The invention relates to a process for the separation of unwanted constituents from undried biomass, which is produced by culturing microorganisms on hydrocarbon-containing substrates and treating the fermentation broth by means of mechanical separation processes.
The growth of microorganisms on hydrocarbonaceous substrates produces biomass suspensions contaminated by unutilized hydrocarbons.
Before biomass can be used as a feed, these unutilized hydrocarbons and biogenic lipophilic cell ingredients, such as fatty acids and glycerides, must be removed. Numerous methods are known for purifying biomass suspensions and dry biomass. According to DD-WP 139 650 dry powdery biomass is extracted with a solvent mixture consisting of hydrocarbons with alcohols, with water or with alcohols and water in countercurrent to improve the Absinkverhaltens and wettability of the particles.
Hach DD-W? 139 651 is contacted with the same objective a biomass suspension with predominantly poorly decreasing solid particles with a solvent mixture consisting of hydrocarbons with alcohols, with water or with alcohols and water, the hydrocarbons fractions preferably in the boiling range of 40 140 0 G.
According to FR patent 1381311, the biomass is subjected to a two-stage extraction.
In the first extraction stage, which may consist of several sub-processes, the biomass is treated with 1.5 to 3 times the amount of alcohol, based on the water content of the biomass suspension, greatly reducing the water content and the content of impurities. In a second extraction stage, which may also consist of several partial processes, v / erden with the 2- to 20-fold amount of a solvent mixture, which is composed of the alcohol used in the first extraction stage and a hydrocarbon, for example η-hexane, residual hydrocarbons and biogenic lipophilic cell ingredients. After extraction, the solvent is removed by evaporation, possibly also under reduced pressure, it is disadvantageous that in addition to the hydrocarbons and undesired cell constituents, for example fatty acids, also nutritionally valuable cell ingredients, such as vitamins and phosphatides, are extracted in these processes, resulting in reduced yields biomass with simultaneous reduction in quality. Another important drawback of the process is the high extraction agent requirement and the resulting high energy expenditure, which is required for regenerating the extractant.
The aim of the invention is the separation of unwanted Beatandteilen from undried biomass, which is produced by 4 growing microorganisms on hydrocarbon substrates by means of a product-friendly, cost-effective and compared to known methods technologically less expensive method for obtaining a veterinary requirements corresponding biomass.
The object underlying the invention lies in the development of a novel process for the separation of unwanted constituents from undried bio-mass, which works effectively, gently and cost-effectively, and in particular serves to remove hydrocarbons and qualitatively reducing biogenic cell constituents. The object of the invention is achieved in that the separation of unwanted constituents from undried biomass with a water content of 95-55 wt .-%, which is produced by culturing microorganisms on hydrocarbonaceous substrates, by contacting with carbon dioxide under supercritical conditions up to 453 fifth 15 K and 40.53 MPa.
The volatile hydrocarbons and the biogenic lipophilic cell constituents, such as, for example, fatty acids and glycerol esters, are selectively dissolved by means of supercritical carbon dioxide, with the formation of a loaded extract phase. The equilibrium concentration of the extracted components in the loaded phase phase is greater by several orders of magnitude in the supercritical range than in the supercritical range.
kritischen Bereich. This effect is based on the fact that the high density of the supercritical extractant and the resulting increased interaction between the components increase the solubility of the volatile components by a multiple.
The inventive method allows in comparison to previously known methods including extraction agent recovery a reduction in the specific energy consumption.
The process can be carried out continuously or discontinuously due to the process.
The contact of the supercritical carbon dioxide with the biomass to be purified can be intensified by intensive stirring or shaking. The carbon dioxide is contaminated with hydrocarbons and cell contents and is discharged with these from the Estraktor and passed into a second apparatus in which the extracted components are deposited by changing the pressure and / or temperature. The uncharged carbon dioxide is returned to the extractor. The extraction time and the ratio ·. Carbon dioxide to biomass are measured so that the required purity of the biomass is achieved.
The purified biomass is further processed by known methods.
The biomass purified by the process according to the invention is distinguished from the biomass produced by known processes in that undesirable components such as hydrocarbons, fatty acids and glycerol esters have been removed, but nutritionally valuable components remain in the biomass, so that a quality improved in its bio-composition, with the ultimately more feed
A particular advantage of the method is that the consistency of the biomass suspension remains essentially unchanged during the extraction process. As a result, the technical equipment and technological complexity can be kept low in comparison with known methods, in particular when using pumpable suspensions.
