Abstract

La invención proporciona un sistema de secuestro de metal pesado mediante bacterias. La bacteria expresa los genes ppk, mt, y/o P-galactosidasa (lacZ) y puede tolerar por lo menos 25 µM de mercurio, 1,000 µM de zinc, 250 µM de cadmio, y 3,000 µM de Pb. El sistema permite la determinación fácil de la presencia de contaminantes de metal pesado en un líquido y la recolección fácil de la bacteria que ha secuestrado grandes cantidades de metal pesado. Se proporciona adicionalmente un sistema de expresión de gen en bacterias que comprende elementos de expresión de gen de fago y plástido y suministra un nivel de expresión de proteína particularmente alto y resistencia al metal pesado.
