Abstract

Eine andere Möglichkeit, Ca-Valproat oder andere in Wasser schwerlösliche Arzneistoffe zu retardieren, besteht in der Verwendung einer hydrophilen Matrix. So sind vor allem Zellulose-Matrices (US 4443428, DE 3045634, DE 3246492, DE 3309516 u.a.) und Einbettungen in verdauliche Hilfsstoffe bekannt. Die von Süß beschriebenen hydrophilen Matrices auf der Basis von Polyvinylalkoholen sind für die Retardierung von schwer löslichen Arzneistoffen —auch beispielsweise Ca-Valproat-nicht geeignet. Erfindungsgemäß wird diese Aufgabe dadurch gelöst, daß man 15 bis 85% Ca-Valproat mit einer Hilfsstoffmischung, bestehend aus 5 bis 95 Gewichtsteilen eines voll verseiften Polyvinylalkoholes mit 0 bis 3% Vinylacetatgehalt, einem k-Wert von 40 bis 60, i einer durchschnittlichen Molmasse von 60000 bis 80000 und einer Gesamtoberfläche von größer 0,1 m2/g und 95 bis 5 Gewichtsteilen eines teilverseiften Polyvinylalkoholes mit 10 bis 18% Vinylacetatgehalt, einem k-Wert von 50 bis 60, einer durchschnittlichen Molmasse von 80000 einer Gesamtoberfläche von größer 0,5 m2/g und einem spezifischen Porenvolumen von größer 0,2cm3/g vereinigt, der bezogen auf die Gesamtmasse dieser Arzneistoff-Hilfsstoffmischung 0 bis 10% Talkum und 0 bis 15% Magnesiumstearat zugesetzt werden; die Arzneiform kann auch durch Direktverpressung hergestellt werden. Überraschenderweise wurde gefunden, daß durch die erfindungsgemäße Mischung zweier Polyvinylalkohole mit unterschiedlichem Vinylacetatgehalt die Nachteile der Retardierungsverfahren auf der Basis diffusionsgesteuerter hydrophober und hydrophiler Matrices überwunden werden, und daß man durch das erfindungsgemäße Verfahren eine Ca-Valproat-Retardarzneiform erhält, die den Wirkstoff über einen gewünschten Zeitraum hinweg gleichmäßig und vollständig in vitro und in vivo freigibt (siehe Abb. 1).
