Description
Die Erfindung betrifft ein Verfähren und eine Vor richtung zur Erfassung von Oberflächenantigenen oder Strukturmerkmalen von Zellen, Partikeln oder Makromolekülen.The invention relates to a method and a direction for the detection of surface antigens or structural features of cells, particles or Macromolecules.

Dabei kann bei Bedarf eine Mehrfach markierung vorgenommen werden, z. B. indem man Anti körper oder Liganden unterschiedlicher Spezifität mit verschiedenen Fluorochromen koppelt, die bei An regung Fluoreszenzlicht unterschiedlicher Wellenlän gen erzeugen. Wird ein derartiger Reaktionsansatz in lichtdurchlässigen Reaktionsgefäßen einem flüssigen oder gelartigen Medium (z. B. aus Mikroglaskügelchen, Latexpartikeln, Dextran oder Zellulose) aufgeschich tet und anschließend zentrifugiert, so sedimentieren die Bestandteile im Medium gemäß ihren unterschied lichen physikalischen Eigenschaften und gemäß der Porengröße im Gel unterschiedlich schnell, so daß sich idealerweise eine bandenartige Auftrennung der Bestandteile abzeichnet. Die verschiedenen Banden repräsentieren z. B. unterschiedliche Zellen, die mit Ausnahme der roten Zellen keine Eigenfarbe besitzen und somit bei Tageslicht nicht sichtbar sind. Durch die spezifische Markierung von Oberflächenantigenen mit fluoreszenzmarkierten Antikörpern können alle Banden mit fluoreszenzmarkierten Bestandteilen (Zel len) gegenüber einer Lichtquelle mit fluoreszenz anregendem Spektrum sichtbar gemacht werden. Die Ablesung der Fluoreszenzbanden kann nach Abschluß der Zentrifugation visuell oder maschinell erfolgen. Dabei können die lichtdurchlässigen Reaktionsgefäße über einen Scanner mit einer beweglichen Optik, die einen punktförmigen monochromatischen Anregungs strahl erzeugt, und einen Fluoreszenzdetektor, der auf den jeweiligen Anregungsort im Reaktionsgefäß fokusiert ist, abgescannt werden. Die Fluoreszenz intensität über den Reaktionsgefäßen wird ortsauf lösend erfaßt. Alternativ kann das Fluoreszenzmuster mit dem CCD-Sensor einer Kamara erfaßt werden und über ein nachgeschaltetes Computerprogramm ausgewer tet werden. Eine weitere Detektionsmöglichkeit ist über eine Zentrifugalanalyse möglich. Dabei werden die Fluoreszenzintensitäten über den Reaktionsgefä ßen, die in radiärer Anordnung auf einem Rotor ange ordnet sind, mittels einer stationären oder radiär beweglichen Optik während der Zentrifugation orts- und/oder zeitaufgelöst registriert. Die Optik er zeugt dabei einen punktförmigen monochromatischen Anregungsstrahl, der auf das Medium innerhalb der lichtdurchlässigen Reaktionsgefäße ausgerichtet ist. Bei Zentrifugation der lichtdurchlässi gen Reaktionsgefäße z. B. in einer radiären Anordnung auf einem Rotor tritt eine Sedimentation und/oder Auftrennung der Bestandteile ein. Schon während der Zentrifugation treten die Bestandteile im Medium in Kontakt mit fluoreszenzmarkierten Antikörpern oder Liganden, sodaß eine Fluoreszenzmarkierung statt findet. Nach der Zentrifugation findet durch eine Lichtquelle mit einem fluoreszenzanregenden spektra len Bereich eine Fluoreszenzanregung der fluores zenzmarkierten Bestandteile in den lichtdurch lässigen Reaktionsgefäßen statt. Dabei tritt eine ortsaufgelöste Änderung der Fluoreszenzintensität im Medium auf, z. B. mit spezifischen Bandenmustern, die visuell ausgewertet werden können oder über einen Scanner mit einer beweglichen Optik, die einen punktförmigen monochromatischen Anregungsstrahl erzeugt und einen Fluoreszenzdetektor beinhaltet, der auf den jeweiligen Anregungsort im Medium fokusiert ist, abgescannt werden. Alternativ kann das Fluoreszenzmuster mit dem CCD-Sensor einer Kamara erfaßt werden und nach der Digitalisierung über ein nachgeschaltetes Computerprogramm ausgewer tet werden. Neben einer Auswertung der ortsaufge lösten Fluoreszenzintensität nach Abschluß der Zen trifugation ist auch eine Auswertung während der Zentrifugation über eine Zentrifugalanalyse möglich. Dabei werden die Fluoreszenzintensitäten über den Reaktionsgefäßen, die in radiärer Anordnung auf ei nem Rotor angeordnet sind, mittels einer stationären oder radiär beweglichen Optik während der Zentrifu gation orts- und/oder zeitaufgelöst registriert. Die Optik erzeugt dabei einen punktförmigen monochro matischen Anregungsstrahl, der auf das Medium inner halb der lichtdurchlässigen Reaktionsgefäße ausge richtet ist. Trifft der Strahl auf fluoreszenzmar kierte Bestandteile, wird eine ungerichtete Fluores zenzstrahlung erzeugt, die durch einen Fluoreszenz detektor erfaßt wird, der auf den jeweiligen Anre gungsort im Reaktionsgefäß fokusiert ist. Dabei ist gewährleistet, daß während der Zentrifugation je weils innerhalb einer Rotorumdrehung die Fluores zenzintensitätswerte in allen lichtdurchlässigen Reaktionsgefäßen erfaßt werden und daß bei radiärer Bewegung der Optik eine orts- und zeitaufgelöste Messung der Fluoreszenzintensitäten über den Reak tionsgefäßen erfolgt. Die Steuerung des Rotors, der radiären Bewegung der Optik und die Meßwertaufnahme erfolgt über eine Schnittstelle durch eine Zentral einheit. Im mittleren Reaktionsgefäß ist die Situation zu Beginn der Zentrifugation dargestellt.
