Bei diesem Verfahren wird mit der Steuereinheit sichergestellt, dass die Scanbewegung des Laserlichts zwei Mal an dem zu untersuchenden Objekt gestreut wird. Sollte das zu untersuchende Objekt beispielsweise im Randbereich des scannenden Laserstrahls liegen, so kann der Fall auftreten, dass nur einmal das gestreute Licht erfasst wird. Bei der Verwendung des reflektierten Laserlichts ist nur eine Abbildungsoptik erforderlich, wodurch sich der Aufwand für die Messvorrichtung verringert. Bei beiden Ansätzen wird das gestreute Licht auf die Detektionsdiode fokussiert. Diese misst die modulierte Lichtleistung. Die Modulationstiefe, also der Unterschied zwischen maximaler und minimal detektierter Intensität schwankt dabei mit der Größe des Moleküls, denn größere Moleküle streuen mehr Licht. Intensitätsmaximum und Minimum unterscheiden sich also nur schwach von der an der Diode gemessenen mittleren Leistung.
