Verfahren zur Gewinnung von sexualhormonhaltigen Extrakten aus den
Exkreten nicht säugender Warmblüter, insbesondere aus Guano Es ist bekannt, daß
aus Faeces von Säugetieren Hormone gewonnen werden können. Es wurde nun gefunden,
daß sich auch in den Exkreten nicht säugender Warmblüter, wie im Guano, Stoffe befinden,
welche mit organischen Lösungsmitteln extrahierbar sind. Diese Extrakte enthalten
alle die Bestandteile, welche im allgemeinen die Hormone im tierischen Organismus
zu begleiten pflegen. Neben den Neutralfetten wurden Fettsäuren, Sterine, Sterinester
gefunden. Wenn man nach den üblichen Verfahren diese bekannten Stoffe aus den Extrakten
entfernt, hinterbleibt ein Rohöl, welches das männliche und das weibliche Geschlechtshormon
enthält. Beispiel i 200g gesiebter Peru-Guano werden im Extraktionsapparat erschöpfend
mit Benzin extrahiert. Der Rückstand beträgt etwa i,5% vom Guano. Durch Nachverseifung
mit alkoholischem Alkali und nachfolgender Ätherextraktion entfernt man die Fettsäuren.
Der Ätherextrakt wird mit Wasser gewaschen, abermals eingedampft, und der Rückstand
wird mit Alkohol oder Aceton gelöst. Die filtrierte Lösung wird durch Ausfrierenlassen
von den kristallisierenden Sterinen befreit. Nach Abdestillieren des Lösungsmittels
hinterbleibt ein Rohöl, welches als Ausgangsmaterial für Hormonpräparate dienen
soll. Beispiel 2 5oog Guano werden mit 51 3%iger Natronlauge in der Wärme etwa 6
Stunden aufgeschlossen. Die erkaltete alkalische Lösung wird in einem Extraktionsapparat
mit Äther erschöpfend extrahiert (neutraler Extrakt). Dann wird unter Ansäuern der
alkalischen Lösung mit Äther weiter extrahiert (saurer Extrakt). Neutraler und saurer
Extrakt werden getrennt oder gemeinsam ,in der unter Beispiel i beschriebenen Art
weiter bis zum Rohöl verarbeitet. Beispiel 3 i 5o g rohe Harnsäure aus Guano werden
mit Alkohol in der Wärme erschöpfend extrahiert. Dieser Alkoholextrakt gibt einen
dunkelbraunen Rückstand, welcher nach Beispiel i oder Beispiel 2 über den Ätherextrakt
zum Rohöl verarbeitet wird. Beispiel ¢ 2 kg frische Hühnerexkrete werden
mit Äthylalkohol erschöpfend extrahiert. Der alkoholische Extrakt wird auf etwa
i l eingeengt, und diese Lösung wird nach Zusatz von etwa 400 9 frisch gelöschtem
Kalk unter Rückfluß auf dem Dampfbad gekocht und dann heiß filtriert. Die abfiltrierten
Kalkseifen werden noch einige Male mit gooloigem
Alkohol :extrahiert.
Die vereinigten alkoholischen Filtrate werden eingeengt, und der wäßrige Rückstand
wird erschöpfend mit Äther extrahiert. Die mit Wasser gewaschene und dann getrocknete
Ätherlösung hinterläßt nach dem Verdampfen einen öligen Rückstand, das sogenannte
Rohöl, m einer Menge von 2,o bis 2,2 g. Dieses Rohöl wird dann nach den bekannten
Trennungsmethoden (vgl. z. B. Zeitschr. f. physiol. Chemie i 9 i, 1930,

ff. und Zeitschr. f. physiol. Chemie i 99, 1931, 243 ff.) weiter gereinigt. Beispiel
5 5 kg frische Geflügelexkrete werden mit 25 kg einer 5%igen wäßrigen Natronlauge
bei 8o bis 9o° innerhalb etwa 6 Stunden verseift. Die warme Lösung wird bis zur
kongosauren Reaktion mit Salzsäure angesäuert und nach- dem Erkalten im Apparat
erschöpfend mit Äther extrahiert. Der so erhaltene Ätherextrakt wird durch Waschen
mit verdünntem, wäßrigem Alkali von der Hauptmenge der Fettsäuren befreit. Die weitere
Reinigung des Ätherrückstandes geschieht durch fraktionierte Ausschüttelung seiner
wäßrigen, alkoholischen Lösung mit Petroläther. Die alkoholische Lösung enthält
die angereicherte Hormonfraktion.Process for obtaining extracts containing sex hormones from the excretions of non-lactating warm-blooded animals, in particular from guano. The cooled alkaline solution is exhaustively extracted in an extraction apparatus with ether (neutral extract). Then the alkaline solution is further extracted with ether (acidic extract). Neutral and acidic extract are processed separately or together, in the manner described under example i, further down to crude oil. EXAMPLE 3 150 g of crude uric acid from guano are exhaustively extracted with hot alcohol. This alcohol extract gives a dark brown residue, which is processed into crude oil via the ether extract according to example i or example 2. Example ¢ 2 kg of fresh chicken excreta are exhaustively extracted with ethyl alcohol. The alcoholic extract is concentrated to about 1 liter, and this solution, after adding about 400 g of freshly slaked lime, is refluxed on the steam bath and then filtered hot. The filtered lime soaps are extracted a few more times with gooloigem alcohol: The combined alcoholic filtrates are concentrated and the aqueous residue is extracted exhaustively with ether. Example 5 5 kg of fresh poultry excreta are saponified with 25 kg of a 5% strength aqueous sodium hydroxide solution at 80 ° to 90 ° within about 6 hours. The warm solution is acidified with hydrochloric acid until it has a Congo acid reaction and, after cooling in the apparatus, is exhaustively extracted with ether. The ether extract obtained in this way is freed from most of the fatty acids by washing with dilute, aqueous alkali. The ether residue is further purified by fractional shaking out of its aqueous, alcoholic solution with petroleum ether. The alcoholic solution contains the enriched hormone fraction.
Beispiel 6 3 kg frische Hühnerfaeces werden mit der gleichen
Menge frisch gelöschtem Kalk gut gemischt. Das ganze wird in der Wärme mit 85%igem
Methylalkohol erschöpfend extrahiert. Der Rückstand vom Methylalkohol wird mit Wasser
und Äther aufgenommen. Aus der Ätherphase gewinnt man als erste Hormonphase das
