen, und die Verbindung wird zu der cis-Form invertiert, wobei cis-1-Amino-2-indanol erhalten wird.
- Die offengelegte Japanische Patentanmeldung Hei 7-316106 offenbart eine Synthese einer cis-Oxazolinverbindung aus 1,2-disubstituierten Indanen oder aus cis-1,2-Indenepoxid unter Verwendung der Ritterreaktion und ein Verfahren zur Herstellung von cis-1-Amino-2-indanol durch Hydrolyse der cis-Oxazolinverbindung.
- Es ist bekannt, dass eine Epoxyverbindung durch Reaktion mit einer Heterocumulenverbindung in Gegenwart eimes metallischen Katalysators Additionsverbindungen liefert.
- Beispielsweise offenbaren J. Org. Chem. 51, 2177 (1986), Chem. Lett. 1986, 1963, und Bull. Chem. Soc, Japan 63, 1069 (1990) Synthesen von Oxazolidinonverbindungen durch eine Addidionsreaktion verschiedener Arten von Epoxyverbindungen mit einem Isocyanat, das eine Heterocumulenverbindung ist.
- Das oben erwähnte Verfahren über das trans-Oxazolidinonderivat weist folgende Nachteile auf, dass viele Herstellungsstufen wegen der Notwendigkeit der Inversion zu der cis-Form erforderlich sind, eine hohe Temperatur notig ist, die Volumeneffizienz und die Ausbeute niedrig sind und eine große Menge von Abfallmaterial gebildet wird.
- Synthesis 541 (1997) betrifft die Synthese von enantiomerenfreiem (1S, 2R)-1-aminoindan-2-ol. das für die Synthese von Indinavir, einem HIV-Proteasehemmer, verwendet werden kann. Das Zwischenprodukt wird mittel einer basisch katalysierten Hydrolyse von (4S, 5R)-Indano[1,2-d]oxazolidin-2-on hergestellt.
- J. Org. Chem. 51, 2177 (1986) offenbart eine Cycloadditionsreaktion von Heterocumulenen mit Oxiranen, welche durch einen Organozinniodid-Lewisbase-Komplex katalysiert wird. Insbesondere wird gelehrt, dass ein Isocyanat mit einem Oxiran zur Reaktion gebracht werden kann, um ein 2-Oxazolidinon zu liefern.
- Tetrahedron Lett. 36, 7619 (1995) betrifft die Synthese eines Oxazolidinons (3) aus einem Aminoindanol (1). Die Verwendung dieses Oxazolidinons in der Synthese von Indinavir mittels einer Diels-Alder-Reaktion liefert eine verbesserte Stereosteuerung der Indinavirsynthese.
- Tetrahedron 47, 4941 (1991) betrifft die Synthese von Aminoalkoholen. Es wird gelehrt, dass eine Hydrolyse von Oxazolidin-2-onen mittels eines Alkalis die entsprechenden 1-Aminoindan-2-ole liefert (Verbindungen 23 und 24).
- JP-A-356073077 betrifft die Herstellung einer 3,5-disubstituierten 2-Oxazolidonverbindung durch Reaktion eines substituierten Isocyanats mit einem substituierten Oxiran in Gegenwart einer Lithiumhalogenidverbindung.
- Synlett 129 (1994) betrifft die Synthese von Oxazolidinonen über eine lanthanidkatalysierte Cycloaddition von Epoxiden mit Isocyanaten. Es wurde gefunden, dass Lanthanidtrichlorid-Katalysatoren für die Katalyse der obigen Reaktion besonders gut brauchbar sind, da sie Oxazolidinone in hohen Ausbeuten liefern.
- EP-A1-05 18 153 offenbart die Reaktion eines substituierten Isocyanats mit einem substituierten Oxiran, um ein substituiertes Oxazolidinon zu liefern. Es wird gelehrt, dass für die Katalyse dieser Reaktion ein Halogensalz eines Alkalimetalls, bevorzugt Lithiumchlorid oder Lithiumbromid, verwendet werden sollte.
- Bioinorganic and Medicinal Chemistry letters, Vol. 6, No. 9, Seiten 1009–1014 (1996), betrifft die Synthese und die antibakterielle Wirksamkeit von neuen troponsubstituierte Phenyloxazolidinone enthaltenden antibakteriellen Mitteln. Es wird offenbart, dass ein substituiertes Isocyanat mit einem Oxiran in Gegenwart eines Lithiumbromidkatalystors zur Reaktion gebracht werden kann, um eine substituierte Oxazolidinonverbindung herzustellen.
- J. Heterocyclic Chem. 25, 1431 (1988) offenbart die Cycloadditionsreaktion eines Isocyanats mit einem Epoxid, um eine Oxazolidonverbindung zu liefern.
- J. Am. Chem. Soc. 109, 792 (1987), betrifft eine Methode, um eine cis-Hydroxyaminierung zustande zu bringen. Es wird beschrieben, wie dies durchgeführt werden kann, indem über eine Reaktion eines Isocyants mit einem Epoxid unter geeigneten Bedingungen ein cis-Oxazolidon gebildet wird und anschließend die so hergestellte Oxazolidonverbindung hydrolysiert wird, um eine cis-hydroxyaminierte Verbindung zu erhalten.
- Chem. Rev. 72, 457 (1972) betrifft Fortschritte in der Isocyanatchemie. Auf Seite 481, Sektion 16, wird offenbart, dass ein 2-Oxazolidon aus einem Isocyanat und einem Epoxid hergestellt werden kann. Es wird festgestellt, dass die Reaktion schnell und mit einer hohen Ausbeute von statten geht, wenn sie in Gegenwart eines löslichen Lithiumbromid-Phosphinoxid-Addukts durchgeführt wird.
- Offenbarung der Erfindung
- Als Resultat der von den jetzigen Erfindern durchgeführten eingehenden Forschung über Verfahren zur effizienteren Herstellung von cis-1-Amino-2-indanol fanden sie eine neue cis-Oxazolidinonverbindung, ein Verfahren zu deren Herstellung und ein Verfahren zur Herstellung des Aminoalkohols mittels einer Hydrolysereaktion der genannten Verbindung, und sie vollendeten die vorliegende Erfindung.
- Die vorliegende Erfindung betrifft nämlich eine cis-Oxazolidinonverbindung der Formel (1) in racemischer Form oder optisch aktiver Form: worin R eine substituierte oder unsubstituierte C1-6-Alkylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C2-6-Alkenylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkoxygruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkylaminogruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte Arylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom) oder ein Halogenatom darstellt, und der Oxazolidinonring in der cis-Konfiguration vorliegt, wobei der cis-Oxazolidinonring insbesondere eine optisch aktive Oxazolidinonverbindung der Formel (2) ist, worin * ein assymmetrisches Kohlenstoff bedeutet und die absolute Konfiguration des Kohlenstoffatoms R oder S ist und der Substituent R die gleiche Bedeutung aufweist, wie sie in der Formel (1) definiert ist.
- Ferner betrifft die vorliegende Erfindung ein Verfahren zur Herstellung der cis-Oxazolidinonverbindung der Formel (1), umfassend die Reaktion von cis-1,2-Indenepoxid der Formel (3), welches in racemischer Form oder oder in optisch aktiver Form vorliegt, worin der Epoxyring in cis-Konfiguration vorliegt, mit einer Sulfonylisocyanatverbindung der Formel (4) worin R die gleiche Bedeutung aufweist, wie sie in der Formel (1) definiert wurde, in Gegenwart von Natriumjodid.
- Ferner betrifft die vorliegende Erfindung ein Verfahren zur Herstellung von cis-1-Amino-2-indanol der Formel (5), welches in racemischer Form oder oder in optisch aktiver Form vorliegt, worin die NH2-Gruppe und die OH-Gruppe sich in cis-Form befinden, durch saure oder alkalische Hydrolyse der cis-Oxazolidinonverbindung der Formel (1).
- Die vorliegende Erfindung wird unten detailliert erläutert.
- Zuerst wird der Substituent R erläutert.
- Die C1-6-Alkylgruppe umfaßt die Methylgruppe, die Ethylgruppe, die n-Propylgruppe, die Isopropylgruppe, die n-Butylgruppe, die Isobutylgruppe, die sek.Butylgruppe, die tert.Butylgruppe, die n-Amylgruppe, die Isoamylgruppe, die Neopentylgruppe, die n-Hexylgruppe und die Cyclohexylgruppe.
- Die C1-6-Alkoxygruppe umfaßt die Methoxygruppe, die Ethoxygruppe, die n-Propoxygruppe, die Isopropoxygruppe, die n-Butoxygruppe, die Isobutoxygruppe, die sek.Butoxygruppe, die tert.Butoxygruppe, die n-Amyloxygruppe, die Isoamyloxygruppe, die Neopentyloxygruppe, die n-Hexyloxygruppe und die Cyclohexyloxygruppe.
- Die C2-7-Alkoxycarbonylgruppe umfaßt die Methoxycarbonylgruppe, die Ethoxycarbonylgruppe, die n-Propoxycarbonylgruppe, die Isopropoxycarbonylgruppe, die n-Butoxycarbonylgruppe, die Isobutoxycarbonylgruppe, die sek.Butoxycarbonylgruppe, die n-Amyloxycarbonylgruppe, die Isoamyloxycarbonylgruppe, die Neopentyloxycarbonylgruppe, die n-Hexyloxycarbonylgruppe und die Cyclohexyloxycarbonylgruppe.
- Die C2-7-Alkylcarbonyloxygruppe umfaßt die Methylcarbonyloxygruppe, die Ethylcarbonyloxygruppe, die n-Propylcarbonyloxygruppe, die Isopropylcarbonyloxygruppe, die n-Butylcarbonyloxygruppe, die Isobutylcarbonyloxygruppe, die sek.Butylcarbonyloxygruppe, die tert.Butylcarbonyloxygruppe, die n-Amylcarbonyloxygruppe, die Isoamylcarbonyloxygruppe, die Neopentylcarbonyloxygruppe, die n-Hexylcarbonyloxygruppe und die Cyclohexylcarbonyloxygruppe.
- Die C2-7-Alkanoylgruppe umfaßt die Acetylgruppe, die Ethylcarbonylgruppe, die n-Propylcarbonylgruppe, die Isopropylcarbonylgruppe, die n-Butylcarbonylgruppe, die Isobutylcarbonylgruppe, die sek.Butylcarbonylgruppe, die, die n-Amylcarbonylgruppe, die Isoamylcarbonylgruppe, die Neopentylcarbonylgruppe, die n-Hexylcarbonylgruppe und die Cyclohexylcarbonylgruppe.
- Die C2-6-Alkenylgruppe umfaßt die Vinylgruppe, die Isopropenylgruppe, die Allylgruppe, die Methallylgruppe, die 1-Butenylgruppe, die 3-Hexenylgruppe, die 1-Cyclopentenylgruppe, die 1-Cyclohexenylgruppe, die Methoxyvinylgruppe und die Ethoxyvinylgruppe.
- Die C1-6-Alkylaminogruppe umfaßt eine Monoalkylaminogruppe und eine Dialkylaminogruppe.
- Konkrete Beispiele für die C1-6-Alkylaminogruppe sind die Methylaminogruppe, die Ethylaminogruppe, die n-Propylaminogruppe, die Isopropylaminogruppe, die n-Butylaminogruppe, die Isobutylaminogruppe, die sek.Butylaminogruppe, die tert.Butylaminogruppe, die n-Amylaminogruppe, die Isoamylaminogruppe, die Neopentylaminogruppe, die n-Hexyl aminogruppe, die Cyclohexylaminogruppe, die Dimethylaminogruppe und die Diethylaminogruppe.
- Die Arylgruppe umfaßt die Phenylgruppe, die o-Tolylgruppe, die m-Tolylgruppe, die p-Tolylgruppe, die 3,5-Dimethylphenylgruppe, die 2,4,6-Trimethylphenylgruppe, die 2,4,6-Triisopropylphenylgruppe, die 4-Isopropylphenylgruppe, die 4-tert.Butylphenylgruppe, die 4-Methoxyphenylgruppe, die 3,5-Dimethoxyphenylgruppe, die 2-Fluorphenylgruppe, die 3-Fluorphenylgruppe, die 4-Fluorphenylgruppe, die 2-Chlorphenylgruppe, die 3-Chlorphenylgruppe, die 4-Chlorphenylgruppe, die 2-Bromphenylgruppe, die 3-Bromphenylgruppe, die 4-Bromphenylgruppe, die 2,4-Dichlorphenylgruppe, die 4-Phenylphenylgruppe, die 1-Naphthylgruppe, die 2-Naphthylgruppe, die 1-Anthracenylgruppe, die 2-Anthracenylgruppe, die 5-Anthracenylgruppe, die 2-Pyridylgruppe, die 3-Pyridylgruppe und die 4-Pyridylgruppe.
- Das Halogenatom umfaßt ein Fluoratom, ein Chloratom, ein Bromatom und ein Jodatom.
- Nachstehend folgt eine Erläuterung der Additionsreaktion von cis-1,2-Indenepoxid der Formal (3) mit der Sulfonylisocynatverbindung der Formel (4).
- Das cis-1,2-Indenepoxid der Formal (3) umfaßt die racemische Form und die optisch aktive Form von (+)-cis-1,2-Indenepoxid und (–)-cis-1,2-Indenepoxid.
- Die Sulfonylisocyanatverbindung der Formel (4) umfaßt Methansulfonylisocyanat, Ethansulfonylisocyanat, n-Propansulfonylisocyanat, n-Butansulfonylisocyanat, Trifluormethansulfonylisocyanat, Benzolsulfonylisocyanat, p-Toluolsulfonylisocyanat und Chlorsulfonylisocyanat. Bevorzugt sind Methansulfonylisocyanat und p-Toluolsulfonylisocyanat.
- Die Menge der verwendeten Sulfonylisocyanatverbindung der Formel (4) liegt im Bereich von der 0,1-fachen bis zur 10-fachen molaren Menge, bevorzugt von der 0,5-fachen bis zur 5-fachen molaren Menge, bezogen auf cis-1,2-Indenepoxid der Formal (3).
- In der vorliegenden Erfindung kann die Reaktion bei gleichzeitiger Anwesenheit einer Lewis-Base durchgeführt werden, um so die Reaktion zu fördern.
- Die Lewis-Base umfaßt Amine, wie Triethylamin, Tri-n-butylamin, 1,8-Diazabicyclo[5,4,0]-7-undecen (DBU) und 1,5-Diazabicyclo[4,3,0]-5.nonen (DBN), Pyridine wie Pyridin, Picolin, Lutidin und Methylethyl-pyridin, Pyridin-N-oxide wie Pyridin-N-oxid, Picolin-N-oxid, Lutidin-N-oxid und 4-Phenylpyridin-N-oxid, Phosphine wie Triphenylphosphin und Tri-n-butylphosphin, Phosphinoxide wie Triphenylphosphinoxid und Tri-n-butylphosphinoxid, Phosphoniumsalze wie Tetra-n-butylphosphoniumbromid, Tetraphenylphosphoniumbromid, Tetra-n-butylphosphoniumiodid und Tetraphenylphosphoniumiodid, Sulfoxide wie wie Dimethylsulfoxid, Diethylsulfoxid, Di-n-propylsulfoxid und Diphenylsulfoxid, Harnstoffe wie Tetramethylharnstoff, Tetrabutylharnstoff und Dimethylpropylharnstoff, Hexamethylphosphorsäuretriamid (HMPA), 2,2'-Azobisisobutylnitril (AIBN). Bevorzugt sind Tri-n-butylphosphinoxid, Tetrabutylharnstoff und Hexamethylphosphorsäuretriamid (HMPA).
- Die Menge des verwendeten Natriumiodids liegt im Bereich von 0,1 bis 50 Mol%, bevorzugt von 1 bis 20 Mol%, bezogen auf das cis-1,2-Indenepoxid der Formal (3).
- Die Menge der verwendetem Lewis-Base liegt im Bereich von der äquimolaren Menge bis zur dreifachen molaren Menge, bezogen auf das Metallhalogenid.
- Das Reaktionslösungsmittel ist nicht besonders eingeschränkt, sofern es nicht an der Reaktion teilnimmt.
- Beispiele für das Reaktionslösungsmittel sind Nitrile wie Acetonitril, Propionitril und Butyronitril. Ketone wie Aceton, Methylethylketon und Methylisobutylketon, aromatische Kohlenwasserstoffe wie Benzol, Toluol, Xylol, Mesitylen, Chlorbenzol und o-Dichlorbenzol, aliphatische Kohlenwasserstoffe wie n-Hexan, Cyclohexan, n-Octan und n-Decan, Ester wie Methylacat, Ethylacetat und Butylacetat, halogenierte Kohlenwasserstoffe wie Dichlormethan, Dichlorethan, Chloroform und Tetrachlorkohlenstoff, Ether wie Tetrahydrofuran, Diethylether, tert.Butylmethylether und Dimethoxyethan, Alkohole wie Methanol, Ethanol, 1-Propanol, 2-Propanol, 1-Butanol, 2-Butanol, Isobutanol und Cyclohexanol, Amide wie N,N,Dimethylformamid, N,N,Dimethylacetamid und N-Methylpyrrolidon, und Harnstoffe wie 1,3-Dimethylimidazolinon, Tetramethylharnstoff und Tetrabutylharnstoff. Bevorzugt sind Toluol, Cyclohexan, Ethylacetat und Tetrabutylharnstoff.
- Diese Lösungsmittel können einzeln oder in Kombination verwendet werden.
- Die Reaktionstemperatur ist nicht besonders beschränkt und sie kann von –20°C bis zum Siedepunkt des zu verwenden Lösungsmittels angewendet werden. Die Reaktion wird jedoch bevorzugt im Bereich von 20°C bis 100°C durchgeführt.
- Die Reaktionszeit liegt normalerweise zwischen 0,1 und 1000 Stunden, obwohl sie in Abhängigkeit von der Reaktivität der Sulfonylisocyanatverbindung der Formel (4) variiert.
- Nach Vollendung der Reaktion wird Wasser zugegeben und eine Extraktion mit einem geeigneten Lösungsmittel wird durchgeführt und das Lösungsmittel wird unter vermindertem Druck konzentriert, um ein Rohprodukt zu erhalten, oder eine feste Substanz wird aus einem geeigneten Lösungsmittel ausgefällt, um ein Rohprodukt zu erhalten.
- Ferner kann eine reine cis-Oxazolidinonverbindung der Formel (1) erhalten werden, indem die Reinigung gemäß einem üblichen Verfahren, wie Umkristallisation oder Silicagel-Säulenchromatographie usw. durchgeführt wird.
- Nun wird die Hydrolysereaktion der cis-Oxazolidinonverbindung der Formel (1) erläutert.
- Bei der Hydrolysereaktion wird eine Säure oder ein Alkali verwendet.
- Die Säure umfaßt anorganische Säuren wie Salzsäure, Schwefelsäure, Phosphorsäure, Salpetersäure, Bromwasserstoffsäure, Jodwasserstoffsäure und Fluorwasserstoffsäure, Carbonsäuren wie Ameisensäure, Essigsäure, Trichloressigsäure und Trifluoressigsäure, Sulfonsäuren wie p-Toluolsulfonsäure, Methansulfonsäre und Trifluormethansulfonsäure.
- Das Alkali umfaßt Hydroxide von Alkalimetallen oder Erdalkalimetallen wie Natriumhydroxid, Kaliumhydroxid, Lithiumhydroxid, Magnesiumhydroxid, Calciumhydroxid und Bariumhydroxid, Carbonate von Alkalimetallen oder Erdalkalimetallen wie Natriumcarbonat, Kaliumcarbonat, Lithiumcarbonat, Magnesiumcarbonat, Calciumcarbonat und Baiumcarbonat. Diese Alkalien können in wässeriger Lösung verwendet werden.
- Die Menge der verwendeten Säure oder des Alkalis ist normalerweise eine äquimolare Menge oder mehr, bezogen auf die cis-Oxazolidinonverbindung der Formel (1). Um die Hydrolysereaktion in kurzer Zeit zu Ende zu bringen, kann ein großer Überschuß, bis zum 20fachen der molaren Menge, verwendet werden.
- Die Reaktion kann in saurer oder alkalischer Lösung durchgeführt werden, aber sie kann auch in einem organischen Lösungsmittel durchgeführt werden.
- Das organische Lösungsmittel ist nicht besonders eingeschränkt, sofern es nicht an der Reaktion teilnimmt. Das organische Lösungsmittel umfaßt Ketone wie Aceton, Methylethylketon, Methylisobutylketon, aromatische Kohlenwasserstoffe wie Benzol, Toluol, Xylol, Mesitylen, Chlorbenzol und o-Dichlorbenzol, aliphatische Kohlenwasserstoffe wie n-Hexan, Cyclohexan, n-Octan und n-Decan, halogenierte Kohlenwasserstoffe wie Dichlormethan, Dichlorethan, Chloroform und Tetrachlorkohlenstoff, Ether wie Tetrahydrofuran, Diethylether, tert.Butylmethylether, Dimethoxyethan und Dioxan, Alkohole wie Methanol, Ethanol, 1-Propanol, 2-Propanol, 1-Butanol, 2-Butanol, Isobutanol und Cyclohexanol. Bevorzugt sind Aceton, Ethanol und Dioxan.
- Diese Lösungsmittel können einzeln oder in Kombination verwendet werden. Die Temperatur der Hydrolyse ist nicht besonders eingeschränkt, aber sie ist normalerweise von 0°C bis zum Siedepunkt eines zu verwenden Lösungsmittels möglich, bevorzugt von 20°C bis 100°C.
- Die Reaktionszeiten liegen normalerweise zwischen 0,1 bis 1000 Stunden.
- Nach Vollendung der Hydrolyse wird die Extraktion gemäß der üblichen Methode durch Zugabe von Wasser und von einem geeigneten Lösungsmittel durchgeführt, das Lösungsmittel wird unter vermindertem Druck konzentriert, um ein Rohprodukt zu erhalten, oder eine feste Substanz wird aus einem geeigneten Lösungsmittel ausgefällt, um ein Rohprodukt zu erhalten.
- Ferner kann eine reine cis-1-Amino-2-indanol-verbindung der Formel (5) erhalten werden, indem die Reinigung gemäß einem üblichen Verfahren wie Umkristallisation, oder Silicagel-Säulenchromatographie usw. durchgeführt wird.
- Beste Methode zur Durchführung der Erfindung
- Die vorliegende Erfindung wird nachstehend detaillierter unter Bezugnahme auf Beispiele erläutert.
- Zu 50 ml Toluol wurden 5,00 g (+)-cis-1,2-Indenepoxid (34 Mmole, 89,4%ige Reinheit, 90,2% ee in optischer Reinheit), 1,65 g (10 Mol% bezogen auf das oben erwähnte Epoxid) Di-n-butylzinniodid (n-Bu2SnI2) und 0,606 g (10 Mol% bezogen auf das oben erwähnte Epoxid) Hexamethylphosphorsäuretriamid (HMPA) unter einer Argonatmosphäre zugegeben. Die erhaltene Mischung wurde auf 50 bis 60°C erhitzt, und 25 ml einer Toluollösung von 6,67 g (34 Mmole) p-Toluolsulfonylisocyanat wurden tropfenweise zu der Mischung innerhalb einer Stunde zugegeben.
- Nachdem die Mischung drei Stunden bei 60°C gerührt worden war, wurde sie unter Rückfluß eine weitere Stunde gerührt.
- Nachdem die Reaktionsflüssigkeit unter vermindertem Druck konzentriert und Acetonitril zugegeben und darin vollständig aufgelöst worden war, wurde das gewünschte Produkt in der Lösung mittels Flüssigkeitschromatographie (Säule: Inertsil ODS-2 4,6Φ × 250 mm, Säulentemperatur: 40°C, Eluent: Acetonitril/Wasser = 50/50, Fließgeschwindigkeit: 1,0 ml/Min., Nachweis: UV 254 nm) bestimmt. Der Gehalt an dem gewünschten Produkt betrug 9,99 g (Ausbeute: 90%).
- Die Lösung wurde konzentriert, dann wurde aus Acetonitril umkristallisiert, um 6,0 g an gereinigtem Produkt zu erhalten.
- Die optische Reinheit des gereinigten Produkts betrug 98,4% ee. (HPLC, Säule: Chiralcel OJ-R 4,6Φ × 250 mm, Säulentemperatur: 35°C, Eluent: Acetonitril/Wasser = 50/50, Fließgeschwindigkeit: 0,5 ml/Min., Nachweis: UV 254 nm).

- Die Reaktion und die Nachbehandlung wurden in der gleichen Weise wie im Beispiel 1 durchgeführt, mit der Ausnahme, dass die Reaktionslösungsmittel variiert wurden, um cis-N-p-Toluolsulfonylindano[1,2-d]oxazolidin-2-on (6) zu erhalten. Die Resultate werden in der folgenden Tabelle gezeigt.
- Beispiele 9 bis 19
- Die Reaktion und die Nachbehandlung wurden in der gleichen Weise wie im Beispiel 1 durchgeführt, mit der Ausnahme, dass verschiedene Arten von Metallhalogenidverbindungen (10 Mol% des oben erwähnten Epoxids) statt des Di-n-butylzinniodids und verschiedene Arten von Lewis-Basen (20 Mol% des oben erwähnten Epoxids, aber bei Tetramethylharnstoff nur 10 Mol%) statt des Hexamethylphosphorsäuretriamids) (HMPA) verwendet wurden, um cis-N-p-Toluolsulfonylindano[1,2-d]-oxazolidin-2-on (6) zu erhalten. Die Resultate werden in der folgenden Tabelle gezeigt.
- Beispiel 20
- Zu 30 ml einer Suspension von Tetramethylharnstoff, enthaltend 324 mg Natriumiodid (10 Mol% des unten erwähnten Epoxids), wurden 3,00 g (+)-cis-1,2-Indenepoxid (22 Mmol, Reinheit 96%, optische Reinheit 90% ee) unter einer Argonatmosphäre bei 25°C zugegeben. Nach Rühren der Mischung bei 25°C über 30 Minuten wurde sie auf 45°C erwärmt.
- Zu der verrührten Flüssigkeit wurden tropfenweise 15 ml einer Lösung von Tetrabutylharnstoff, enthaltend 4,58 g (23 Mmole) p-Toluolsulfonylisocyanat, bei 45 bis 50°C innerhalb einer Stunde zugegeben. Anschließend wurde die erhaltene Mischung über 3 weitere Stunden bei dieser Temperatur gerührt.
- Die verrührte Reaktionsflüssigkeit wurde zu Hexamethylphophorsäuretriamid und Acetonitril gegeben und darin vollständig aufgelöst. Danach wurde das gewünschte Produkt in der Lösung mittels Flüssigkeitschromatographie (Säule: Inertsil ODS-2 4,6Φ × 250 mm, Säulentemperatur: 40°C, Eluent: Acetonitril/Wasser = 60/40 (0,2% Essigsäure), Fließgeschwindigkeit: 1,0 ml/Min., Nachweis: UV 254 nm) bestimmt. Als Ergebnis betrug der Gehalt des gewünschten Produkts 7,10 g (Ausbeute; 99%).
- Beispiel 21
- Zu 125 ml einer Suspension von Tetramethylharnstoff, enthaltend 2,89 g Natriumiodid, (10 Mol% des unten erwähnten Epoxids) wurden 25,0 g (+)-cis-1,2-Indenepoxid (0,153 Mole, Reinheit 81%, optische Reinheit 93% ee) unter einer Argonatmosphäre bei 25°C zugegeben. Nach Rühren der Mischung bei 25°C über 30 Minuten wurde sie auf 45°C erwärmt.
- Zu der verrührten Flüssigkeit wurden 31,7 g (0,161 Mole) p-Toluolsulfonylisocyanat bei 45 bis 50°C innerhalb einer Stunde zugegeben. Anschließend wurde die Mischung über drei weitere Stunden bei dieser Temperatur gerührt.
- Nach Abkühlen der verrührten Reaktionsflüssigkeit auf 4°C wurden die Ausfällungen abfiltriert und die Kristalle wurden mit Methanol gewaschen.
- Die erhaltenen Kristalle wurden getrocknet, um 46,2 g des gewünschten Produkts zu erhalten (Reinheit: 98%, Ausbeute; 90%).
- Die optische Reinheit der erhaltenen Kristalle betrug 98% ee. (HPLC, Säule: Chiracel OJ-R 4,6Φ × 250 mm, Säulentemperatur: 35°C, Eluent: Acetonitril/Methanol/Wasser = 3/2/5, Fließgeschwindigkeit: 0,9 ml/Min., Nachweis: UV 254 nm).
- 1 ml von 97%iger Schwefelsäure wurde zu 1,00 g cis-N-p-Toluolsulfonylindano[1,2-d]-oxazolidin-2-on (6) (3,0 Mmole, 99%ige Reinheit, 100% ee optische Reinheit) gegeben und bei 60°C eine Stunde gerührt.
- Dann wurde eine wässerige Natriumhydroxidlösung zu der Mischung zugegeben und der pH-Wert wurde auf 10 bis 11 eingestellt. Nach Rühren der Mischung über 30 Minuten bei 80°C, wurde durch Zugabe von Chloroform eine Extraktion durchgeführt.
- Die Chloroformschicht wurde mittels Natriumsulfat getrocknet und dann konzentriert, um 0,41 g des gewünschten Produkts zu erhalten (Ausbeute: 91%). Die optische Reinheit war 100% ee.
- Beispiel 23
- Die Reaktion und die Nachbehandlung wurden in der gleichen Weise wie im Beispiel 17 durchgeführt mit der Ausnahme, dass 3 ml einer wässerigen Lösung von 40%igem Natriumhydroxid statt Sulfonsäure verwendet wurden und Rühren über drei Tage unter Rückfluß durchgeführt wurde, um 0,16 g (–)cis-1-Amino-2-indanol (8) zu erhalten (Ausbeute: 35%). Die optische Reiheit betrug 100% ee.
- Beispiel 24
- 28 g von 85%iger Sulfonsäure wurden zu 9,9 g cis-N-p-Toluolsulfonylindano[1,2-d]-oxazolidin-2-on (6) (29 Mmole, 96,7%ige Reiheit, 100%ige optische Reinheit) unter einer Argonatmosphäre gegeben und drei Stunden bei 72°C gerührt.
- Nachdem die Mischung auf 20°C gekühlt worden war, wurden 51 g Wasser zu der Mischung zugegeben und die ausgefallenen Kristalle wurden abfiltriert.
- Die erhaltenen Kristalle wurden mit Wasser gewaschen und dann unter vermindertem Druck getrocknet, um 5,16 g des Oxazolidinon-Zwischenbprodukts (cis-[1,2-d]-Oxazolidin-2-on) (92%ige Reinheit, Ausbeute: 94%) zu erhalten.
- 3,26 g (17,1 Mmole) des Oxazolidinon-Zwischenprodukts wurden zu einer wässerigen Lösung von 1,44 g Natriumhydroxid, gelöst in 15,0 g Wasser, unter einer Argonatmosphäre gegeben und die Mischung wurde 6 Stunden bei 85°C gerührt.
- Nach Vollendung der Reaktion wurde zur Mischung Chloroform gegeben, es wurde extrahiert und die Chloroformschicht wurde konzentriert, um 2,54 g des gewünschten Produkts zu erhalten (Ausbeute: 99%).
- Die optische Reinheit betrug 100% ee.
- Industrielle Anwendbarkeit
- Gemäß der vorliegenden Erfindung können neue cis-Oxazolidinonverbindungen der Formel (1) leicht aus cis-1,2-Indenepoxid der Formel (3) und einer Sulfonylisocyanatverbindung der Formel (4) hergestellt werden, und ferner kann cis-1-Amino-2-indanol der Formel (5) durch Hydrolyse der cis-Oxazolidinonverbindung hergestellt werden. cis-1-Amino-2-indanol der Formel (5) ist als Zwischenprodukt für Arzneimittel, z. B. von Arzneimitteln gegen HIV, brauchbar.

- Cis-oxazolidinon-Verbindung der Formel (1) in racemischer Form oder optisch aktiver Form: worin R eine substituierte oder unsubstituierte C1-6-Alkylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C2-6-Alkenylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkoxygruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkylaminogruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte Arylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom) oder ein Halogenatom ist, und der Oxazolidinonring liegt in der cis-Konfiguration vor.
- Verbindung gemäss Anspruch 1, worin die cis-Oxazolidinon-Verbindung eine optisch aktive Oxazolidinonverbindung der Formel (2) ist: worin * ein asymmetrisches Kohlenstoffatom bedeutet, und die absolute Konfiguration des Kohlenstoffatoms ist R oder S, und der Substituent R hat die gleiche Bedeutung wie in Formel (1) definiert.
- Optisch aktive Oxazolidinonverbindung gemäss Anspruch 2, worin R eine p-Tolylgruppe ist.
- Verfahren zur Herstellung einer cis-Oxazolidinon-Verbindung der Formel (1): worin R eine substituierte oder unsubstituierte C1-6-Alkylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C2-6-Alkenylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkoxygruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkylaminogruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte Arylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom) oder ein Halogenatom ist, und der Oxazolidinonring liegt in der cis-Konfiguration vor, das die Umsetzung von cis-1,2-Indenepoxid der Formel (3) in racemischer oder optisch aktiver Form: worin der Epoxyring in der cis-Konfiguration vorliegt, mit einer Sulfonylisocyanatverbindung der Formel (4): worin R die gleiche Bedeutung hat wie in Formel (1) definiert, in Gegenwart von Natriumiodid umfasst.
- Verfahren zur Herstellung von cis-1-Amino-2-indanol der Formel (5) in racemischer oder optisch aktiver Form: worin die NH2-Gruppe und die OH-Gruppe in der cis-Konfiguration vorliegen, durch saure oder alkalische Hydrolyse der cis-Oxazolidinon-Verbindung der Formel (1) worin R eine substituierte oder unsubstituierte C1-6-Alkylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C2-6-Alkenylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkoxygruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkylaminogruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte Arylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom) oder ein Halogenatom ist, und der Oxazolidinonring liegt in der cis-Konfiguration vor.
- Verfahren zur Herstellung einer cis-Oxazolidinon-Verbindung gemäss Anspruch 4, worin das cis-1,2-Indenepoxid ein optisch aktives (+)-cis-1,2-Indenepoxid ist.
- Verfahren zur Herstellung einer cis-Oxazolidinon-Verbindung gemäss Anspruch 4, worin das cis-1,2-Indenepoxid ein optisch aktives (–)-cis-1,2-Indenepoxid ist.
- Verfahren zur Herstellung einer cis-Oxazolidinon-Verbindung gemäss Anspruch 4, worin R eine p-Tolylgruppe ist.
- Verfahren zur Herstellung einer cis-Oxazolidinon-Verbindung gemäss Anspruch 5, worin die Verbindung der Formel (1) optisch aktiv ist.
- Verfahren zur Herstellung von cis-1-Amino-2-indanol der Formel (5) in racemischer oder optisch aktiver Form: worin die NH2-Gruppe und die OH-Gruppe in der cis-Konfiguration vorliegen, das Verfahren umfasst: Erhalt einer cis-Oxazolidinon-Verbindung der Formel (1): worin R eine substituierte oder unsubstituierte C1-6-Alkylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C2-6-Alkenylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkoxygruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte C1-6-Alkylaminogruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom), eine substituierte oder unsubstituierte Arylgruppe (der Substituent ist eine C1-6-Alkylgruppe, C1-6-Alkoxygruppe, C2-7-Alkoxycarbonylgruppe, C2-7-Alkylcarbonyloxygruppe, C2-7-Alkanoylgruppe, Phenylgruppe oder ein Halogenatom) oder ein Halogenatom ist, und der Oxazolidinonring liegt in der cis-Konfiguration vor, durch Umsetzung von cis-1,2-Indenepoxid der Formel (3) in racemischer oder optisch aktiver Form: worin der Epoxyring in der cis-Konfiguration vorliegt, mit einer Sulfonylisocyanatverbindung der Formel (4): worin R die gleiche Bedeutung hat wie für Formel (1) definiert, in Gegenwart eines Metallhalogenid-Katalysators, ausgewählt aus Alkalimetallhalogenidverbindungen, Zinnhalogenidverbindungen und Zinkhalogenidverbindungen, und Hydrolysieren der Oxazolidinonverbindung.
