Description
So far, ergosterol was considered the only substance that can be converted into an irradiation product showing the antirachitic effect of the D vitamin by treatment with activating light. It has been found that the Lumisterin occurring as a by-product during the activation of ergosterol (cf. Ann. 492, p. 229, lines 4 to 8 and footnote and Chem. Zeitung 1931, p. 956, right column, 2nd paragraph, line 7 to 11), which in itself is completely ineffective antirachitic, 'can be converted into an antirachitic product by treatment with activating rays, in particular from the ultraviolet part of the spectrum. The irradiation product of Lumisterine can be like that of Ergosterin z. B. win geao on the way via the dinitrobenzoate in crystallized form. The purified irradiation product proves to be identical in all chemical, physical and physiological properties to the antirachitic conversion product of ergosterol described in the literature under the name vitamin D 2.
Like the Lumisterin itself, the esters of the Lumisterine, ζ. B. the lumisteryl acetate, enableable.
Um sie gegen Luftoxydation zu schützen, nimmt man sie zweckmäßig in an sich bekannter Weise in einem Pflanzenöl auf.ι g Lumisterin is dissolved in a quartz hollow roller in 50 ecm peroxide-free ether and irradiated with a magnesium 3S spark gap for 1 hour with the absolute exclusion of air oxygen. After the ether has evaporated, an oily substance remains. This also shows the absorption characteristic of the antirachitic conversion product of ergosterol, with a pronounced maximum at 265 m «. It is highly effective antirachitic. In order to protect them against air oxidation, they are expediently taken up in a vegetable oil in a manner known per se.
ι g Lumisterin wird in ätherischer Lösung,ι g Lumisterin is in an essential solution,
The oily substance obtained after evaporation of the ether is dissolved in hot absolute pyridine, heated with a solution of 1 g ~ 3,5-dinitrobenzoyl chloride on a water bath for 20 minutes and then left to stand for several hours at room temperature. When the solution is sprayed with water, an oil separates which is absorbed in ether. The ethereal solution, carefully washed with water, is evaporated, the residue is dissolved in acetone and allowed to crystallize. After a short time a yellow substance crystallizes out, which melts after recrystallization at 147 0th
Their optical rotation in acetone solution is a ™ = + 1O 3> 7 ° · The new product thus has the same melting point and the same rotation as the 3, 5-dinitro-
benzoat des Vitamin D2 genannten Bestrahlungsproduktes des Ergosterins. Der Mischschmelzpunkt zeigt keine Schmelzpunkterniedrigung. Benzoate of vitamin D 2 called radiation product of ergosterol. The mixed melting point shows no lowering of the melting point.
Danach hat das antirachitisch wirksame Bestrahlungsprodukt des LumisteTo convert the dinitrobenzoate into pure vitamin D, its solution in 5% methyl alcoholic potassium hydroxide solution is boiled on a water bath for ro minutes and then left to stand for 24 hours at room temperature. Then it is evaporated to a small volume at room temperature in a vacuum and, after adding water, shakes out with petroleum ether. The petroleum ether solution a crystalline residue which is recrystallized ° / 0 ethyl methyl alcohol of 95 and needles of melting point provides ii6 ° leaves behind after evaporation. The specific rotation capacity is 0 ^ = 81.4 ° (geao measure in · acetone solution). The absorption spectrum shows a pronounced maximum at 265 mw. After that, the anti-rachitic radiation product of the Lumiste has
Die Ausbeute an reinem Vitamin aus dem oben beschriebenen 30 Diriitrobenzoat nähert sich dem berechneten Wert.rine the same melting point. the same rotation and the same absorption maximum as the crystallized vitamin D 2 obtained by irradiating ergosterol. The mixed melting point shows no lowering of the melting point. The yield of pure vitamin from the diriitrobenzoate described above approaches the calculated value.
