Description
Verfahren zur Herstellung von Impfstoffen aus menschlichen Tuberkelbazillen oder Rindertuberkelbazillen zur Schutzimpfung gegen Tuberkulose. Die Erfindung betrifft ein Verfahren zur Herstellung von Impfstoffen aus menschlichen oder Rindertuberkelbazillen, bei denen lebende Tuberkelbazillen so lange verrieben werden, bis ein Präparat entsteht, das lebend aufgeschlossenes Bakterienprotoplasma in Verbindung mit einer geringen Zahl lebender Tuberkelbazillen enthält. Die bisher bekannten Verfahren zur Herstellung von Präparaten aus Tuberkelbazillen gingen von dem Gedanken aus, entweder lebende, völlig avirulente und andersartige Bazillen zu verwenden (z. B. zur Schutzimpfung von Rindern menschliche Tuberkelbazillen), oder Präparate aus zerriebenen Bazillen zu gewinnen, wobei die Zerreibung so weit getrieben wurde, daß in dem Endprodukt kein lebender Bazillus mehr nachweisbar war. Um ganz sicher zu gehen, wurden sogar diesen letzteren Präparaten noch besondere Sterilisationsmittel (Carbolsäure) zugesetzt. Im Gegensatz hierzu werden bei meiner Erfindüng die lebenden Tuberkelbazillen nur so weit verrieben, daß im Endprodukt stets noch eine gewisse Anzahl von ihnen im lebenden Zustand enthalten sind. Das neue Verfahren wird in der Weise ausgeübt, daß im. Achatmörser mit der Hand die Tuberkelbazillenmasse verrieben wird, wobei durch mikroskopische Untersuchung der verriebenen Masse von Zeit zu Zeit geprüft werden muß, ob und inwieweit das Endziel erreicht ist. Der Zeitpunkt des Rufhörens -richtet sich nach den zur Verwendung kommenden Kulturen. . Durch Untersuchungen (s. S e 1 t e r »Die Immunitätsverhältnisse bei Meerschweinchentuberkulose«, Zeitschr. f. Hygiene, Bd.95, S. 159 und »über die Wirkung abgetöteter Tuberkelbazillen«, Zeitschrift f. Hygiene, Bd. 95, S. 233) wurde bewiesen, daß die beobachtete Wirkung nur eintritt, wenn das lebend aufgeschlossene Protoplasma in Verbindung mit einer bestimmten Anzahl lebender Tuberkelbazillen zur Anwendung kommt, da weder das Protoplasma noch die in dem Präparat vorhandene Anzahl lebender Bazillen für sich allein den gewünschten Erfolg hervorbringen. Zur Erfindung gehört weiter, daß zur Schutzimpfung nur arteigene Tuberkelbazillen mit genügender Virulenz benutzt werden, so zur Schutzimpfung der Rinder Rindertuberkelbazillen, zur Schutzimpfung der Menschen menschliche Tuberkelbazillen.Process for the preparation of vaccines from human tuberculosis bacilli or bovine tubercle bacilli for protection against tuberculosis. The invention relates to a method for the production of vaccines from human or bovine tubercle bacilli, in which live tubercle bacilli are ground until a preparation is obtained which contains living disrupted bacterial protoplasm in connection with a small number of live tubercle bacilli. The previously known processes for the production of preparations from tubercle bacilli were based on the idea of either using live, completely avirulent and different types of bacilli (e.g. human tubercle bacilli for the vaccination of cattle), or to obtain preparations from ground bacilli by trituration so far that no living bacillus was detectable in the end product. To be on the safe side, special sterilizing agents (carbolic acid) were even added to these latter preparations. In contrast to this, in my invention the living tubercle bacilli are only ground so far that the end product always contains a certain number of them in the living state. The new procedure is carried out in such a way that im. In an agate mortar, the mass of tubercle bacilli is rubbed by hand, whereby a microscopic examination of the rubbed mass must be checked from time to time to see whether and to what extent the end goal has been achieved. Hygiene, vol. 95, p. 233 ) it has been proven that the observed effect only occurs when the live, disrupted protoplasm is used in conjunction with a certain number of live tubercle bacilli, since neither the protoplasm nor the number of living bacilli present in the preparation alone produce the desired result. The invention also includes the fact that only native tubercle bacilli with sufficient virulence are used for the vaccination, for example bovine tubercle bacilli for the vaccination of cattle and human tubercle bacilli for the vaccination of humans.
