Abstract
Der Nachweis von antinukleären Antikörpern in Körperflüssigkeiten dient der Diagnose von Kollagenosen sowie von anderen Erkrankungen des rheumatischen Formenkreises. Als Nachweismethode ist die indirekte Immunfluoreszenz gebräuchlich, bei der auf Objektträger fixierte Kryostatschnitte von Rattenleber oder epitheliale Zellinien mit Patientenserum überschichtet werden, so daß sich vorhandene antinukleäre Antikörper an die nukleären Antigene binden. Diese Fixierung ist aufwendig und zur modernen Routine kaum geeignet. Aufgabe der vorliegenden Erfindung ist es daher, Zellkerne an eine feste Phase zu fixieren, um damit eine effektive Bestimmung von antinukleären Antikörpern im Immunoassay zu ermöglichen. Die Lösung erfolgt durch kovalente oder adsorptive Bindung isolierter Zellkerne an chemisch aktivierte, feste Phasen insbesondere Polystyrenoberflächen. Die Vorteile des Verfahrens liegen in der effektiven Bestimmung großer Probenzahlen unter standardisierten Bedingungen, verbunden mit wesentlich geringeren Kosten in der klinischen Praxis.The detection of antinuclear antibodies in body fluids is used to diagnose collagenosis and other rheumatic diseases. Die Erfindung betrifft ein Verfahren zur qualitativen und/oder quantitativen Bestimmung von antinuklnären Antikörpern. Das Verfahren findet Anwendung in der pharmazeutischen Industrie, den Biowissenschaften und in der Medizin. Antinukleäre Antikörper (ΆΝΑ, ANF) sind Autoantikörper unterschiedlicher Spezifität, die gegen Zellkernantigene gerichtet sind. Der Nachweis von ANA ist besonders bedeutsam für die Diagnose von Kollagenosen, so vor allem dos systemischen Lupus erythematodes und der mit dieser Erkrankung eng assoziierten „mixed connective tissue disease (MCTD)", sowie von anderen Erkrankungen des rheumatischen Formenkreises.The invention relates to a method for the qualitative and / or quantitative determination of antinuclear antibodies. Die so gebundenen Antikörper werden durch ein FITC-markiertes Antihuman-Immunglobulin nachgewiesen und mit einem geeigneten Fluoreszenzmikroskop sichtbar gemacht. Die Präparation der Kryostatschnitte von Rattenleber erfordert neben dem Aufwand der Tierhaltung eine kostenintensive Technik (Kryostat). Der Einsatz von Zellen ist an din kontinuierliche Züchtung derselben unter definierten und standardisierten Bedingungen gebunden und somit für die immunologische Routinediagnostik wenig geeignet. Dieser nicht unerhebliche Aufwand sowie die hiermit verbundenen Kosten limitieren die Zahl der Bestimmungen in der hochspezialisierten immunoloijischen Diagnostik, so daß diese den Anforderungen der Kliniken nicht mehr gerecht werden kann. Hinzu kommt, daß die Durchführung des Testes, die Beurteilung der Muster unri die Interpretation der Befunde viel Erfahrung erfordern und somit dem Spezialisten vorbehalten sind. Die Aufgabe wird erfindungsgemäß dadurch gelöst, daß mittels Gradientenzentrifugation gewonnene intakte Zellkerne unterschiedlicher Herkunft, jedoch vorzugsweise aus Rattenleber, an feste Phasen adsorptiv oder aber kovalent gebunden werden, so daß diese zur spezifischen Bindung von antinukleären Antikörpern aus Körperflüssigkeiten im (Enzym-) Immunoassay eingesetzt werden. Hierzu wird die zu bestimmende Körperflüssigkeit, vorzugsweise Serum, in einer geeigneten Verdünnung mit den Zellkernen inkubiert, so daß sich vorhandene antinukleäre Antikörper an die Kernantigene der Zellkerne binden. Die so gebundenen Antikörper werden nunmehr mit einem markierten Anti-human-Antikörper nachgewiesen, wobei vorzugsweise Enzyme zur Markierung eingesetzt werden. Der als feste Phase dienende Träger kann unterschiedliche geometrische Formen aufweisen, wobei die bekannten Mlkrotitratlonsplatten besonders geeignet sind. Jedoch sind auch Röhrchen, kugelförmige oder flächenförmlge Träger einsetzbar. Die Bindung der antinukloären Antikörper erfolgt durch Inkubation des Serums in einer geeigneten Verdünnung mit den immobilisierten Zellkernen. Durch unterschiedliche Verdünnungen des zu bestimmenden Serums ist eine Ermittlung des Titers und somit eine semiquantitative Bestimmung möglich. Eine quantitative Bestimmung ist bei Vorhandensein eines Standards prinzipiell möglich. Zur Unterstützung unspezifischer Bindungen gestaltet sich der Einsatz von Puffern, die nichtionische oder ionische Detergenzien, Inertproteine, wie Rinderserumalbumin, ober aber Serum anderer Spezies, wie Kälborserum, enthalten als vorteilhaft.The isolation of the core antigens is carried out by means of pitch-gradient centrifugation, so that the antigenic structures are retained.
