Abstract
An injectable solution of [i23I]-labeled iodoamphetamine is used in nuclear medicine for the determination of cerebral blood flow. The method of preparing iodoamphetamine labeled with [123I] consists in adding palladium chloride as a catalyst to a solution of non-radioactive iodoamphetamine, a radioactive solution of [123I] sodium iodide, and heating the reaction mixture to 100 °C for 20 minutes. The use of palladium chloride as a catalyst is new, which significantly reduces the temperature of the reaction mixture. This prevents the formation of decomposition products and shortens the time required for the reaction.
Description
Vynález se týká způsobu přípravy injekčního roztoku jodamfetaminu (IAMP) značeného radioaktivnímu izotopy jodu. The present invention relates to a process for the preparation of an iodamphetamine (IAMP) solution for injection labeled with radioactive iodine isotopes.
IAMP značený radioaktivními izotopy jodu je možné připravit celou řadu způsobů. Pro případ I123, vzhledem k jeho krátkému poločasu rozpadu, z časových důvodů, neboť 123I má krátký poločas 13,,3 hod, je prakticky výhradně používána výměnná reakce mezi organickým jodderivátem a anorganickým [123I] jodidem sodným.IAMP labeled with radioactive iodine isotopes can be prepared in a number of ways. For case I 123 , due to its short half-life, for time reasons, since 123 I has a short half-life of 13, 3 hours, the exchange reaction between the organic iodo derivative and the inorganic [ 123 I] sodium iodide is practically exclusively used.
Značně složitý způsob přípravy IAMP, popisuje Kuhl D. E. a spolupracovníci v J. Nucl. Med. 23 (1982), str. 196 až 203 a Lassen N. A. a spol· v J. Nucl. A considerably complex process for preparing IAMP is described by Kuhl D.E. and co-workers in J. Nucl. Copper. 23 (1982), pp. 196-203 and Lassen N.A. et al., J. Nucl. Copper. 24, (1983), pp. 17-21. This method is based on melting the org. with iodide at 150 ° C for 30 minutes. Its main disadvantage is that it can lead to decomposition products. Radioactive IAMP is isolated from the reaction mixture by repeated extraction with ether.
J. Nucl. Med. 7, (1982), str. 820, Kunst E. J. a spol. v Third International Symposium on Radiopharmacology, Frelburg, Sept. 1983. Eli P· J. a spol. v Eur. J. Nucl. Med,8, (1983), str. 114 a Kunst E. J. a další v Nukleařmedizin 23, (1984), str. Somewhat more preferred is a preparation process in which catalysis of the exchange reaction with CuI salts is used to improve the yield and purity of the preparation . as reported by Carlsen L. and Andersen K. in Eur. J. Nucl. Copper. 7, (1982), p. 820, Kunst EJ et al. at the Third International Symposium on Radiopharmacology, Frelburg, Sept. 1983. Eli P. J. et al. v Eur. J. Nucl. Med., 8, (1983), p. 114 and Kunst EJ et al. In Nuclearizin 23, (1984), p. 31-34. The reaction takes place at 175 ° C in acetic acid for 30 min. As it does not proceed quantitatively, the product must be purified from unreacted [ 123 I] sodium iodide by high pressure liquid chromatography.
Využití katalytických vlastností Cu1 pro výměnnou reakci popisuje Merteus a spol, v J. Labelled Compd. RAdiopharm. 22 (1985), str. 89 až 93, kde CU1 ionty připravuje redukcí chloridem cínatým přímo v reakční směsi· Nevýhodou tohoto způsobu jsou vysoké používané teploty a nutnost odstraňování Cu1 vysokotlakou kapalinou chromatografií.The use of the catalytic properties of Cu 1 for the exchange reaction is described by Merteus et al. In J. Labeled Compd. RAdiopharm. 22 (1985), pp. 89-93, wherein Cu 1 ions are prepared by reduction with stannous chloride directly in the reaction mixture. The disadvantages of this method are the high temperatures used and the necessity to remove Cu 1 by high pressure liquid chromatography.
P. Angelberg a spol. popisují v J. Lab. Comp. Radiopharm. 21, (1984), str. 1110 až 1111 výsledky různých způsobů příprav IAMP, při nichž se teplota reakčních směsí pohybovala v rozmezí 120 až 160 °C, doba zahřívání 30 až 90 minut a výtěžky v rozmezí 7 až 85 %.P. Angelberg et al. described in J. Lab. Comp. Radiopharm. 21, (1984), pp. 1110-1111 results in various IAMP preparation methods in which the temperature of the reaction mixtures was 120-160 ° C, the heating time was 30-90 minutes, and the yields were 7-85%.
Obecnou nevýhodou všech těchto metod jsou nízké výtěžky, vysoké používané teploty vedoucí k částečnému rozkladu látky a nutnost složitého čištění produktu. The general disadvantage of all these methods is low yields, high temperatures used leading to partial decomposition of the substance, and the need for complex product purification.
Reakce probíhá s vysokým výtěžkem (95%) bez vzniku nežádoucích vedlejších produktů a případně přítomný nezreagovaný [123I] jodid sodný lze velmi snadno* odstranit filtrací produktů přes papír sycený chloridem stříbrným.The main of these disadvantages - the high temperature - is eliminated by the process of preparing the IAMP of the invention, wherein the palladium chloride catalyst is added to the non-radioactive iodamphetamine solution, the radioactive [ 123 I] sodium iodide solution and the reaction mixture is heated to 100 ° C for 20 minutes and IAMP is formed. The reaction proceeds in high yield (95%) without the formation of unwanted by-products and any unreacted [ 123 I] sodium iodide present can easily be removed by filtering the products through silver-saturated paper.
Mezi hlavní výhody způsobu podle vynálezu patří použití chloridu paládnatého jako katalyzátoru, což umožňuje podstatně snížit teplotu reakční směsi, tím zabránit vzniku rozkladných produktů a zkrátit dobu potřebnou pro reakci. The main advantages of the process according to the invention include the use of palladium chloride as a catalyst, which makes it possible to substantially reduce the temperature of the reaction mixture, thereby avoiding the formation of decomposition products and reducing the time required for the reaction.
The preparation is used in nuclear medicine to determine local cerebral blood flow, taking advantage of the fact that its uptake from blood to brain tissue is rapid, the ratio of radioactivity in the brain to blood is high and the elution is slow. The use of this preparation in nuclear medicine makes it possible to refine the diagnosis in some very serious cerebral diseases such as schizophrenia, manic-depressive psychoses and the like.
PříkladExample
Κ 1 ml vodného- roztoku chromatograficky čistého N-isopropyl-p-jodamfetaminu o koncentraci 3,4 mg/ml se přidá 10 μΐ roztoku chloridu paládnatého o koncentraci 1,2 mg/ml.
