Abstract
Cosmetic or pharmaceutical composition with lamellar structures using long-chain sucrose fatty acid esters, adapted to the conventional recipe of the DAC base cream.
Description
Die neuartige Formulierung wird hier als modifizierte DAC-Creme aufgeführtThe subject of the present invention includes the revision of the DAC base cream that is often used in pharmacy, with all raw materials used now being of natural origin. Thanks to the use of PEG-free emulsifiers from the group of sucrose fatty acid esters, the novel formulation also has lamellar skin-repairing structures. The novel formulation is listed here as a modified DAC cream
The original base cream according to DAC 2011 is a popular dermatics base for the production of semi-solid preparations and is also suitable for preventive skin care. It is prescribed as a hydrophilic base, but it also has amphiphilic properties and is therefore easily miscible with both water and lipids. Since it is compatible with most ionic and non-ionic active ingredients, the NRF 2011 (New Formulation Formulation) contains 22 dermatological formulations based on the DAC cream.
The DAC base cream, which has been used for over 34 years, is characterized by the two points listed by numerous dermatologists: on the one hand, the recipe contains a small number of raw materials (water, white petrolatum, propylene glycol, medium-chain triglycerides,
However, time seems to have stood still with the pharmaceutical DAC base cream. In today's innovative cosmetic formulations, the ingredients used are as completely natural as possible and also biodegradable. Such formulations therefore have no PEG-containing components, no parabens, no silicones or microplastics, among other things. Furthermore, a new innovative base cream itself should also have a lasting positive effect on the skin.
The present invention is based on the object of developing a new base cream which consists of natural raw materials and which can be used both pharmaceutically and cosmetically as a cream base.
For example, the invention is intended to be replaced by plant-based skin-protecting and barrier-forming fats and waxes, on the one hand, for petroleum jelly (petroleum product), which reduces transepidermal water loss through the formation of a protective film on the skin. This task is solved, among other things, by using "Vaseline" on a plant basis such as "Pionier 38005" from Hansen & Rosenthal or "Vegeline 65" from Sophim. These products are free from mineral oil or synthetic ingredients. They have a moisturizing effect, sensory properties, prevent transepidermal water loss and make the skin feel smooth. The main component of both products is Ricinus Communis Seed Oil and Hydrogenated Castor Oil. Furthermore, other vegetable waxes, lipids and essential fatty acids used alone or in a mixture can act as vegetable vaseline, e.g. carnauba wax, candellilla wax, shea butter, jojoba wax, berry wax (Japan wax) and vegetable oils with a high content of omega fatty acids (e.g. evening primrose oil) and / or their hydrogenated (saturated fatty acids) form. The following waxes are popular in natural cosmetics: e.g. rice wax, rose flower wax or sunflower seed wax.
(http://www.dejayu.de/dms-creme-wie-man-eine-basiscreme-zur-individuellen-hautpflegenutzt/).
The DSM (Derma Structure Membrane) cream forms such a basis, which has lamellar structures similar to those of the skin lipids (patent
(http://www.dejayu.de/dms-creme-wie-man-eine-basiscreme-zur-individuellen-hautpflegenutzt/).
These formulations are mainly of interest for cosmetics and are implemented, for example, in the Physiogel from the company Stiefel.
The fact that the DMS creams have not yet achieved triumph in the mass market and pharmacies is due to the complex manufacturing method that includes high-pressure homogenization. This process is very energy-intensive and can only be carried out with special equipment, which is normally not available in pharmacies (H. Lautenschläger, Österreichische Apothekerzeitung, 56 (2002) 679).
For example, with regard to the invention, the PEG-containing O / W emulsifier is to be replaced by a natural O / W emulsifier which also has lamellar structures in the formulation. That is why emulsifiers from the group of sucrose fatty acid esters are mainly used in the new, modified DAC base cream.
So zeichnet sich die neue lamellare DAC-Basiscreme durch die Verwendung pflanzlicher Wachse und Fette und den Gebrauch von Emulgatoren der Familie der Saccharose-Fettsäurenester aus (siehe Beispiel 1 und 2).Sucrose tristearate, which has only a very small proportion of monoesters and, accordingly, a high proportion of di- and higher esters such as tri- and tetra-esters (e.g. from the companies Sisterna and Mitsubishi-Kagaku Foods Corporation) was selected. Due to the long-chain fatty acids, the sucrose stearate variant is a very skin-friendly and mild emulsifier that can also be used on damaged skin. The high water-binding capacity of the hydrophilic sugar head group leads to an increase in skin moisture and reduces the roughness of the skin. The new lamellar DAC base cream is characterized by the use of vegetable waxes and fats and the use of emulsifiers from the sucrose fatty acid ester family (see Examples 1 and 2).
UV-Filter.Cosmetic and / or pharmaceutical active ingredients, which can be both lipophilic and hydrophilic in nature, can be present as further components in the lamellar DAC formulation. Such substances are all active ingredients that can be incorporated into the lamellar formulation and have cosmetic and / or pharmaceutical effects in or on the skin, e.g. moisturizers such as urea and hyaluronic acid; Antioxidants such as vitamins and coenzyme Q10; anti-inflammatory agents such as incense; oxygenating substances and / or special lipids such as ceramides. Furthermore, substances can be used that protect the skin, such as UV filters.
Nur sehr wenige Wirkstoffe können die Hautbarriere - das Stratum corneum- überwinden und gelangen dann in die darunterliegenden Hautschichten und an den Wirkort. Das sind kleine Substanzen mit einem Molekulargewicht unter 500 Da und einen Verteilungskoeffizienten von log Pow von 1 bis 3 (Löslichkeit in Octanol/Wasser). Um Wirkstoffe aus diesen lamellaren Lipidstrukturen in die tieferen Hautschichten (Epidermis und Dermis) zu transportieren, eignen sich im besonderen Falle die Nanoemulsionen (NE) wie die Hydro-Tops (W/O/W-NE;
Diese neuheitliche Kombination aus der hier beschriebenen modifizierten lamellaren DAC-Creme und den aufgeführten Nanoemulsionen ermöglicht eine gezielte Behandlung auf und in der Haut.Very few active ingredients can overcome the skin barrier - the stratum corneum - and then reach the underlying skin layers and the site of action. These are small substances with a molecular weight below 500 Da and a distribution coefficient of log P ow from 1 to 3 (solubility in octanol / water). In order to transport active ingredients from these lamellar lipid structures into the deeper skin layers (epidermis and dermis), the nanoemulsions (NE) such as the hydro-tops (W / O / W-NE) are particularly suitable;
In particular, active ingredients such as water-soluble substances cannot pass the lipophilic skin barrier and are dependent on effective transport systems such as esculin (log P of -1). This water-soluble active ingredient from horse chestnut has a capillary-sealing effect in the case of weak veins and, when used in cosmetic products, protects against UV-B rays. Although lipophilic substances can penetrate the stratum corneum (lipophilic skin barrier), they are retained there due to their lipophilicity. In this way, they do not get into the living skin layers underneath (water content> 70%) without an effective carrier system such as the active ingredient Coenzyme Q10 (log P of> 10), an energy donor for the cells.
Less efficient results of the penetration of active ingredients into the living skin layers and the areas below were achieved with the aid of commercially available liposomes.
This innovative combination of the modified lamellar DAC cream described here and the listed nanoemulsions enables targeted treatment on and in the skin.
The simple and rapid production of such a formulation with lamellar structures also enables an inexpensive production that can be carried out by anyone and that can also be carried out in pharmacies.
Beispiel 1:Example 1:
Modifizierte DAC-Formulierung mit dermalen Strukturen (Pharnazeutische Anwendung)Modified DAC formulation with dermal structures (pharmaceutical application)
The modified DAC base cream was made in two steps. First, sucrose monostearate and cetyl alcohol were melted and homogenized together with the vegetable vaseline in a water bath at 60 ° C. The hydrophilic phase, consisting of water, propylene glycol and sucrose tristearate was also heated to 60 ° C. and additionally homogenized for two minutes using an Ultra-Turrax at 9,500 rpm. The hydrophilic phase was then added to the lipophilic phase and the cream was stirred until it was cold. After cooling, the carrier systems (nanoemulsions) were incorporated into the new modified DAC base cream with simple homogenization at 3500 rpm.

Herstellung wie in Beispiel 1Production as in example 1
Beispiel 3:Example 3:
