Description
Process for the production of stable hydrochloric acid trypsin preparations. It is known that pepsin-hydrochloric acid renders trypsin ineffective; this phenomenon also occurs gradually. by hydrochloric acid alone (cf. Hanimarsten, Physiologische Chemie. 1907, p. 39i). However, according to recent experiments, the destructive effect of free hydrochloric acid is not so strong that the digestive power of the trypsin given per os would be completely destroyed after passage through the stomach (Long and Hull, Journ; Amer. Chemical Soc. 39, 14_g3 to 1500 ). From the well-known fact that trypsin is ineffective even in a very weakly acidic solution, it does not follow that it is destroyed by small amounts of acid. Rather, after being exposed to a moderate acidic effect for a short time, it can digest protein after producing a neutral or alkaline reaction.
In fact, experiments have confirmed that the digestive power is weakened of trypsin by the hydrochloric acid of gastric juice both in the case of trypsin itself as well as 'partly happened to me in his acid-insoluble yannates.
It would be desirable to have a permanent preparation which the effect of larger hydrochloric acid or hydrochloric acid 1'ellsin quantities with that of trypsin, as in many gastric and intestinal diseases one Simultaneous administration of hydrochloric acid or hydrochloric acid-pepsin and Trvpsin indicated is.
It has now been found that such preparations can be made if betaine chlorohydrate or betaine chlorohydrate pepsin with a dry trypsin preparation, such as B. the Pancreatinum activum or absolutum of the trade or its tannic acid compound mixed. Mixtures of this type can be used in any desired proportions produce and retain their tryptic effect even when stored for a longer period of time, while such retention of an aqueous hydrochloric acid in therapeutic. usable Amount containing trypsin preparation is excluded.
The almost complete shelf life seems to be based on the fact that the strongly acidic, solid bctaine chlorohydrate particles in the powder mixture are only allowed to act on the acid-sensitive trypsin particles at the touch points and therefore the vast majority of the trypsin, as long as the mixture is kept completely dry, the otherwise long-term damaging effect of the hydrochloric acid of the betaine chlorohydrate remains withdrawn. The stability of the new trypsin preparations was demonstrated by a series of tests that extended over a period of time. The preparation was examined after 5 months according to the method of Fuld-GroB (`Vohlgemuth, plan of the fermentation method. Igi3, p. 187). The test was carried out as follows: 0.6 g of the new preparation corresponding to o, ig pancreatinum activum were dissolved in 20 ccxn water and, for the purpose of neutralization, 1.3 cc of double normal sodium hydroxide solution, corresponding to the 0.4 g of betaine chlorohydrate, was added. From the solution made to 50 cc with water, 1 cc was removed for testing. The digestion sample of the freshly prepared preparation and the one stored for 5 months gave the same result, so the digestive power of panlreatine remained unchanged.
