The invention relates to a method for producing a technetium-99m generator, in which a pretreated chromatographic aluminum oxide column with a solution of from cleavage product Moiybden-99 Molybdate ions formed at a pH value of 3 to 4 brought into contact and the loaded column to Removal of contaminant ions is washed with a physiological saline solution.
The radionuclide technetium-99m wins because of its organ specificity and the minimal physiological Risks associated with its short half-life are increasing in importance on the Field of nuclear medicine. The most common form of Technetium-99m for diagnostic purposes consists of a solution of sodium pertechnetate in sterile, pyrogen-free physiological saline solution.
The sodium pertechnetate solution is generally used because of its short half-life of 6 hours made as soon as it is needed by eluting a technetium generator which is essentially is a chromatographic column on which molybdenum-99 is adsorbed in the form of molybdate ions is. The substrate generally used for the column is alumina. According to the usual way of working such generators for the adsorption of molybdenum are activated by the fact that the aluminum oxide is first brought into contact with dilute hydrochloric or nitric acid with a pH of 1.1. A sodium molybdate solution that contains molybdenum-99 and has a pH of approximately 3 to 5 is then used passed through the column, which adsorbs the molybdate ions from the solution. The loaded column is then washed successively with dilute acid and physiological saline solution to add molybdenum to the column to "fix" and remove unwanted ions. With an improved fixing wash, a Acetate solution is used as described in more detail in U.S. Patent Application Serial No. 1,77,269 (pending on September 2, 1971).
Usually technetium generators are mi? Molybdenum-99 was charged by neutron bombardment obtained from molybdenum-98. A smaller part of the molybdenum-98 is made by a Neutron capture converted into its radioisotope molybdenum-99. The radioactive molybdenum produced in this way is hereinafter referred to as "neutron product" for short.
Molybdenum-99 can also be obtained in the form of a fission product from neutron bombardment of uranium-235. This "fission product" is available in the form of sodium molybdate solutions with a much higher specific radioactivity than the highest activity that can be obtained from a neutron product. When using this highly active fission product for charging, technetium generators can be produced which give sodium pertechnetate eluates with exceptionally high levels of radioactivity and which are therefore very suitable for certain medical applications. When such a generator is manufactured in the conventional manner, however, it has been found that the eluates obtained therefrom disadvantageously contain undesirably high levels of molybdenum-99 and aluminum ions. Because of the high specific activity of the cleavage product molybdenum-99 and its relatively long half-life, the molybdenum content of the eluate is of particular importance. High aluminum ion content also creates difficulties and the extent of aluminum "strike through" or elution can be exacerbated by the radialytic attack of the alumina resulting from the high level of specific radioactivity in the generator.
Attempts have been made in this area to "strike through" or elute Molybdenum-99 when eluting fission product generators by using a relatively high pH value (i.e. from 4 to 9) of the sodium molybdate solution as it is being fed to the column. For example, British Patent 12 65 769 indicates that this approach can be used to reduce the in molybdenum found in sodium pertechnetate eluate Has success. For a suitable column charging, however, the pH value should be between 3 and 4 and am best around 3.5. This pH is to achieve maximum loading of the column with Molybdenum-99 required. Therefore, according to the aforementioned British patent, a certain type Pre-treatment and post-treatment of the alumina substrate are required to make this substrate a pH value below 4 to be able to load successfully. However, so far there is no suggestion of any effective one Pre-treatment or post-treatment procedures have been made, and the teachings of the aforementioned British Patents are directed to the pH range of 4 to 9 at which, as indicated, such Treatment is not required.
Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur Herstellung eines Techneüum-99m-Generators zur Verfügung zu stellen, bei dem eine derartThe invention is based on the object of a method for producing a Techneüum-99m generator to make available in which such a
The pretreated chromatographic aluminum oxide column is brought into contact with a solution of molybdate ions formed from the fission product molybdenum-99, so that there is no risk of elution o of molybdenum-99 together with sodium pertechnetate when using the generator, but on the other hand because of the usability of a molybdate solution with a pH of 3 to 4 the greatest possible activity of Mo! ybdän-99 is obtained.
To solve this problem, the invention is based on a method for producing a technetium-99m generator, in which a pretreated chromatographic alumina column with a solution of Molybdate ions formed from the fission product molybdenum-99 are brought into contact at a pH value of 3 to 4 and the loaded column for removing contaminant ions with a physiological saline solution is washed off and suggests that the alumina column be activated by heat a temperature of 100 to 150 ° C, preferably by heating to about 120 ° C for about 20 minutes, is pretreated.
By the invention it is achieved that the heat-activated alumina column according to the invention Has high adsorption capacity for molybdate ions and does not contain molybdenum-99 in the eluate when using the Generator is discharged, even with a high molybdenum-99 charge, which is due to use a molybdate solution with a pH value of 3 to 4 is made possible.
The generators produced according to the invention are essentially similar in structure to those that have hitherto been commonly used, with the exception, however, that the use of the cleavage product Molybdenum-99 allows for much smaller dimensions of the column than the dimensions of a neutron product generator with appropriate capacity. If z. B. with a molybdate solution with a pH of 3 to 4 is charged, only 1 to 2 grams of alumina are required for a 100 millicurie generator running with the fission product molybdenum-99 is charged, while 20 g or more for a generator of the same Capacity required with the neutron product-molybdenum-99 is loaded. Chromatographic alumina of the same quality can be used for the Fission product generator as used for the neutron product generator.
The manufacture of fission product generators according to the invention is manufacturing in several respects similar to conventional neutron product generators. The main difference between the Methods of the invention and the methods heretofore used to manufacture neutron product generators lies in the activation of the aluminum oxide used for the generator column. When conventional acid activation techniques in the manufacture of a fission product molybdenum-99 / technetium-99m generator If used, substantial and undesirable amounts of molybdenum-99 are common eluted with the sodium pertechnetate when the generator is put into use. When on the other hand, alumina is used as it is, d. H. without prior activation, it is impossible to make molybdenum-99 with the desired high degree of activation.
It has now been found that the alumina is very effective for adsorbing molybdate through it Heat activation can be activated and that this type of activation results in subsequent »penetration« or elution of molybdenum-99 when the column is eluted. Such eluting will Avoided even with extremely high molybdenum-99 charges, which are given when the Column is charged with a molybdate solution with a pH value of 3 to 4.
Nach dem Verfahren der Erfindung wird das Aluminiumoxid relativ kurze Zeit bei einer TemperaturAccording to the method of the invention, the alumina is at one temperature for a relatively short time
After the treatment, the aluminum oxide is able to adsorb very high proportions of fission product molybdenum-99, while the molybdenum-99 content of the eluate is kept well below undesirable proportions. In practice, the molybdenum content of such eluates is below the limit of quantification when using methods which are used to specifically determine whether an eluate complies with current safety regulations.
ausgesetzt, in dem bzw. durch das dann dieThe degree of activation achieved by heating alumina is a function of the time the alumina has been exposed to a particular temperature. The heating of the column by Behar.delii same in the autoclave at a temperature of 121 ° C for 20 minutes is sufficient to achieve a satisfactory activation. If lower temperatures are used, the time must be increased somewhat for maximum activation. Temperatures above 150 ° C should be avoided because at temperatures around 160 ° C or above, dehydration of the alumina can occur, which has adverse effects on the chromatographic properties of the alumina.
To produce a generator, a glass column containing heat-activated aluminum oxide and a porous carrier material at its lower end is produced. A narrow layer of silica over the alumina is advantageous and any voids in the column are simply filled with glass wool. Conveniently, the alumina is added to the column prior to activation and the filled column is then placed in an autoclave or some other heating means in which or through which the
4«; Aktivierung stattfindet. Loading or loading the column is effected by making an acidic solution of sodium molybdate with a pH of 3.5, the Cleavage product containing molybdenum-99, is passed through the activated alumina column. Because of the high specific activity of the cleavage product is compared with the volume required to produce a Neutron product generator of appropriate capacity is required, a relatively small volume of the Solution used.
Das Waschen mit der Acetatlösung dient dazu, den Aluminiumionengehalt aufAfter the column is loaded with molybdenum-99, it is used to remove residual acid and others unwanted ions washed with physiological saline solution. Before washing with the physiological saline solution, the column can advantageously with an acetate solution with a pH value between 2.8 and 6.0, and preferably 4.0, as in the U.S. patent application Serial No. 1 77 268 is indicated. The meaning is, residual solution from the column after washing with the physiological saline solution through Directing air to suck through the column. When excess unadsorbed molybdate is not off removed from the column by suction, a radiolytically induced hydrolytic attack of the Alumina due to the very high level of radiation emitted in the after the process Generators manufactured according to the invention can be achieved. The following example explains the invention.
Six 100 millicury technetium generators were made Manufactured in the following way: From a certain amount of chromatographic alumina (Woelm) the abrasion was removed by the aluminum oxide on a sieve made of corrosion-resistant steel ij Brought with a mesh size of 0.074 mm and the retained portion is repeated with water Was washed until the protruding liquid wedge was clear.
A slurry containing about 1.25 g of washed ta alumina was placed in each of the 6 glass generator columns (10 mm outer diameter χ 65 mm) which contained a sintered glass disc fused to one end of the column and neck pieces connected to both ends were suitable for receiving normal rubber stoppers. A thin layer of silica was placed on top of the alumina and the remaining cavity in each column was filled with glass wool, previously boiled in a 10 volume percent hydrogen peroxide solution, was introduced into the necks at each end of the column.
Each generator was then kept in an autoclave at a temperature of 12-20 minutes, to heat-activate the alumina.
Each activated column was charged by passing a solution of fission product sodium molybdate, the molybdenum content of which was almost entirely composed of molybdenum-99, through hollow inlet and outlet needles which were driven through the upper and lower plugs of the generator column. For this purpose, the pH of a commercially available sodium molybdate 99 solution was adjusted to 3.5 ± 0.1 with hydrochloric acid and diluted so that a specific radioactivity of approximately 245 millicuries per m! was given. Each generator, when charged with 2 ml of this solution, with approximately 470 millicuries / g aluminum present, had an activity of approximately 100 millicuries 6 days thereafter. After adding the feed solution, each column was allowed to stand for 30 minutes.
Approximately 1 ml of pH 4.0 acetic acid solution was then added to each column this solution was introduced through the inlet needle and withdrawn through the outlet needle. If more than About 2 ml of acetic acid is added, as has been determined by the generator obtainable yield of technetium-99m adversely affected, without these disadvantages by advantages would be compensated. After washing with acetic acid, 10 ml of 0.9% sodium chloride solution was added. The residual solution was sucked off by passing 20 ml of air through the column. The needles were with a cover, and the generators were sterilized by once again at 121 ° C 20 Minutes were heated in the autoclave.
The generators produced according to the above description gave high yields with values of sodium pertechnetate that were almost theoretically expected when they were then eluted with physiological saline solution. The molybdenum content of the eluate was only 0.001 to 0.005 μαΜο / mCi Tc-99m. However, when an alumina column was activated by conventional acid treatment, the eluate contained 0.01 to 0.05 % Mo / mCi Tc-99m, or about ten times as much. The pH of the obtained eluates ranged from 5.0 to 5.3 and the aluminum ion content was less than 0.3 g / ml.
Generators with even greater activity can, of course, be made; H. by using of sodium molybdate solutions with greater specific activity. You can also wash with Acetic acid and the aspiration of the remaining wash solution are omitted if the sodium pertechnetate to be used for purposes where there is a higher content of aluminum ions in the eluate, such as z. B. a two to four times higher salary is not disadvantageous.
