Abstract

人羊膜基底膜侵袭模型,是一种肿癌研究的专用容器,包括玻璃圆杯体1、筒2和罩3,其特征是杯1中放置的筒2一端有向外扩展的沿,其外径与杯体1的直径相等,沿的内径是筒2的内径,筒2的长度短于杯体1,罩3的直径大于杯体1罩的沿可下沿。它可用来观察肿瘤的侵袭过程、粘附性等,同时也易制成各种标本;便于用它筛选高侵袭癌细胞亚系,如需较长时间的细胞培养,因该容器操作简便,易于消毒,易于更换培养液,污染机会较现有方法少,成功率高。

Description

人羊膜基底膜侵袭模型
本实用新型涉及B65D 3/00类,一种医学实验用容器,人羊膜基底膜侵袭模型。
在基础医学研究领域中,肿瘤侵袭研究除需要科学的研究方法外,更需要良好的模型作保障。Liotta提出了人羊膜基底膜的侵袭模型(New method torprepairing large surface of intact human basement membrane tor tumorinvasion studies.″Cancer Lett″,1980,11:141),用于体外癌细胞侵袭行为的研究。但是他在文中仅仅是提出一种操作原则,并没有具体化。从许多国外文献上也只能看到模式图,以说明其原理,并没有实物图型,国内更无侵袭模型商品。国内研究者采用了一些方法自制该模型,如两个大小相近的试管或蛋壳相套、两块24孔培养板叠加在一起,去除上层培养孔底等。这在消毒、减少污染及换液等方面存在一定的困难,使该项研究有一定难度。
本实用新型的目的是提供一种用于肿瘤研究的专用容器,人羊膜基底膜侵袭模型。
该模型包括玻璃圆杯体1、筒2和罩3,其特征是杯1中放置的筒2一端为向外扩展的沿,沿的外径与杯体1的直径相等,沿的内径是筒2的内径,筒2的长度短于杯体1,罩3的直径大于杯体1的直径,罩3的沿可沿杯体1延展。
该模型为玻璃制品,便于观察,可用来观察肿瘤的侵袭过程、粘附性等,同时在制作光镜和电镜标本时,羊膜易取下,容易制成各种标本;便于用它筛选高侵袭癌细胞亚系,如需较长时间的细胞培养,因该容器操作简便,易于消毒,易于更换培养液,所以污染机会较现有方法少,成功率高。

实施例同上述方案,省略。
使用方法:参照Liotta及Mignatt法对羊膜进行处理,取足月正常分娩或剖腹产新鲜胎盘,立即剥离羊膜,用PH7.2的PBS-PS(含青霉素1000u/mL,链霉素1000u/mL)缓冲液充分清洁后,室温下0.25M氢氧化胺溶液浸泡2小时,溶解表层细胞,然后在无菌条件下于超净工作台上用橡皮铲轻轻地刮去羊膜上残余的表皮,再经灭菌的的PBS-PS缓冲液充分清洗后,置于含PS的细胞培养液中,4℃保存。该玻璃器皿高压消毒,将制备好的羊膜剪成适宜大小,包于筒2的无沿端,用消毒橡皮套扎紧,将此端向下置于杯1中,如图2所示,在筒2和杯1中加入细胞培养液,将生长状态良好的细胞制成所需细胞数的细胞悬液,取2mL接种于筒2内,盖上罩3。根据实验设计所需的时间,观察所需要的内容,取下的羊膜可根据常规方法制成标本,进行光镜、电镜观察,侵袭羊膜后穿过的细胞落于杯1内,进一步进行传代,可获得高侵袭癌细胞亚系。

1、一种人羊膜基底膜侵袭模型,包括玻璃圆杯体(1)、筒(2)和罩(3),其特征是杯(1)中放置筒(2),筒(2)的一端为向外扩展的沿,沿的外径与杯体(1)的直径相等,沿的内径是筒(2)的内径,筒(2)的长度短于杯体(1),罩(3)的直径大于杯体(1)的直径,罩(3)的沿可沿杯体(1)延展。
