Abstract
The invention relates to a method for the immobilization of antibiotics or other therapeutic or diagnostic substances on surfaces, in particular surgical auxiliary materials. The deposited substances are located as a crystalline precipitate on the surfaces to be loaded. The invention is based on the fact that antibiotics and other therapeutic or diagnostic substances which exhibit a p H-value-dependent solution behavior precipitate as a crystalline precipitate on the surfaces to be loaded when the p H value is changed, with chemical groups not more closely defined than Serve crystallization cores. The crystals adhere firmly to the surface and the substances are even after extensive rinsing for several days delivered to the environment in sufficient concentration.
Description
Verfahren zur Immobilisation von Antibiotika an Oberflächen·Process for the immobilization of antibiotics on surfaces
Anwendungsgebiet der Erfindung:Field of application of the invention:
The invention relates to a method for the immobilization of antibiotics or other therapeutic or diagnostic substances on surfaces of surgical sutures, catheters or dressings for use in human or veterinary medicine.
Characteristic of the known technical solutions: To provide surfaces of materials for implantation (surgical suture material, catheters, allografts or dressing materials, hereinafter referred to as materials) with antibacterial, antifungal properties, or these materials with other diagnostic or therapeutic substances (hereinafter referred to as substance It has hitherto been customary to load the materials into a solution of the substances in question and to use them immediately thereafter. Catheter material had to be rinsed with the appropriate substance solutions immediately before or during use (with the catheter lying).
The substances deposited thereby quickly diffuse into the surrounding tissue or into the body fluid and are therefore only available for a short time at the place of application.

,Wax (US Pat. 2751910/1956), or by introducing antibiotic-containing foam or powder in the suture (US Pat. 3674901/1972, A 61 L 17/00) try to accumulate large amounts of the substances to the surface of materials the danger that the substances are either chemically altered and thereby lose their activity, or that the bioavailability of these substances is considerably limited. Since some 'of the processes mentioned in each case only "a: molecule of the substance to a binding site of Mate. Is rials annealed, the annealed amount of substance is severely limited a required minimum Wixkstoffkönzentration in the immediate vicinity of the material can not be achieved.
On the other hand, by particularly firm binding of the substance or its inclusion in a polymer, the period of delivery of the substance can be considerably extended. This is associated with a loss of drug concentration. Particularly in the case of the prophylactic use of antibiotics, it is necessary to deliver an effective amount of substance at the time of the greatest risk of infection and only for a few days in order to avoid disadvantages such as toxic local damage (wound healing disorder), toxic-r-allergic effects and resistance development.

Darstellung des Wesens der Erfindung: , 'Presentation of the essence of the invention:
The invention is based on the provision of a method which is characterized by a change in pH in an aqueous solution of an antibiotic or other therapeutic or diagnostic, which zur Konzentrationsände-. According to the invention, the object of loading surgical auxiliary materials with antibiotics or therapeutic or diagnostic substances is achieved in that the material to be loaded is placed in an aqueous solution of the antibiotic or another dissociable substance. By gradual addition of a strongly acidic or strongly alkaline buffer, depending on whether the antibiotic, therapeutic or diagnostic substance is an anion or a cation, the substance precipitates precipitously in the form of crystals.
The pH of the solution is 5.5.
Under sterile conditions, several Teflon pieces are placed in the oxacillin solution.
By adding an acidic KCl / HCl buffer pH 1.0 (25 ml 0.2 η KCl + 54 ml 0.2 η HCl, water ad 100 ml), the lowering of the pH initially takes place rapidly up to a Value of 1.8, then slowly over a period τοη 1O minutes to pH 1.6. The addition of the buffer is carried out under constant Euhr and under constant pH control. During the procedure, an ultrasonic finger dives into the solution, which generates an ultrasonic field of 170 watts and 0.02 MHz. After the procedure, the teflon pieces are removed from the oxacillin solution. The adherent solution comes with. Sterile filter paper removed and the Teflon pieces come in a rinsing cell, which is traversed over several days of sterile water.
