Die Erfindung betrifft ein Verfahren zur kontinuierlichen Züchtung von Saccharomyces cerevisiae zum Zwecke der Eiweißgewinnung auf der Basis von Essigsäuremutterlauge der Zelluloseazetatproduktion in Verbindung mit kohlenhydrathaltigen Rohstoffquellen. Die Erzeugung von Biomasse auf Kohlenhydratbasis ist zur Zeit bekanntlich vorwiegend auf die Verarbeitung verschiedener Abfallstoffe wie Melasse, Melasseschlempen, Zellstoffablaugen und deren Schlempen, Vorhydrolysaten usw. beschränkt. Im Interesse einer Erweiterung der Rohstoffbasis wurde deshalb die Verarbeitung von Säften zuckerhaltiger Pflanzen, z. B. aus Zuckerrübe oder Zuckerrohr bzw. anderer Produkte der Zuckerindustrie vorgeschlagen. Neben der stark in den Vordergrund gerückten Kohlenwasserstoff-Verhefung wurde u.a. auch die Verwendung von fettsäurehaltigen Abwässern der braunkohleverarbeitenden Industrie, der Paraffinoxydation sowie der Cyclohexanonproduktion vorgeschlagen.The invention relates to a process for the continuous growth of Saccharomyces cerevisiae for the purpose of protein production based on acetic acid mother liquor of the cellulose acetate production in conjunction with carbohydrate-containing raw material sources. Die anfallenden Mengen kohlenhydrathaltiger Abfallstoffe können den Bedarf für die Eiweißerzeugung nicht decken. Das Aufkommen an fettsäurehaltigen Abwässern ist gleichfalls beschränkt und es ergeben sich technologische Probleme aus dem z.T. recht niedrigen Gehalt an Fettsäuren sowie dem Vorhandensein von Phenolen und übergroßen Ammoniak-Mengen (Braunkohlenabwässer). Die früher als potentielle Möglichkeit erwähnte aber nicht durch Versuche belegte Verhefung von essigsäurehaltigen Abwässern der Zelluloseazetatproduktion führt nach eigenen Versuchen mit Saccharomyces-Hefen zu keiner technologisch praktikablen Lösung, da diese Abwässer Hemmstoffe bis zu solchen Konzentrationen enthalten können, die eine kontinuierliche Arbeitsweise nicht ermöglichen, da eine völlige Vermehrungshemmung auftritt. Zur Verarbeitung von Produkten der Zuckerindustrie ist zu bemerken, daß die verwendeten Kohlenstoffquellen sehr kostenintensiv sind; der Kampagnecharakter der Zuckerrübenverarbeitung bringt zusätzliche Probleme der Lagerhaltung der Folgeprodukte mit sich. Eine Vorbehandlung der Rohstoffe wie Aufschluß, Klären etc. ist notwendig. Im Verlaufe dieser Prozesse sind Substanzverluste durch Mikroorganismenbefall möglich. Andererseits wird ein Nebenprodukt der essigsäureverarbeitenden Industrie, das sich gut für eine mikrobielle Verwertung zum Zwecke der Eiweißsynthese eignet, mit relativ sehr hohen Kosten wieder aufgearbeitet. Bei der Erzeugung von Zelluloseazetat durch Reaktion von Zellulose mit Essigsäureanhydrid sowie technischer 99-100%iger Essigsäure werden als primäres Nebenprodukt größere Mengen einer ca. 20%igen verdünnten Essigsäure (auch als verdünnte Essigsäuremutterlauge bezeichnet) gewonnen, welche als anorganische Begleitstoffe vor allem Natrium- und Sulfationen enthält. Nach herkömmlicher Technologie wird die Essigsäure von diesen und anderen Begleitsubstanzen während des Aufkonzentrierungsprozesses mit Äthylazetat befreit, so daß am Ende dieses, wie schon erwähnt recht kostspieligen Prozesses, eine technische ca. 99-100%ige Essigsäure erhalten wird. Die vorliegende Erfindung ermöglicht die gleichzeitige Nutzung von Essigsäuremutterlauge (ζ. B. in Form des oben erwähnten primären Nebenproduktes der Zelluloseazetatproduktion) und Zucker als Kohlenstoffquellen für die aerobe Kultivierung von Saccharomyces cerevisiae mit hohen Zuwachsraten und günstigen Biomasseausbeuten. Die Essigsäurekomponente stellt dabei eine Kohlenstoffquelle dar, deren Produktion nicht von klimatischen Bedingungen abhängig bzw. beeinflußbar ist und keinen Kampagnecharakter aufweist. Die Nutzung von Essigsäuremutterlauge für die mikrobielle Eiweißsynthese stellt die Aufwertung eines Nebenproduktes der chemischen Grundstoffindustrie im Hinblick auf die Gewinnung proteinreicher Nahrungsmittel für die menschliche Ernährung dar. Die gleichzeitige Nutzung dieser Essigsäuremutterlauge mit bekannten Kohlenhydratquellen bringt im Vergleich zur alleinigen Verhefung von Essigsäuremutterlauge deutlich höhere bioökonomische Koeffizienten. Erfindungsgemäß erfolgt die kontinuierliche Verhefung von Essigsäuremutterlauge der Zelluloseazetatproduktion durch Saccharomyces cerevisiae, indem neben einer anteilmäßig in weiten Grenzen variierbaren Zuckerquelle (z. B. Melasse, Rohzucker oder andere Produkte der Zuckerwirtschaft einschließlich Zuckerrohrprodukte, Hydrolysate von Getreide, Holz oder Stroh) wie üblich eine Stickstoff- und eine Phosphatquelle in den Prozeß eingegeben und für ein ausreichendes Wuchsstoff- und Mineralstoffangebot Sorge getragen wird. Durch Wahl geeigneter Fermentationsparameter (Luftdurchsatz, Verdünnungsrate) wird unter aeroben Stoffwechselbedingungen ständig eine gleichzeitige fast hundertprozentige Assimilation der Essigsäuremutterlauge und des Zuckers erreicht, wodurch die Kultivierung von Mikroorganismen ermöglicht wird, die wie Saccharomyces cerevisiae gegenüber höheren Essigsäurekonzentrationen empfindlich sind. Die stoffliche Zusammensetzung von verdünnter Essigsäuremutterlauge der Zelluloseazetatproduktion bringt keine toxikologischen Bedenken gegenüber der darau s gewonnenen Biomasse, wodurch eine Verwendung dieser Kohlenstoffquellen für die Produktion von neuen proteinogenen Ausgangsmaterialien möglich wird.The resulting amounts of carbohydrate-containing waste materials can not meet the demand for protein production.
