Description
BEREICH DER ERFINDUNGFIELD OF THE INVENTION
The present disclosure relates to a system and composition for developing a transethosomal gel loaded with neem oil for a prolonged effect using phosphatidylcholine as the phospholipid and ethanol included in the formulation to give the vesicular system a unique identity and also contribute to increasing the lipid fluidity of the vesicles.
HINTERGRUND DER ERFINDUNGBACKGROUND OF THE INVENTION
Much attention has been paid in recent decades to the development of novel drug delivery systems (NDDS). The NDDS should ideally meet two requirements: first, it should deliver the active ingredient at a rate that suits the body's needs throughout the treatment, and second, it should deliver the active ingredient to the site of action. Novel drug delivery attempts to either maintain the effect of the drug at a certain rate or to maintain a relatively constant, effective level of the drug in the body while minimizing undesirable side effects.
Various novel vesicular systems have been developed for transdermal drug delivery, with TEs (TEs) being one of these vesicular systems. TEs are the new generation of ethosomal systems. This vesicular system contains the basic components of classic ethosomes such as phospholipids and ethanol with an additional compound, e.g. B. a penetration enhancer or an edge activator (surfactant) as a component. These novel vesicles were developed in an attempt to combine the advantages of classical ethosomes and deformable or elastic liposomes (transferosomes) into one formula to make TEs. This means that the TEs are superior to the classical ethosomes. Neem seed oil (Azadirachta indica) has been used for centuries in traditional Indian medicine to heal skin conditions such as eczema, psoriasis, rashes, acne, dry skin, burns and abrasions. Neem oil, with its natural antibacterial properties, has been used for centuries as a safe and gentle skin care agent.
In view of the foregoing, it is clear that a system and composition are required for the development of a neem-loaded transethosomal gel.

Charakterisierung der Formulierungen:Characterization of the formulations:
Charakterisierungen von TEs:Characterizations of TEs:
2) Analyse der Vesikelgröße und PDI2) Analysis of vesicle size and PDI
Vesicle size analysis of transethosomal formulations is performed with the Zetasizer (Malvern Instruments Ltd. UK). The analysis provides the mean vesicle size (z-average) and the polydispersity index (PDI) as a measure of the breadth of the distribution. The analysis is carried out after appropriate dilution with double-distilled water and subsequent filtration through Whatman filter paper.
3) TEM-Analyse von TES3) TEM analysis of TES
Schließlich wird die Probe auf dem Kupfergitter mit dem Transmissionselektronenmikroskop untersucht.TEM studies of the formulations are performed with the aim of analyzing the surface morphology of the TEs (Titan G2 60-300 KV TEM, Thermo Fisher Scientific, US). A drop of the diluted sample is placed on a copper grid and dried. The dried sample is then stained with 2% phosphotungstic acid. Finally, the sample on the copper grid is examined with the transmission electron microscope.
Charakterisierung von TEs gel:Characterization of TEs gel:
1) Bestimmung der Viskosität1) Determination of the viscosity
The viscosity of the gel is determined using a Brookfield viscometer (S-64, model LVDV-E) at 25°C and a spindle speed of 12 rpm.
2) Extrudierbarkeit2) extrudability
This test determines the amount of gel extruded from a tube at a constant weight. A closed tube containing 10 g of gel is pressed with a constant weight of 1 kg at the kinked end. When removing the cap, the gel extrudes until the pressure is released. The extruded gel is collected and weighed.
3) Verteilbarkeit3) distributability
Die Vergrößerung des Durchmessers aufgrund der Ausbreitung des Gels wird notiert.0.5 g of gel are placed in a 1 cm diameter circle pre-marked on a glass plate, over which the second plate is placed. A weight of 500 g is allowed to rest on the top plate for 5 minutes. The increase in diameter due to spreading of the gel is noted.
Der pH-Wert der Dispersion wird mit einem digitalen pH-Meter (pH-Meter, Mettler Toledo) gemessen.100 mg of the gel formulation is weighed and placed in a 50 mL volumetric flask. The volume is made up to 50 ml with distilled water. The pH of the dispersion is measured using a digital pH meter (pH meter, Mettler Toledo).
In-vitro-Freisetzungsstudie:In vitro release study:
In vitro drug release studies for optimized TEs gels are performed using a Franz diffusion cell with an effective area of 1.76 cm2 and a receptor chamber capacity of 25 ml. A dialysis membrane with a molecular weight cut-off of 1200 and a flat width of 25 mm is used for the study. The membrane is saturated with the medium of the receptor cell (phosphate buffer pH 5.5). The receptor compartment is filled with degassed receptor medium and the activated membrane placed on top of the receptor compartment. The donor compartment is placed over the membrane and the formulation equivalent to 10mL of neem oil (calculated based on entrapment efficiency) is applied and evenly distributed in the donor compartment. The receiver medium is kept at 33±1°C and stirred constantly with a magnetic stirrer at 400 rpm. A 1 mL sample aliquot is withdrawn at regular intervals, every hour for 8 hours. After each withdrawal, the receptor compartment is filled with 1 ml of fresh receptor medium. The aliquots taken are analyzed with a UV spectrophotometer (Shimadzu 800) at 222 nm for further investigations. The release study is performed in triplicate.
In
0.884 aufwiesen. The encapsulation efficiency results show that formulations F1, F2 and F3 have an encapsulation efficiency of more than 50% and were therefore used to determine the vesicle size and PDI. F1 had an average particle size of 116.1 nm with a PDI of 0.774, while formulations F2 and F3 had an average particle size of 252 nm and 990 nm and a PDI of 0.307 and 0.884, respectively.
Charakterisierung von TEs-Neemöl-GelCharacterization of TEs neem oil gel
Based on the evaluation parameters, F1 was selected as the optimized formulation as it exhibited acceptable entrapment efficiency, the smallest vesicle size, and resulted in the formation of vesicles with spherical morphology and well-defined border.
Die Viskosität der optimierten Formulierung beträgt ----cps, was im idealen Bereich für topische Gelformulierungen mit Carbopol als Geliermittel liegt [Kim].The viscosity of the formulations (F1 with different proportions of Carbopol gel) was measured with a Brookfield viscometer (S-64, model LVDV-E). The viscosity of the various formulations is shown in Table 2. The viscosity of the optimized formulation is ----cps, which is in the ideal range for topical gel formulations using Carbopol as the gelling agent [Kim].
Extrudierbarkeitextrudability
When examining the extrudability of a gel formulation, the force or pressure required to extrude the gel from the tube is measured. The more gel that is extruded, the better the quality, but too much of an amount is also undesirable as it results in wasting the formulation. The F1G3 formulation was selected for further study because of its ability to be extruded from the tube in the desired manner. The results of the extrudability study are presented in Table 2.
Verteilbarkeitdistributability
Spreadability studies are performed to determine the size of the area over which the gel spreads when applied to the skin. This is an essential property that the gel must have and also promotes patient compliance due to the aesthetic feel. The increase in diameter by spreading the best formulation is 6.7±0.08 cm. This value indicates good spreadability. Table 2 shows the spreadability results.
pH-MessungpH measurement
The pH measurement of the formulated gels showed that all formulations have a pH value that is suitable for topical application and also does not cause skin irritation.
The
