Gewebeersatz. The present invention describes the encapsulation of bioactive materials in matrices
Biopolymers. The bioactive materials can be enzymes, microorganisms, cell cultures
or parts / organelles from microorganisms and cell cultures. The matrices are
complexes of the biopolymer Gellan. The capsules formed can be used for the biosynthesis of
Metabolites are used for biotransformation or for biodegradation. On
Another point of use is the controlled release of substances, for example when used as
Tissue replacement.
Gellan is a polymer [-3 -) - β-D-glucose- (1-4) -β-D-glucuronic acid- (1-4) -β-D-glucose- (1-4) -α-L- Rhamnose- (1-] n (see Fig. 2), which is primarily used as a gelling agent in the food industry, is formed by the microorganism Sphingomonas eloedea (formerly: Pseudomonas eloedea) and is present in large quantities in a constant, defined and constant manner The glucose subunits can have different degrees of acetylation.
einer Konzentration zwischen 1 und 10%, vorzugsweise 5%ig.The capsule formation is carried out analogously to the alginate capsules described in the literature by
Complex formation with cations, preferably multivalent cations. The Gellan is solved
at temperatures between 70 ° C and 100 ° C in ultrapure water in a moving bath. The concentration
of the gellan is between 0.5% and 10%, preferably 1%. The addition of the
encapsulating component takes place in the dissolved gellan. The mixture is in through a capillary nozzle
a bath containing a complex-forming substance. The complexing agent can be a cation,
preferably a polyvalent cation, preferably a calcium salt or barium salt. The
Complexing agent is preferably dissolved in a buffer such as PBS in Tris buffer, namely in
a concentration between 1 and 10%, preferably 5%.
The immobilization in capsules takes place with the aid of an encapsulation apparatus (see Fig. 1). The suspension consisting of the material to be immobilized and the gel-forming component is dripped through a capillary. This can be done with the help of a pump or with the help of a pressure-overlaid reservoir. A fluid flow (e.g. air) can also be given via the apparatus, with the aid of which the capsule size can be varied.

1. Verkapselung1. Encapsulation
A 1% (w / v) solution of the gellan in ultrapure water is prepared for the production of capsules. Calcium chloride is made up to 10% (w / v) in 20 mM Tris buffer, the pH of which is adjusted to 6.3 with HCl. A cell suspension of SF21 cells is added to the gellan, the cell concentration of which is 5 × 10 7 cells / mL. The resulting suspension is pumped through the encapsulation apparatus shown in Fig. 1. The resulting drops fall into the agitated calcium chloride bath and are stirred there for 30 minutes. The stirring speed must be set so that undeformed capsules can form.
The capsules are then separated from the Tris buffer and three times with phosphate buffer (100 mM,
pH 6.3) washed. This is followed by a washing step with medium (SF900II). The cultivation
the cells enclosed in the capsules are carried out in SF900II medium at 27 ° C. Vitality test with
MTT and EZ4U demonstrate the growth of the cells in the gellan capsules.
