Abstract
Description
Die Erfindung betrifft ein Verfahren zur Herstellung von NDP-Zucker (Nucleosiddiphosphat-Zucker) die auch als Zucker-Nucleotide bezeichnet werden. Diese Verbin dungen sind wichtige Intermediate der Biosynthese von Glycokonjugaten. Ihre Synthese in größerem Maßstab ist somit sehr bedeutend zur in vitro Synthese von Oli gosacchariden, besonders im Hinblick auf mögliche Be deutungen von Oligosacchariden in medizinisch diagno stischen und therapeutischen Anwendungen.The invention relates to a method for manufacturing of NDP sugar (nucleoside diphosphate sugar) which too are referred to as sugar nucleotides. Es ist eine Vielzahl von derartigen Reaktionen bekannt, bei denen verschiedene Zucker-1-Phosphate mit unterschiedlichen Nucleosidtriphosphaten umgesetzt werden. So können die Triphosphate von Uridin (U), Guanosin (G), Cytidin (C) sowie Thymidin (T) jeweils mit den -1-phosphaten von Glucose, Galactose, Fucose und Mannose aber auch von N-Acetylglucosamin oder N-Acetylgalactosamin umgesetzt werden. Für die jeweiligen Reaktionen kommen jeweils diejenigen Pyrophosphorylasen zum Einsatz, die die Re aktion des jeweiligen Zucker-1-phosphates und des NTP's katalysieren. Diese sind dem Fachmann bekannt und kön nen durch moderne Klonierungs- und Expressionsmethoden erhalten werden. Durch Variation der Verweilzeit des Reaktionsgemisches kommt es zu unterschiedlichen Umsätzen und Raum-Zeit-Aus beuten. Die durch die vertikalen gestrichelten Lini en der Fig. 7 abgegrenzten Abschnitte stellen Ver suchsabschnitte dar in denen die Durchflußgeschwindig keit der Reaktionslösung, die umgekehrt proportional der Verweilzeit τ ist, als Parameter variiert wurde. Der Figur ist zu entnehmen, daß bei mit gleichen Ver weilzeiten charakterisierten Segmenten gleiche Umsätze erzielt werden. Die jeweils mit zunehmender Betriebs zeit des Durchflußreaktors absinkenden Umsätze sind auf natürliche Denaturierung der Pyrophosphorylase zu rückzuführen. Das Ansteigen des Umsatzes im Segment 2 der Fig. 7 ist lediglich auf ein Ansteigen der Tempe ratur während des Betriebs zurückzuführen.2.
