Description
The invention relates to a method for the surface treatment of a carrier which is adapted to receive particles, a carrier with a functionalized carrier surface, and a method for the preparation of a carrier for examination purposes, in particular for electron microscopy or optical examination methods. More particularly, the invention relates to methods and substrates in which particles are primarily disposed adjacent to one another along the substrate surface. Applications of the invention are in the preparation of carriers (sample carriers, substrates), in particular for electron microscopy or optical investigation methods, for. As spectroscopy, and the investigation of nanoparticles given.
It is generally known to investigate inorganic and / or organic particles by means of electron microscopy. The particles are deposited on a carrier adapted for electron microscopy (so-called grid), transferred to the electron microscope and in the electron microscope z. B. irradiated for imaging or for a diffraction examination with an electron beam. The carrier has such a small thickness for transmission electron microscopy that weakening of the electron beam by the carrier does not interfere with the examination of the particles.
The deposition of the particles takes place, for example, by pipetting a liquid drop having a volume of a few μl, which contains the particles, onto the surface of the support. After waiting for excess liquid is removed, for. B. sucked with filter paper. It has proved to be disadvantageous in practice that in particular macromolecular particles with a polarizability or charge on the carrier surface preferably form aggregates or clusters and lie relative to the carrier surface one above the other or possibly chemically react with each other. As a result, the electron microscopic examination can be made difficult or even impossible.
The known functionalization processes can have a number of disadvantages which result from the most complex procedure, a complicated handling of the carriers and their low durability. For example, the charges formed on the surface can be undesirably neutralized if handled improperly before use.

Gemäß dem zweiten allgemeinen Gesichtspunkt der Erfindung wird insbesondere ein Träger, der zur Aufnahme von Partikeln eingerichtet ist, mit einer Funktionalisierungsschicht bereitgestellt, die eine mittels Photopolymerisation gebildete Polymerschicht umfasst. Vorzugsweise ist der Träger mit einem Verfahren gemäß dem ersten allgemeinen Gesichtspunkt der Erfindung hergestellt.According to a second general aspect of the invention there is provided a support adapted to receive particles comprising a support surface having a functionalizing layer formed from at least one photopolymer and having such an effect on particles deposited on the support surface, that the particles are distributed on the surface side by side. In particular, according to the second general aspect of the invention, a carrier adapted to receive particles is provided with a functionalizing layer comprising a polymer layer formed by photopolymerization. Preferably, the support is made by a method according to the first general aspect of the invention.
According to a third general aspect of the invention, there is provided a method of preparing a carrier with particles for examination of the particles comprising the steps of depositing drops of a liquid containing the particles with a microdispensing device on a carrier surface of the carrier and removing the liquid in which the carrier surface of the carrier has such a functionalization that the particles form a planar distribution on the surface of the carrier. According to the third general aspect of the invention, the functionalization may include the formation of the functionalization layer according to the first general aspect of the invention or another, especially known as surface modification method, in particular a deposition of a two-dimensional monolayer, preferably of organic molecules, on the support surface, and / or include a plasma treatment. The two-dimensional monolayer is also referred to as 2D Self Assembling Monolayer. It is z. By silanization of the support surface in solvent environment or in gas phase environment, e.g. B. formed with chloro or alkyloxysilanes. The microdispenser is designed for non-contact dispensing. It comprises a dispenser with which drops with a volume of less than 1 nl, preferably less than 500 pl, are generated and transferred to the carrier (spotting). The microdispensing device is preferably equipped with a piezoelectrically operated dispenser. Alternatively, a vapor bubble-powered dispenser (bubble-jet dispenser) or an electrospray dispenser may be provided.
The functionalization layer (or: functionalization coating, functionalization film) generally has a composition (chemical composition and / or surface geometry) such that the particles have a different, preferably more attractive, interaction with the carrier surface, in particular with the functionalization layer, than with adjacent particles. The functionalization layer causes a change in the surface energy in comparison to the uncoated carrier surface. The functionalization layer is in particular formed in such a way that particles which are deposited on the carrier surface, preferably supplied to the carrier surface in a drop of liquid, are distributed over the surface (two-dimensionally). In other words, the functionalization layer provides a surface that is suitable for arranging particles along the carrier surface with priority to one another. The functionalization layer has a multiplicity of adhesion sites to which particles collect individually. A majority or all of the particles form the areal distribution, i. H. a distribution which is free of aggregates (accumulations of particles above the support surface) or which contains negligible aggregates when examining the particles on the support. With the functionalization layer advantageously takes place adjustment of the wettability of the surface of the carrier and / or an electrostatic fixation of particles. It is preferably provided a targeted design of the functionalization layer, with which the distribution of the particles is adjusted on the support.
The functionalization layer is preferably configured such that the particles on the carrier surface are exposed to a stronger interaction by the functionalization layer than by other particles. As a result, the particles on the support surface provided with the functionalization layer form a monolayer or sub-monolayer. The particles are preferably singulated on the carrier by the interaction with the carrier, but alternatively may be arranged directly adjacent to one another in the lateral direction.
Advantageously, with the surface treatment according to the invention, which can also be referred to as surface functionalization, surface coating or surface modification, disadvantages of conventional techniques are avoided by simplifying the provision and / or handling of the carriers and more reliably forming a two-dimensional particle distribution. The invention offers the particular advantage that in the first and second aspects of the invention, the functionalization layer or in the third aspect of the invention, by combining with the supply of particles by means of the Mikrodispensiereinrichtung, generally any functionalization fulfills the separation function of the functionalization more effective and reliable in conventional carriers. Aggregate formation is minimized and the particles are placed primarily as a monolayer or sub-monolayer. Advantageously, the deposition density of the particles be optimized. The particles can be arranged evenly distributed. In other words, a homogeneous distribution of the particles is formed.
The functionalization layer is a layer formed from at least one photopolymer. The photopolymer is composed of monomers whose polymerization is effected by exposure, if appropriate in the presence of initiator molecules. The layer may cover the carrier in sections and z. B. form islands of the photopolymer and / or uncoated uncoated sections. Preferably, a continuous, closed covering of the carrier is provided by the functionalization layer. The functionalization layer can be formed from a single photopolymer or from several photopolymers.
Unlike conventional surface modifications, eg. B. in the form of a 2D Self Assembling monolayer, the functionalization layer is preferably formed three-dimensional. Advantageously, this offers more possibilities of interaction for the particles. More trapping sites for the particles may be provided and / or the particles may even be partially embedded in the functionalization layer. In the photopolymerization, the functionalization z. B. be formed to form so-called polymer brushes, in which the particles are partially embedded or fixed in the polymer layer. This effect can lead to a better separation of the particles on the functionalized areas of the carrier.

Partikel biologischer Materialien (insbesondere biologische Makromoleküle), Partikel aus anorganischen oder organischen Substanzen, oder anorganische oder organische Partikel in Verbindung mit biologischen Materialien. Bevorzugt sind die Partikel Nanopartikel, insbesondere Partikel mit einer charakteristischen Größe, z. B. Durchmesser oder Länge, die geringer als 1000 nm, besonders bevorzugt geringer als 500 nm sind.Applications of the invention relate to carriers for receiving particles, in particular for examination purposes. Die auf einem Träger abgeschiedenen Partikel können vom gleichen Typ, insbesondere von gleicher Zusammensetzung, oder verschieden sein. Die Funktionalisierungsschicht kann so gestaltet werden, dass Partikel sowohl positiv und/oder negativ geladene Partikel als auch neutrale Partikel auf der Oberfläche der Funktionalisierungsschicht zweidimensional verteilt werden.Inorganic particles include, for. As an electron microscope (eg., Transmission electron microscope, TEM, scanning electron microscopy, SEM, imaging and / or diffraction measurement) or spectrometer (eg for fluorescence or Raman scattering measurements) designed. For example, the support for electron microscopy has a lateral size such that it fits the sample receptacle of the electron microscope, and a thickness such that a weakening of the electron beam by the support constitutes a negligible attenuation for the result of the examination of the particles. The carrier is z. As a SEM and / or TEM support (or: grid, window, membrane, filmed network for TEM and / or SEM), preferably comprising a support, for. B.

Vorzugsweise ist die Trägeroberfläche aus Siliziumoxid, Siliziumdioxid, Siliziumnitrid oder Kohlenstoff hergestellt.Ring, grid or mesh, and a continuous, electron-transparent film (film), which forms the support surface and on which the functionalization is arranged. As such, the carrier surface may be structured (eg, with gradations) or unstructured. Preferably, the support surface is made of silicon oxide, silicon dioxide, silicon nitride or carbon.
Besonders bevorzugt wird die Photopolymerisation unter Verwendung von olefinisch ungesättigten Monomeren (z. B. Acryl- bzw.- Methacrylmonomeren) in Gegenwart von Photoinitiatoren (z. B. Benzophenon) in wässriger oder organischer Lösungmittelumgebung durchgeführt. Most preferably, the photopolymerization is carried out using olefinically unsaturated monomers (eg, acrylic or methacrylic monomers) in the presence of photoinitiators (eg, benzophenone) in an aqueous or organic solvent environment.

Die Funktionalisierungsschicht kann vollständig neutral oder abschnittsweise neutralen und/oder positiv und/oder negativ geladen sein.According to a further preferred embodiment of the invention, the functionalization layer can be formed with an electrically neutral surface and / or a surface with positive and / or negative charge. The functionalization layer can be completely neutral or partially neutral and / or positively and / or negatively charged.

The inventors have found that the functionalizing layer does not interfere with the function of the wearer in the study. According to a further preferred embodiment of the invention, the functionalization layer can be formed after or during its production with a surface structure. Advantageously, the deposition structure of the particles can be optimized with the surface structure, for. B. can be maximized without an undesirable interaction of the particles, eg. B.
