Verfahren zur Darstellung von Dinitrodiphenylmerkuridicarbonsäuren.Process for the preparation of dinitrodiphenylmerkuridicarboxylic acids.
Zusatz zum Patent 249725 vom 4. Dezember 1910.Addendum to patent 249725 from December 4, 1910.
Patentiert im Deutschen Reiche vom 28. Mai 1911 ab. Längste Dauer: 3. Dezember 1925.Patented in the German Empire on May 28, 1911. Longest duration: December 3, 1925.
The patent 249725 relates to a process for the preparation of diamidodiphenylmerkuridicarboxylic acids. The mercury compound of the nitrobenzoic acids is first prepared and then subjected to reduction to the diamidophenyl mercuridicarboxylic acids. It has now been shown that too the gradual reduction of mercury nitrobenzoic acids using two Atoms of hydrogen on two molecules of nitro compound succeeds, taking with deposition Dinitrodiphenylmerkuridicarbonsäuren arise from mercury. This process is very strange, since it was to be expected that the nitro group would first be replaced by reducing agents would be attacked. The escape of mercury bound in the core instead of a reduction the nitro group is surprising. The properties of the obtained dinitrodiphenylmerkuridicarboxylic acids are unexpected. The salts of the mercury compounds of nitrobenzoic acids used as starting material are highly poisonous, so animals can only be injected with very small doses. Against are the Dinitrodiphenylmerkuri dicarboxylic acids (or their salts, ζ. B. the sodium salt) of essential less toxicity, while the therapeutic effectiveness is extraordinarily great. 1.5 g of the sodium salt can be injected subcutaneously into a rabbit without the animal shows even the slightest signs of poisoning, although the mercury content is 0.5 g amounts to. The non-toxicity is all the more striking, as nitro compounds are for him Are generally harmful to the organism. Despite being non-toxic, the salts of dinitrodiphenyl mercuridicarboxylic acids has a strong effect on spirochetes and spirils, so that a single injection is sufficient to or to heal spirochete animals.
Bei dem Zusatz von Zinnchlorür scheidet sich sofort das Quecksilber in grauen Flocken aus, die beim Erwärmen kompakter werden, so daß sie sich dann leicht filtrieren lassen.115 g of the mercury compound of p-nitrobenzoic acid are dissolved in about I 1 Z 2 1 of water with sodium hydroxide solution and an alkaline solution of 100 to 150 g of tin chloride is added in the cold or in the warm until a sample is filtered off further addition of tin chloride prevents mercury from precipitating. The entire reaction product is then advantageously heated on the steam bath for some time. When tin chloride is added, the mercury immediately separates out in gray flakes, which become more compact when heated, so that they can then be easily filtered.
B. Salzsäure an. Das zuerst ausgefällte Zinnoxyd geht als Chlorid in Lösung, wäh-Then the precipitated mercury is filtered off and the yellow solution is acidified z. B. hydrochloric acid. The tin oxide precipitated first goes into solution as chloride, while
After filtering, you can remove the acid Dissolve traces of tin, which it may still contain, in soda. The tin oxide then remains unsolved. If you then precipitate the clear yellow solution with hydrochloric acid or acetic acid, the pure p-dinitrodiphenyl mercuridicarboxylic acid is obtained in this way as canary yellow,
ίο viel Wasser enthaltenden, schleimigen Niederschlag. After drying on clay or at 100 °, the acid forms a dark yellow colored, solid powder that does not dissolve in water, alcohol or organic solvents, such as benzene, toluene, gasoline, ethyl acetate, etc., dissolves. The acid is also insoluble in mineral acids. It differs in this from the diamino compound of the main patent. She solves but can easily be found in alkalis such as caustic soda, potassium hydroxide, ammonia. These solutions are colored yellow. If possible concentrated dissolution of the acid in z. B. caustic soda with alcohol, the disodium salt of the acid turns out to be yellow, flaky Powder like. The same is pure and corresponds to the following after drying at 100 ° Formula, as the analysis shows:

Alle diese Salze sind in der Kälte gefällt gelatinös, ballen sich aber beim Erwärmen zu flockigenFrom the solution of the sodium salt the dinitrodiphenylmercuridicarboxylic acid naturally falls off again easily with acid the end. The solution of the sodium salt and almost all metal salt solutions give precipitates which are differently colored are. This is how the yellow-colored silver salt is created with silver nitrate, and that with barium chloride yellow colored barium salt, with nickel chloride the yellow-green colored nickel salt, with cobalt potassium sulfate the gray-pink colored cobalt salt, with iron chloride the red-brown iron salt. All These salts are gelatinous in the cold, but clump together to form flakes when heated

The free acid behaves in a very similar way, and it does not melt, but rather when it is warmed up decomposed under deflagration with phenomena of fire. In contrast to the Sodium salt has the smell of nitrobenzene.
In exactly the same way, the 0- and m-dinitrodiphenyl mercuridicarboxylic acids become manufactured. Chemically, the appearance and properties are identical.

Dasselbe hat nach der Analyse folgende Zusammensetzung :200 g of the mercury compound of m-nitrobenzoic acid are mixed with about 2 liters of water Sodium hydroxide solution dissolved. About 150 g of 15 percent sodium hydroxide solution are required for this. In the solution which contains the sodium salt of the mercury compound of m-nitrobenzoic acid, add about 9 g of aluminum shavings and let them work while stirring for so long until a filtered sample of the solution with alkaline tin chloride solution does not show any mercury can fail more. There is then aluminum amalgam at the bottom of the vessel contaminated with alumina flakes. The solution is then advantageously filtered and acidified with hydrochloric acid. The m-Dinitrodiphenylmerkuridicarbonsäure then falls in yellow, voluminous flakes that are difficult to filter. After suction and Washing with water, the acid still holds back a lot of water. After drying on clay or at 100 ° the acid is a yellow, solid powder that dissolves in water and organic Solvents such as alcohol, ether, acetone, benzene, etc., does not dissolve. Hot hydrochloric acid dissolves Traces on. The acid dissolves easily in alkalis and ammonia. From the hot concentrated solutions, the corresponding salt precipitates on cooling, z. B. with caustic soda the disodium salt of m-Dinitrodiphenylmerkuridicarbonsäure falls as indistinct crystalline yellow powder. According to the analysis, it has the following composition :
NO,NO,
CO2NaCO 2 Na
CO2NaCO 2 Na
The sodium salt does not melt when heated, but instead evaporates with phenomena of fire. Here, mercury is deposited. The solution of the sodium salt gives with metal salt solutions very similar precipitates as the p-compound. The silver salt is yellow, as is the barium salt: 'The nickel salt is yellow-green, the iron salt is red-brown.
