Verfahren zur Herstellung von Carbonsäurechloriden der Thiazolreihe
Aus den bekanntgemachten Unterlagen des belgischen Patents 618 751 ist bekannt,
daß man Carbonsäurehalogenide von Thiazolen, die in 2-Stellung ein Chloratom und
in 4,5-Stellung einen aromatischen Rest. ankondensiert enthalten, erhält, wenn man
die entsprechenden Carbonsäuren, die in 2-Stellung eine Hydroxy- oder Mercaptogruppe
enthalten, mit einem Phosphorsäurechlorid in Gegenwart von wenigstens 0,5 Äquivalenten,
bezogen auf die Carbonsäure, eines Formamids bei wenigstens 90°C umsetzt. Auf Seite
3 der bekanntgemachten Unterlagen des belgischen Patents ist ausgeführt, daß bei
Verwendung von weniger als 0,5 Äquivalenten Dimethylformamid, bezogen auf die Carbonsäure,
nicht das der Carbonsäure entsprechende, ü12-Stellung chlorierte Säurechlorid, sondern
das Dithiazolylsulfidcarbonsäurechlorid erhalten wird. Aus diesem Grund sollen wenigstens

Rest stehen, wobei wenigstens einer der beiden Reste eine Carboxylgruppe trägt,

können, der wenigstens eine Carboxylgruppe trägt, mit einem Halogenierungsmittel
erhält, wenn man die 2-Mercaptothiazolcarbonsäuren mit Phosgen bei Temperaturen
zwischen etwa 20 und 150°C in Gegenwart von wasserfreien, inerten Lösungs- und/
oder Suspensionsmitteln und in Gegenwart von 0,1 bis 5 Molprozent eines N-disubstituierten
Carbonsäureämids, bezogen auf die 2-Mercaptothiazolcarbonsäure, umsetzt, das Umsetzungsgemisch
gegebenenfalls auf Temperaturen zwischen 60 und 150°C erwärmt und das gebildete

dieser Verbindungen mit Phosgen erfolgt in inerten, wasserfreien Lösungs- oder Verdünnungsmitteln,
wie Dioxan, Benzol, Ligrom, Toluol, Chlorbenzol, Chloroform oder Tetrahydrofuran.
Die Reaktion erfolgt bei gewöhnlichem oder erhöhtem Druck. Die obere Temperaturgrenze
ist innerhalb des oben angegebenen Bereichs meist durch die Siedetemperatur des

mit einem Stickstoffstrom und filtriert nach Zugabe von Tierkohle. Aus dem Filtrat
wird das Lösungsmittel abdestilliert. Der Rückstand wird unter vermindertem Druck
destilliert. Man erhält 18,5 Teile 2-Chlor-4-methylthiazol-5-carbonsäurechlorid
mit einem Siedepunkt von 96 bis 99°C bei 11 Torr. Die Ausbeute beträgt 94,40% der
Theorie, bezogen auf angewandte 2-Mercapto-4-methylthiazol-5-carbonsäure. Analyse
für C5H3C12NOS (196). Berechnet ..... Cl 36,20%; gefunden ..... C136,10/0.
Beis-piel2 Man bereitet unter Rühren bei 0°C eine Mischung von 10 Teilen 2-Mercaptobenzthiazol-5-carbonsäure,
75 Teilen Chloroform sowie 10 Teilen Phosgen und setzt 0,2 Teile Dimethylformamid
zu. Es wird 1 Stunde bei 0 bis 5°C gerührt, dann heizt man in einer Stunde unter
ständigem Einleiten eines schwachen Phosgenstromes auf Siedetemperatur und kocht
7 Stunden unter weiterem Phosgeneinleiten unter Rückfluß. Nach Abkühlen auf Raumtemperatur
wird das Chloroform zusammen mit noch vorhandenem Phosgen mit trockenem Stickstoff
abgetrieben. Der feste weiße Rückstand wird mehrmals mit jeweils 100Teilen Ligroin
ausgekocht. Aus den vereinigten Ligroinfiltraten scheiden sich beim Abkühlen weiße
Kristalle ab, die abgesaugt und mit wenig Ligroin gewaschen werden. Man erhält 8,8
Teile 2-Chlorbenzthiazol-5-carbonsäurechlorid vom Schmelzpunkt 131'C. Die
Ausbeute beträgt 80% der Theorie, bezogen auf angewandte 2-Mercaptobenzthiazol-5-carbonsäure.
