Description
Process for the production of active ingredients from spleens by tabular abuse or through toxic side effects of properly indicated drugs, But especially with the new cvtostatic substances that are used clinically for treatment Unlimited tumor growth are used, it often comes to not inconsiderable Impairments in the production of white blood cells. The bone marrow finds out a disturbance of its normal function, and leukopenic conditions result, which often take on considerable proportions and lead to death. The previous therapy consists of blood transfusions or the use of certain corticoids without this it has so far been possible to restore the normal function of the Restore bone marrow.
The invention relates to a process for the production of the Active substances that promote the formation of white blood cells in healthy bone marrow. It is based on the knowledge gained through clinical and experimental studies, that in the healthy spleen of warm-blooded animals there are not only such substances which cause an inhibition of the production of white blood cells in the bone marrow, but likewise other substances that affect the bone marrow function and especially the formation of white Stimulate blood cells. The latter substances are used in leukemic cases injected, there will be tremendous increases in the number of white blood cells. If this substance is injected in leukopenic conditions, in the course of one or 2 weeks, the production of healthy white blood cells is strongly mobilized Patients recover, and above all the fever and the weakness in defense against infections go away back in agranulocytosis.
According to the main patent application W 18273 IV a / 3Qh, a becomes the malignant White blood cell growth inhibiting agent obtained by finely chopping, spleens of healthy animals freed from fat in water with strongly acidic or alkaline water Reaction homogenized, slowly heated with stirring up to about 80 ° C, which is not separates dissolved components, the protein precipitates, the filtrates up acidified further to pH 4 to 4.5, the resulting precipitates are separated off, the mother liquor salted out, the collected precipitates dissolve in glacial acetic acid and fractionated by repeated Precipitation with ether, acetone and petroleum ether broken down into the petroleum ether fraction isolates the highly enriched active ingredient.
It has now been found that this method also simultaneously Another active ingredient is produced, which is the growth of white blood cells in healthy bone marrow, which promotes leukopoiesis and which is involved in the procedure of Hau.ptp.atentanmeldung enriched in the ether acetone precipitation, which in the case of the Obtaining the inhibitor-directed process of the main patent application because of their low content of this substance was discarded. According to the present invention the procedure of the main patent application is further expanded in the sense that also the ether-acetone fractions are worked up in order to convert them to the new ones, the formation gain the active substance that promotes white blood cells in healthy bone marrow, which is of considerable importance for therapy. It works particularly well with him in people whose bone marrow function by means of fighting life-threatening The medication administered during the disease process paralyzed the bone marrow function and to stimulate and get the production of white blood cells going again.
The process is carried out with the same starting materials and in the same white as after the main patent application, only that the ether-acetone precipitate at the end worked up by repeated fractional extraction and reprecipitation will.
It was also found that the active ingredients of interest here are not contained only in the spleens but also in the livers of healthy animals, so that livers can also be used as starting material.
From the German patent specification 714 707 it is known from Livers build leukocytes promoting active component together with a factor that stimulates erythrocyte formation. The separation from the purely aqueous extract is done by adding one with water in each Ratio of miscible organic solvents, such as acetone, up to concentrations of 60%. In contrast, however, according to the present invention, it is instantaneous hydrolysis brought about by acid or alkali, which is the decisive prerequisite for a detachment of the sought-after active ingredients from the accompanying proteins bound to them secures. The protein precipitation carried out according to the state of the art without prior hydrolysis must also precipitate most of the active ingredients before they are obtained can. According to the present invention, the active ingredients are not distributed from aqueous solution, but only from various organic solvents, see above that protein-free products are actually obtained.
According to US Pat. No. 2,662,047, one goes through benzene defatted spleen dry powder, extracted the residue with ether and niniint after evaporation of the ether with water. The active ingredients according to the present Invention obtained are soluble in benzene. So you will in the application of the process of the patent mentioned already removed with the degreasing. Indeed The aforementioned patent also wants a completely different active ingredient, namely an against Allergies gain effective principle, and the resulting substances are in toluene, Acetone and cold ethyl alcohol are insoluble, so basically different from the active ingredients obtained by the process of the present invention.
One works as according to the example of the main patent application W 18273 IVa / 30h until the clear, centrifuged glacial acetic acid crude extract is obtained. Then moved 100 cc of clear glacial acetic acid extract with 250 cc of a mixture of equal parts Acetone-ether, sucks off the coarse precipitate and dries. The filtrate will mixed with 500 ml of petroleum ether and the corresponding fine precipitate as well sucked off and dried. The precipitates turn into glacial acetic acid again dissolved and fractionated with acetone-ether and petroleum ether precipitated. The on .this Well-obtained precipitates can be used for injection.
In principle, these active ingredients can be used in the manner described from all reticular tissues such as spleen and liver. It is crucial that in the case of the representation in the alkaline range up to a reasonably clear one Solution reached. Only in this way is it possible to use those present in the acidic extraction and the resulting colloidal, suspensoid and emulsoid substances become almost molecularly disperse Form from. Which one can more easily secrete the accompanying proteins by acidification can without the active ingredients being precipitated.
