Pharmazeut.ι · The two most important for digestion of the stomach Substances are known to be pepsin and hydrochloric acid. One has therefore tried many times, To produce preparations which combine these two substances; but the attempts aimed at this have so far only given unsatisfactory results. ' It. Has namely shown (see pharmacist.

A process has now been found which allows completely stable pepsin salt ^ to produce acid preparations with any acid content. That . Procedure consists in that pepsin is processed with betaine chlorohydrate instead of aqueous hydrochloric acid.
It is based on the observation that betaine chlorohydrate, which has not yet found any commercial use, has the property of being very strongly hydrolyzed by water, so that aqueous solutions of the salt act like free hydrochloric acid. Compared to other strongly hydrolyzable chlorohydrates of organic bases, betaine chlorohydrate has the advantage that the base it contains is completely non-toxic and has a very pleasant, more fruity than mineral-acid taste. The non-toxicity of betaine has been confirmed by extensive clinical trials. After monthly administration of betaine chlorohydrate to stomach sufferers the slightest side effect never occurred, while the therapeutic success was the same as with corresponding doses of hydrochloric acid. It has now also been demonstrated that pepsin becomes just as easily soluble in water through the addition of a small amount of betaine chlorohydrate as through the action of hydrochloric acid, and that solutions of betaine chlorohydrate and pepsin are able to digest protein just like hydrochloric acid-pepsin solutions.
The technical progress now lies in the fact that the betaine chlorohydrate, a beautiful crystallizing, air-resistant substance, mix in all proportions with pepsin and can be rubbed in, with or without the addition of other substances, and that the so produced Preparations can be stored completely undecomposed for any time.
Folgender Versuch erweist die technische Bedeutung der Erfindung:The following experiment proves the technical importance of the invention:
Das Betainchlorhydrat-There were (1.05 g of hydrochloric acid of 38 percent), in the other mixed 10 g pepsin with 6 '/ 4 percent H Cl even in the form of aqueous hydrochloric acid in the form of Betainchlorhydrat (2.6 g), and both preparations in stored in well-closed vessels. After four weeks of storage, the preparation made with aqueous hydrochloric acid had turned deep brown. The betaine chlorohydrate
The preparation was completely colorless and remained unchanged. Digestion experiments were carried out with 10 g of protein each. Addition of the amounts corresponding to 0.2 g of pepsin both preparations carried out in the usual way; it was found that the hydrochloric acid-pepsin preparation was only able to digest a small part of the protein, while the greater part remained unaffected. The betaine chlorohydrate pepsin, on the other hand, showed exactly the same digestive effect as fresh prepared pepsin hydrochloric acid and brought all the protein in a few hours Solution.
To carry out the process of the present invention, pepsin and betainchlo.rhydrate are ground together in the ratio which corresponds to the desired hydrochloric acid content of the finished product. Since betaine chlorohydrate ■. contains approximately 25 percent HCl , z. B. to produce a pepsin-hydrochloric acid preparation containing 10 percent HCl , 4 parts of betaine chlorohydrate with 6 parts of pepsin. A preparation which, on the other hand , should contain as 20 percent HCl and 20 percent pepsin is obtained by rubbing together 8 parts betaine chlorohydrate and 2 parts pepsin.
A very small addition of betaine chlorohydrate is sufficient to produce water-soluble pepsin; a flawless preparation is obtained by intimately mixing, for example, 97Y 2 parts of pepsin with 2 1 and 2 parts of betaine chlorohydrate.
.The mixtures of pepsin and betaine chlorohydrate, which one other suitable powdery substances, such as. B. lactose, can be easily pressed into tablet form or into pills, coated tablets, etc. to process. .
