Arzneimitteldrug
The subject of the invention is the elimination of side effects occurring with normal administration in the case of orticosteroids (e.g. glycosteroids), which have proven to be valuable drugs in various diseases. the Elimination of these side effects, which are usual with normal administration, is achieved by simultaneously with the corticosteroid or shortly before or after, an effective amount of beta (2- or 4-pyridyl-alkyl) amine or administered a substance with a similar effect. It has been shown that - for reasons not yet completely have been clarified, but for which some theoretical explanation has been found - compounds such as beta (2- or 4-pyridil-alkyl) amines cause the corticosteroid as with normal administration and contributes to the relief of the disease, but without the known side effects appear.
Das häufige AuftretenThe use of corticosteroids for many purposes, such as acute inflammation and allergic diseases of the Eyes, skin, mucous membrane, rheumatoid arthritis, bronchial agthma, ulcerative colitis and other diseases, the complete list of which would lead too far here has increased considerably in recent years. The frequent occurrence

Attempts have also been made to reduce the known undesirable side effects by developing various new steroid derivatives. However, since these new steroid derivatives led in some cases to an increased activity per molecule, the Efficacy per biological unit in relation to the biological basis essentially parallel to the side effects. So it has so far not been possible to eliminate or at least reduce the undesirable Side effects of the corticosteroids to find a satisfactory solution.
In many cases the side effects are so great that the continued administration of corticosteroids is no longer necessary However, the need for further administration would be urgently needed. Then you have calibrated often in such a way that one administered smaller amounts than were necessary, or that one often co-administered the treatment Corticosteroids completely stopped, although continuation of the treatment would have been advisable. It is now a success Finding a method by which the side effects of the corticosteroids are turned off by one at the same time with the corticosteroid, or shortly before or shortly after an additional agent administered. This invention Agent eliminates the known side effects without eliminating the desired effect of the corticosteroids

oder eingeschränkt wird.or is restricted.
Accordingly, it is primarily on gate of the invention that Eliminate the side effects of administering corticosteroids to achieve the desired ones essential effects of the corticosteroids are preserved at the same time.
Eine weitere wesentliche Aufgabe der Erfindung besteht darin, ein überlegeneres Arzneimittel zu schaffen, dasAnother essential object of the invention is to provide a more superior drug that

Für Predison liegt für die Erzielung der eindämmenden Wirkung die orale Dosis bei 20 bis 60 mg pro Tag, die dann stufenweise erhöht werden kann, bis die optimaleThe exact dose of the corticosteroid to be administered depends on the type of corticosteroid used and the disease to be cured. The invention is applicable to all effective amounts. For example, in the case of cortisone and oral administration, the dose can be 2.5 to 100 mg, for intramuscular administration 5 - 300 mg, for intravenous administration up to 100 mg, and for ointments for local treatment and for suspensions 0.5 - 2.5 96. In the treatment of cattle for acetonemia, the dose can be between 1.0 and 1.5 g i.d. m. amount. In the treatment from dogs for arthritis it can be between 25 and 75 mg when taken orally. When using Hydrocortisone the dose is generally 2/3 the dose of cortisone. For predison lies for achieving the containment Effect the oral dose at 20 to 60 mg per day, which can then be increased gradually until the optimal one

Maintenance dose has been reached. When using dexamethasone, which is approximately 7 times as strong, than prednisolone and 20 to 35 times as strong as hydrocartisone, the dose is 0.5 to 1.5 mg orally, while a solution of $ 0.1 is appropriate for topical application. As already indicated above, the present one is Invention applicable to all effective doses and for all corticosteroids.
It is believed that the action of the corticosteroids comes about through passive binding to microvascular smooth muscle cells and through interference with the anticonvulsant effects of histamine. Histamine is usually found in small amounts in smooth muscle cells. ä The generation of histamine is, however, achieved by strong stimuli either local or systemic nature (local or systemic nature). In general, there is an increase in histamine production when there is significant irritation (stress), such as; B. major injury, and then causes astringent substances such as adrenaline and noradrenaline to be released.
It could therefore be that the corticosteroid's intended counteraction against the increased histamine * production is the main function (essential stress function) of these hormones, the susceptibility to injuries in animals suffering from adrenaline deficiency and the wide variation in the requirement for the Corticosteroid effects (underly).
Daraus ergibt sich, daß die Corticosteroid-It therefore seems to be the case that the serious side effects of corticosteroid therapy by the tendency of these hormones to affect the microvascular muscles as a result of the Counteraction caused by the normal relaxation effects of the histamine. It follows that the corticosteroid

gäbe den cappilaren Blutstrom vermindern, daß das Gewebe unterernährt wird und infolgedessen vielfältige (widespread) abnorme Reaktionen hinsichtlich der chemischen Vorgänge in den Zellen und anderer Funktionen entstehen, welche die Nebenwirkungen der Corticosteroidtnerap^ie verursachen.the cappilar blood flow would diminish that tissue is malnourished and as a result diverse (widespread) abnormal reactions to the chemical processes in cells and other functions that cause the Cause side effects of corticosteroid therapy.
The agents according to the invention apparently counteract the tendency of the hormones, which consists in the capillary Decrease blood flow, while on the other hand increasing blood flow side effects of corticosteroid treatment prevented, but at the same time an interaction with the anti-inflammatory effect of the corticosteroid does not occur. It is also advantageous that the agents according to the invention only have a small amount Have toxicity and, more importantly, that they can be administered orally.
The following examples are provided for further explanation the invention. However, the scope of protection of the invention is not intended to be restricted by the exemplary embodiments cited will.
An injured adrenalectomized rat (a stressed adrenalectomized rat), which was exposed to parts of the adrenal gland 5 mg of hydrocortisone are injected. The increased closing time of the microvascular muscles of the Liver causes an abnormally rapid overflow of the open remaining capillaries. The process causes the activation of the metabolism of the over-nourished liver cells, whereby the general non-specific liver anabolic characteristics of the animal treated with the corticosteroid increases.

Another rat prepared in this way was given the same dose of hydrocortisone and 1 mg of beta-2-pyridyl-methylethylene hydrochloride, Beta-histine hydrochloride is injected in physiological saline solution. It has been shown that despite the restoration the puncture (restoration) of the open and closed Sphincter muscles the extent of the side effects is significantly reduced.
