Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung grobkristalliner 4-Amino-3-(p-chlorphenyl)-buttersaeure, die durch Behandeln der auf bekanntem Wege durch Hofmann-Abbau aus 3-(p-Chlorphenyl)-glutarsaeureamid herstellbaren mikrokristallinen Form mit Alkanol-Wasser-Mischungen bei erhoehter Temperatur hergestellt wird. Ohne Zwischenisolierung kann die Suspension der mikrokristallinen Form eingesetzt werden. Der Vorteil der Erfindung liegt in der Erhoehung der Ausbeute an Produkt in pharmazeutischer Qualitaet und in der Vereinfachung des Verfahrens, insbesondere in den Reinigungs- und Trennstufen. Nach Beendigung des Hofmann-Abbaus wird mit 22 Liter konzentrierter Salzsäure pH 6 eingestellt und die Suspensionen des mikrokristallinen Rohproduktes sofort mit 50 Liter Methanol = 30 Ma.-%, bezogen auf die bereits vorliegende flüssige wäßrige Phase versetzt. Anschließend wird bis zu einer Sumpftemperatur von 60°C unter Rühren geheizt. Nach 30min der Behandlung der ca. 23Ma.-% Methanol enthaltenden wäßrigen Mischung, bezogen auf alle Bestandteile derflüssigen Phase, hat sich ein grobkristallines Produkt gebildet. Nach Abkühlung auf Raumtemperatur wird dieses über ein Nutschenfilter abgetrennt und mit 30 Liter 50%igem wäßrigem Methanol nachgewaschen. Nachdem der Hofmann-Abbau beendet ist, wird mit ca. 245 ml konzentrierter Salzsäure auf pH 6 eingestellt und die gebildete Suspension des mikrokristallinen Rohrproduktes mit 700 ml Ethanol = 38Ma.-%, bezogen auf die bereits vorliegende flüssige Phase, versetzt. Danach wird bis zu einer Sumpftemperatur von 70°C erhitzt und 20 min bei dieser Temperatur gerührt. Nach dieser Behandlung hat sich in der ca. 27,5 Ma.-% Ethanol enthaltenen wäßrigen Mischung, bezogen auf alle Bestandteile der flüssigen Phase, ein grobes Kristallisat gebildet. Nach dem Abkühlen auf Raumtemperatur wird dieses auf einer Glasfiltemutsche abgetrennt, mit 400 ml 30%igem wäßrigem Isopropanol gewaschen und getrocknet.3. Nach Abkühlung auf 20°C wird dieses über ein Nutschenfilter abgetrennt, mit 30 Liter 50%igem wäßrigem Methanol nachgewaschen und getrocknet. Ausbeute: · 8,55 kg ^ 96,7 %d. Th., bezogen auf Amid4.
