Description
It is known that in the fermentation of a nutrient medium with certain molds of the Genus Cephalosporium (for example with the one in the American Type Culture Collection under the ATCC number 14,553 deposited organism) antibiotic activity is developed. Such fermentations are in British Patent 810196, U.S. Patent 3,184454 and U.S. Patent No. 3,184454 French patent 1,353,113.
These patents also describe certain processes for the recovery and purification of cephalosporin C from the crude fermentation liquor, for example the following: (1) decolorization by passing the filtered fermentation mixture through a first column containing activated carbon and then through a second column, containing aluminum oxide, (2) acidifying the decolorized filtrate with an acidic buffer solution, for example pyridine and sulfuric acid or with an ion exchange resin in acid form, (3) removing inorganic anions by reading through a column containing a basic synthetic resin anion exchanger (trademark Amberlite IRA 400 ) (OH form), (4) Adsorption of the active cephalosporin on a column containing a basic synthetic resin anion exchanger (trademark Amberlite IR 4 B) (H + form), (5) Eluting with an aqueous buffer, for example ammonium acetate and (6) recovering cephalosporin C from the eluate.
The invention aims to provide an improved process for the recovery of cephalosporin C from fermentation liquids in fewer steps and in higher overall yield.
The method according to the invention for obtaining cephalosporin C from fermentation liquids, wherein the fermentation products are separated off by filtration, the crude culture broth filtrate through passed an ion exchanger and this is then eluted, is characterized in that the crude culture broth filtrate is mixed with 1 to 4 times the volume of acetone, filtered and then poured into usually processed further.
This modification of the known recovery process, in which the original filtrate with the 1 to 4 times the volume of acetone is added, leads to the unexpected result that many of the undesirable Precipitated impurities from the broth while the cephalosporin C remains in solution.
Dowex 50) (H+-Form) geleitet,The method according to the invention is carried out as follows: the crude cephalosporin C fermentation broth is filtered to remove the mycelium and other insoluble matter, and the filtrate is diluted with about 2 to 3 times the volume of acetone while stirring. The mixture will be about Let sit for 30 minutes and refilter to a gummy precipitate of low activity to remove. The filtrate is passed through a bed of an ion exchange resin made from styrene-divinylbenzene copolymer (Wz.Dowex 50) (H + form) headed,
The effluent from this treatment stage is then passed through a column made of a synthetic resin ion exchanger (acetate or formate form), the amount of which is so large that 25 to 60 g of the activity come into contact with 11 resin each time. From The liquid flowing off the column is collected by distillation to obtain the acetone, and that Adsorbate is, based on the applied volume
ao men an Ionenaustauscher mit etwa dem 40- bis 50fachen Volumen einer Natriumformiat-Pufferlösung vom pH 3,5 bis 4,0 eluiert. Das Eluat wird bei einer Temperatur von nicht mehr als 30° C zu einem dicken Sirup eingeengt, falls nötig zur Einleitung der Kristallisation mit Kristallen von Cephalosporin C angeimpft und kristallisieren gelassen.ao men on ion exchangers with about 40 to 50 times the volume of a sodium formate buffer solution eluted from pH 3.5 to 4.0. The eluate becomes at a temperature not higher than 30 ° C concentrated in a thick syrup, if necessary to initiate crystallization with crystals of cephalosporin Seeded with C and allowed to crystallize.
The resulting slurry is a syrupy crystalline mass used to improve its filterability and to produce a drier, more manageable product of about twice the volume Methanol is diluted.
This dilute slurry is filtered and the solid crystalline product is air dried.
The method according to the invention leads to implementation to improved yields of purified cephalosporin C in the manner described above high quality obtained by any of the known processes for the production of 7-aminocephalosporanic acid, commonly known as the cephalosporin nucleus can be used.
