Description
Bereich der Erfindung Field of the invention
The present invention relates to novel steroid carboxylic acid esters having improved properties and compositions containing these steroid-carboxylic acid esters with topical local application for cosmetic or dermatological purposes. The invention also relates to the use of the steroid carboxylic acid esters or of a composition containing these steroid carboxylic acid esters with local topical application for cosmetic or dermatological purposes for the treatment and alleviation of itching.
Stand der Technik State of the art
The skin, as the largest human organ, performs many important functions. It provides protection against heat, cold, radiation, the ingress of pathogens and chemical substances. In its role as a barrier organ, the skin is particularly subject to external agents. The barrier function is maintained, among other things, by skin lipids released in the keratinization process of the keratinocytes. If the barrier function of the skin is disturbed, water-binding capacity, elasticity, ability to regenerate or the resistance to environmental influences are lost. As a result, both local irritation and holistic discomfort and pain may occur.
It is known that certain steroid hormones and their carboxylic acid esters can be used for cosmetic and dermatological purposes because of their positive effects on the body. Thus, according to the current state of the art, the occurrence of itching and skin irritations with corticosteroids is counteracted. For the topical use of steroids, these are used in the form of steroid carboxylic acid esters, which serves for a targeted release of the steroids at their preferred site of action in the skin by the human carboxylesterase 2 (hCE2) located in the keratinocytes.
However, the known side effects of corticosteroids are of great disadvantage. For example, skin depletion of collagen (atrophy of the skin) and deterioration of barrier function (increase in transepidermal water loss) have been observed. In addition, in all likelihood, topical use of corticosteroids in very low concentrations also causes these steroids to enter the bloodstream, leading to a negative feedback on steroid synthesis in the adrenal cortex. This is especially important in children and the elderly. In diseased skin with reduced barrier function, there is an increased systemic absorption. This can lead to the development of iatrogenic Cushing's syndrome. In children, growth may be delayed or aseptic femoral head necrosis may develop. Even low levels of corticosteroids may be sufficient to induce vision loss glaucoma.
In addition, an application of corticosteroids over a longer period is usually required in order to observe a mitigation of itching. This is particularly disadvantageous because the side effects described increase with prolonged use.
Aufgabe der Erfindung Object of the invention
The object of the invention is therefore to overcome the disadvantages of the prior art and to provide compounds that reduce itching quickly and reliably in topical local application for cosmetic or dermatological purposes and also have no undesirable steroid effects.
Offenbarung der Erfindung Disclosure of the invention
The object is achieved by a new steroid carboxylic acid ester for use in local topical application for cosmetic or dermatological purposes with the formula (I) where R is a branched acyl radical.
The invention is based on the observation that 4-O-carboxylic acid esters of androst-4-ene-3,17-dione, which have a branched acyl radical R, cause a significant reduction in itching after a short-term application. In addition, it has been observed that the 4-O-carboxylic esters of androst-4-ene-3,17-dione are reacted more rapidly with a branched acyl radical R in human keratinocytes than 4-O-carboxylic esters of androst-4-en-3, 17-dione in which the acyl radical is unbranched. Moreover, the 4-O-carboxylic acid esters of androst-4-ene-3,17-dione with a branched acyl radical R have no undesirable side effects as caused by corticosteroids.
In addition, it has been found that skin irritations based on allergies or due to insect bites or UV or X-radiation can be very favorably influenced by the steroid carboxylic esters according to the invention. Furthermore, it was found that the skin moisture and the transepidermal water loss and thus the barrier function of the skin are positively influenced.
Besonders bevorzugt ist das 4-O-2-Methyl-n-butyryl-androst-4-en-3,17-dion According to an advantageous embodiment of the invention, the radical R of the steroid carboxylic acid ester is selected from 2-methyl-n-butyryl and isocaproyl. Both 4-O-2-methyl-n-butyryl-androst-4-ene-3,17-dione and 4-O-isocaproyl-androst-4-ene-3,17-dione are compared to steroid carboxylic acid esters with unbranched acyl radicals an improved cosmetic and pharmaceutical effect. Particularly preferred is 4-O-2-methyl-n-butyryl-androst-4-ene-3,17-dione
The steroid carboxylic acid esters 4-O-2-methyl-n-butyryl-androst-4-ene-3,17-dione and 4-O-isocaproyl-androst-4-ene-3,17-dione in the context of the present invention include both the "S" enantiomers and the "R" enantiomers of the steroid-esterified carboxylic acid. Preferably, the "S" enantiomers are 4-O-2-methyl-n-butyryl-androst-4-ene-3,17-dione and 4-O-isocaproyl-androst-4-ene-3,17-dione , These are reacted faster by the human carboxylesterase 2 (hCE 2) than the corresponding "R" enantiomers.
Another object of the invention is a composition for use in topical local application for cosmetic or dermatological purposes, which contains at least one steroid carboxylic acid ester of the formula (I), wherein R is a branched acyl radical. After 28 days, the skin moisture improved by 80% in all subjects. Of the Transepidermal water distress, which is increased with poor barrier function, decreased by 36% after 28 days in all subjects.
The better implementation of the inventive steroid carboxylic acid esters with branched acyl radicals is exemplified by the example of 4-O-2-methyl-butyryl-androst-4-ene-3,17-dione in comparison to a steroid carboxylic acid ester with unbranched acyl radical, the 4th O-propionyl-androst-4-ene-3,17-dione.
HaCaT human keratinocyte cell line was used to cleave the two carboxylic acid esters of 4-hydroxyandrostenedione, 4-O-2-methylbutyryl-androst-4-ene-3,17-dione and 4-O-propionyl-androst-4- en-3,17-dion. At time points 0, 3, 8, 12 and 15 minutes, the enzymatic reaction of cleavage of the esters by the human carboxylesterase (hCE2) expressed in the cells was stopped by addition of sodium fluoride and the cells were lysed by ultrasound. In the lysates, the quantification of the starting materials and products of the enzyme reaction were carried out with the aid of LC / MS (liquid chromatography with mass spectroscopy coupling).
Ergebnis: Result:
It was found that the concentration of 4-O-propionyl-androst-4-ene-3,17-dione only decreased by 3% within 15 minutes, while the cleavage of 4-O-2-methyl-butyryl-androst 4-en-3,17-dione caused the concentration of the ester to decrease by 25% within 15 minutes. Accordingly, a significantly higher conversion was achieved within 15 minutes with 4-O-2-methylbutyryl-androst-4-ene-3,17-dione than with 4-O-propionyl-androst-4-ene-3 , 17-dione.
Bei einer topischen Anwendung der Steroid-Carbonsäureester kann somit die Wirkung des 4-O-2-Methyl-butyryl-androst-4-en-3,17-dion wesentlich schneller eintreten, als beim 4-O-Propionyl-androst-4-en-3,17-dion. In a topical application of the steroid carboxylic acid esters, the action of 4-O-2-methyl-butyryl-androst-4-ene-3,17-dione can thus occur much faster than with 4-O-propionyl-androst-4-. ene-3,17-dione.
Die Herstellung der erfindungsgemäßen Steroid-Carbonsäureester wird nachfolgend anhand der Herstellung des 4-O-2-Methyl-butyryl-androst-4-en-3,17-dion beschrieben: The preparation of the steroid carboxylic acid esters according to the invention is described below with reference to the preparation of 4-O-2-methylbutyryl-androst-4-ene-3,17-dione:
Darstellung von 4-O-2-Methyl-butyryl-androst-4-en-3,17-dion (2-Methyl-buttersäureester von Formestan, 4-OMA) Preparation of 4-O-2-methylbutyryl-androst-4-ene-3,17-dione (2-methylbutyric acid ester of formestan, 4-OMA)

9.57 g of formestane (31.7 mmol) are dissolved in 35 ml of dry DCM, 6 ml of triethylamine (1.01 eq based on acid chloride) and 23 mg of DMAP are added. 5 ml of 2-methyl-butyric acid chloride (1.25 eq.) Are dissolved in 20 ml of dichloromethane. It is cooled slightly with a water bath and the acid chloride is added dropwise. The addition is slightly exothermic (20-25 ° C). After a short time, turbidity occurs. TLC control is possible after dilution with DCM with the mixture DCM / MeOH 50: 1. After one hour, 50 ml of 1M hydrochloric acid are added dropwise and the phases are separated. The organic phase is washed twice with 50 ml of sodium bicarbonate solution and once with 50 ml of brine, and dried over sodium sulfate. After filtration and concentration at 40 ° C to obtain 12.72 g of crude product (09-14-I69 P6065), which solidifies after smoking with DCM to a white wax. It is recrystallized at boiling heat from 30 ml of cyclohexane. The product is mechanically minced and rinsed with 3 times 3 ml of cyclohexane on the frit.
