Beispiel 2: Wasserextraktion und SäulentrennungExample 2: Water extraction and column separation
Ein Kilogramm trockener Wurzel-Rinde von Morus australis Poir. wurde einmal aufgekocht beziehungsweise zweimal mit 10 L Wasser für eine Stunde unter Rückflusskühlung erhitzt. Die Rohextrakte wurden anschließend mit einem 350 Maschensieb filtriert beziehungsweise die Filtrate wurden gesammelt. Die Reste bzw. Rückstände wurden jeweils mit der zehnfachen Menge Gewichtswasser für einen zweiten Extraktionsvorgang (1 Stunde) gemischt, und anschließend mit einem 350 Maschensieb filtriert, um ein anderes Filtrat zu erhalten. Die vorstehend erwähnten Wasserextrakte wurden jeweils durch eine Diaion HP20 Säule (Diaion, Mitsubishi Chemistry Inc.), wobei das Verhältnis der Trockenbasis des Extrakts und des Harzes 1/30 betrug. Nach dem Laden der Extrakte auf die Säulen wurden die Diaion HP20 Säulen der Reihe nach mit der vierfachen Volumenmenge RO Wassers und der dreifachen Volumenmenge einer 35–95%-igen Ethanollösung, um entsprechende Ethanoleluate (Proben 1-1 bis 1-6) zu erhalten. Die Ethanoleluate werden konzentriert und gefriergetrocknet, um das Ethanol in den Eluaten zu entfernen und trockene Pulver zu erhalten. Die Ausbeute der vorstehend erwähnten Eluate ist in Tabelle 1 gezeigt. Die Beurteilung des Hustenmittels der vorstehend erwähnten Eluate ist in Tabelle 2 gezeigt. Tabelle 1

Die vorstehend erwähnten Wasserextrakte wurden jeweils durch eine Diaion HP20 Säule (Diaion, Mitsubishi Chemistry In.) aufgetrennt, wobei das Verhältnis der Trockenbasis des Extraktes und dem Harz 1/20 bis 1/30 betrug. Nach dem Laden der Extrakte auf die Säulen, wurden die Diaion HP20 Säulen sequentiell jeweils durch 2 bis 4 fache Volumenmengen RO Wassers und 2 bis 3 fache Volumenmengen einer 50% Ethanollösung eluiert, um Ethanoleluate (Proben 1-7 bis 1-14) zu erhalten. Die Ethanoleluate wurden dann konzentriert und gefriergetrocknet, um das Ethanol in den Eluaten zu entfernen und trockne Pulver zu erhalten. Die Ausbeute der vorstehend erwähnten Eluate ist in Tabelle 3 gezeigt. Die Beurteilung des Hustenmittels der vorstehend erwähnten Eluate ist in Tabelle 4 gezeigt. Tabelle 3

Es wurde ein Kilogramm trockener Wurzel-Rinde von Morus australis Poir. einmal aufgekocht und zweimal mit 10 L einer 95% Ethanollösung für 1 Stunde unter Rückflusskühlung erhitzt, um einen Rohextrakt zu erhalten. Der Rohextrakt wurde anschließend mit einem 350 Maschensieb filtriert, wobei das Filtrat gesammelt wurde. Der Rückstand wurde mit der zehnfachen Menge Wasserwassergewicht für eine zweite Extraktionsvorgang (1 Stunde) gemischt, und anschließend mit einem 350 Maschensieb filtriert, um ein anderes Filtrat zu erhalten. Die zwei Filtrate wurden vereint, um ein Ethanolextrakt (Probe 2-1) zu erhalten. Der Ethanolextrakt wurde konzentriert und gefriergetrocknet, um ein Trockenpulver zu erhalten.
