本检测药盒使用方法简便,灵敏、快速、所用血清样品少,容易为患者所接受。

Description

活性维生素D检测药盒。
本发明属生物化学领域。
活性维生素D也称为1.25-双羟胆钙化醇。(1.25-Di-hydrOXychO    LecaLciferOL.DHec)。是维生素D在体内经肝脏和肾脏羟化后转变而成的一种高活性形式。在正常情况下,体内合成量充足,合成后经血液运输到靶器官,通过受体而发挥作用,所以它是一种新的甾族激素。活性维生素D参与免疫调节,促进某些肿瘤细胞(如白血病细胞)的正常分化以及对生长素,泌乳素分泌等都有重要影响,对佝偻病的早期诊断和防治,对老年性骨病和某些内分泌疾病的研究有应用价值。同时,血清中活性维生素D含量的检测对奶牛产奶率,家禽产蛋率,淡水养鱼及实验动物的饲养条件,饲料配方等均有重要的指导意义。
活性维生素D检测测方法国外已开展,但方法的技术难度较大,需要昂贵的仪器设备,不经专门的训练难以掌握操作技术。受体蛋白制作技术复杂,周期长,不稳定,测量血清量大,不易为患者所接受。目前国内尚没有理想的测试手段和方法,更没有试剂盒的供应和使用。
本发明的目的是为了在国内开展活性维生素D检测技术,制备国产化试剂盒,使此项超微量检测技术尽快地为经济建设服务。
本实用新型的结构示意图由附图给出。图1为盒盖,图2为盒底。盒内容物装入盒底(1)中,内容物包括:受体蛋白(2),标准品(3),标记物(4),磷酸缓冲液(5),活性炭(6),各种试剂的制备如下:
1.1.受体蛋白(2)的制备:以正常兔小肠胞浆受体蛋白制备为例。用日本大白兔活体积取小肠环,灌洗肠腔,纵向剖开,刮下粘膜层,用2倍体积的高渗缓冲液制备匀浆,离心,取上清液超速离心,上清液测蛋白浓度后,分装,通氮条件下抽干,冷冻于玻璃瓶中。
2.标准品(3)的制备:以结晶维生素D3为原料,采用体外羟化的方法进行孵育,经葡聚糖凝胶LH-20柱层析及高压液相分离,制得1.25-(OH)-D,分装于玻璃瓶中。
3.标记物(4)的制备:活性维生素D的氚标记物采用低温交换法。以催化剂为钯,氚离子由氚分子激发获得。粗标记样品经真空脱游离氚后,经葡聚糖凝胶LH-20柱层析分离,纯化,分装于玻璃瓶中。-20℃水箱保存。比活性:87ci/mmOL。
4.磷酸缓冲液(5)的配制:用磷酸二氢钠和磷酸氢二钠加氮化钠溶于重蒸水中,溶均即可,分装于玻璃瓶中。
5.活性炭(6)的制备:取活性炭经酸,碱及重蒸水清洗后烘烤,分装瓶中,密封保存。
上述试剂装入盒底,用泡沫塑料固定,封上盖即可。
使用时取静脉血离心,取血清1mL,按侧定程序及计算公式便可可测定血清中活性维生素D含量。本试剂盒测定正常值为20-65pg/mL。
本发明药盒使用方法简便,灵敏,快速,不需要高效液相等昂贵贵的仪器,一般市级医院均可开展此项技术。检测方法稳定,精密度及回收率均合格,所用血清样品量少,容易被患者所接受。

1、一种由盒底(1)和盒盖组成的活性维生素D检测药盒,其特征在于盒底内受体蛋白(2),标准品(3),标记物(4),磷酸缓冲液(5),活性炭(6)所组成。
