Description
Process for the preparation of a tasteless quinine salt of 2-phenylquinoline-4-carboxylic acid The patent 563 457 is the. Production of tasteless quinine salt the 2-phenylquinoline-q.-carboxylic acid protected. The gist of this procedure is the implementation of the starting materials at temperatures from 6o to 70 ° in aqueous solution or in the presence of chloroform. The other procedures that have become known lead as could be proven, not unnerving to as good as clunk-free Bodies, but rather too strongly bitter-tasting salts.
Beispiel i 32 Teile Chinin werden in ioo Teilen Alkohol gelöst. It has now been found that with the help of other organic Solvents such as alcohol; Benzene, etc., tasteless - quinine salt with the mentioned Acid is formed when the .influential when combining the starting materials Temperature limits from 6o to 70 ° are initially ignored, d. H. at low temperatures works, and then the salt obtained in this way by washing out with a suitable washing liquid Treated from 6o to 7o °. According to the invention, the starting materials can therefore be used temperatures lower than 6o to 70 ° in organic solvents, if necessary using solubilizers such as ether. The follow-up of the reaction product consists in the fact that it is separated from the mother liquor with water or organic solvents, in which the quinine salt is only slightly dissolves, is quickly washed out at temperatures of 6o to 70 °. The previous or subsequent treatment with cold washing liquids is without disadvantage. Afterward the reaction product can also be recrystallized from suitable solvents. Example i 32 parts of quinine are dissolved in 100 parts of alcohol. To this solution a slurry of 34 parts of phenylquinoline carboxylic acid is brought with stirring in 35o parts alcohol. When adding 150 parts of ether, stirring occurs if necessary after gentle warming, immediate solution, and after a short time the compound falls in flakes. After separation from the solvent, the reaction product can be mixed with water or organic solvents from 6o to 7o ° and washed out of suitable solvents be recrystallized.
Example 2 Equimolecular amounts of quinine base and phenylquinoline carboxylic acid are dissolved in benzene with gentle warming and left to crystallize. After filtering off the reaction product and after removing the adhering benzene the quinine salt with water or any suitable one organic Solvent washed out from 60 to 70 °. This can lead to recrystallization connect.
Example 3 32 parts of quinine base become ethylene glycol in zoo parts dissolved with warming. A solution of is added to this solution with stirring 24 parts of phenylquinoline carboxylic acid in 300 parts of ethylene glycol. After adding the twice the amount of water, the compound precipitates in flakes. After separating from the Mother liquor becomes the reaction product with water or a suitable organic Solvent washed out from 6o to 7o °.
Example 4 20 parts of quinine hydrochloride are converted into zoo parts of alcohol dissolved with warming. A solution of 1.3 parts is given for this purpose. phenylquinolinecarboxylic acid Sodium in zoo parts alcohol. When washing out with water or a suitable one organic solvents from 6o to 70 ° the quinine salt is obtained completely pure.
Example 5 20 g of quinine hydrochloride dissolved in 150 g of glycerol, and 13 g of phenylquinoline carboxylic acid sodium, dissolved in zoo g of glycerine, are taken under Stir combined. Here the connection fails, which is then with water or a suitable organic solvent from 6o to 70 ° washed out 'vvird.f example 6 Equimol.ecular amounts of quinine hydrochloride and phenylquinoline carboxylic acid Sodium is dissolved in ethylene glycol, mixed and left to crystallize. It is useful to add water here. After filtering off the reaction product the quinine salt is washed out with water or organic solvents at 6o to 70 °. Example 7 32 parts of quinine are dissolved in 300 cc of xylene with heating. To this The solution is then added to a solution of 24 parts of phenylquinolinecarboxylic acid with stirring in 35o parts xylene. The compound crystallizes out in needles and is washed with water or washed out with a suitable organic solvent at 6o to 7o °.
