Abstract

Description
背景技术
切向流过滤(TFF)是使用膜来根据尺寸或者分子量不同来分离液体溶液或者悬浮液中的成分的分离过程。应用包括蛋白质和诸如核苷酸、抗原和单克隆抗体之类的其它生物分子的浓缩、澄清,以及脱盐;缓冲剂交换;过程发展;膜选择研究;预先色谱分析澄清来去除胶体微粒;诸如右旋糖和抗生素之类的小分子的去除热原法;细胞培养物、溶胞产物、胶体悬浮液和病毒培养物的收获、洗涤或者澄清;以及试样制备。
在TFF中,要过滤的溶液或者悬浮液以交叉流的模式通过膜的表面。用于过滤的驱动力是跨膜压力,其通常由蠕动泵产生。滤出液通过膜表面的速度也控制过滤速度,且有助于防止膜的阻塞。因为TFF穿过膜表面再循环滞留物,所以最小化膜积垢,维持了高过滤速率和增强了产品回收。
传统的TFF装置由多个元件形成,包括泵、供给溶液容器、过滤模块和用于连接这些元件的导管。在使用中,供给溶液从供给溶液容器引导到过滤模块,同时来自过滤模块的滞留物从过滤模块再循环到供给溶液容器,直到获得需要量的滞留物。膜夹在顶部和底部歧管或者保持器之间,该歧管或者保持器用于提供精确的机械约束防止装置的内部液压压力,以及也用于分配穿过装置内的多个流道的过滤流。这些歧管或者保持器通常由不锈钢制成,且必须在每次使用前清洁和验证,特别是在生物制药学和其它卫生的应用中。这是昂贵的和费时的过程。
需要提供一种过滤装置,其在更换过滤介质时不需要如前所述的清洁和验证步骤。
发明内容
根据本教示,提供了一种切向流过滤装置,其中,在过滤元件与顶部和底部保持器或者歧管之间提供衬垫。该衬垫包括原先存在于歧管中的流通道以及入口和出口端口。该衬垫由不贵的材料制成,因此在单次使用后可任意处理,使得处理它们比清洁传统的歧管更加节约成本。另外,衬垫可以是预先消毒的。

具体实施方式
首先参考图1,显示了根据本教示的过滤装置10的分解视图。装置10包括顶部保持器板12和隔开的底部保持器板13。保持器板12、13优选由不锈钢制成,且充分刚性和耐久,以提供组件的精确和有效的机械约束,防止内部液压操作压力,诸如50-60磅/平方英寸。孔隙28设置在保持器板12、13中,且在组件的每一层中,以容纳连杆或者带螺纹的销或者螺栓14或者其它夹紧装置,以将组件固定在一起。提供垫片15,且其可以是弹簧加载的。在保持器板12、13中不存在过滤流通道。
在组装状态下定位在保持器板12下的是一次性的衬垫16。衬垫16优选由不贵的材料制成,适合于应用,其对于药物检定是可接受的(且优选是政府批准的)。结构的合适材料包括塑料,诸如聚苯乙烯,优选为聚烯烃,诸如聚丙烯、聚乙烯、其共聚物和混合物。衬垫16优选模制有通道和开口。替代地,且较不优选的,其可以通过铣磨、钻孔和其它这样的方法来形成。
如图2-5最好的显示,衬垫16包括第一端口17A、五个子端口17C、第二端口17B和四个子端口17D。端口17A根据其在组件内的定向用于引入供给物或者去除滞留物,端口17B用于去除渗透物,而防止滤出液与滞留物或者供给物混合,如传统的那样。端口17A连接到以歧管排列的五个子端口17C。端口17B以类似方式连接到四个子端口17D。优选的,端口17A和17B位于衬垫的相对侧,如所示的,以便提供足够的间隔,以及避免与其它部件干扰。然而,在其它应用中,特别是在间隔充分或者没有干扰出现的情况下,它们可以位于相同侧。每个端口17A、17B与流道或者通道流体连通,该流道或者通道与相应的孔隙连通,以容纳供给物、滞留物或者渗透物的流,如传统的那样,从而限定用于装置内的过滤流的多个流道。
如图3所示,当通道离开它们相应的端口前进时,它们优选是逐渐变细变窄的,以在子端口17C和17D的每一个处标准化压力。
返回图1,显示了定位在衬垫16下的过滤元件20。过滤元件20可以是单一的膜,且优选是多个堆叠的膜,诸如堆叠的超滤或者微滤膜,最优选以盒的形式设置。尽管显示了膜的单一盒,本领域中的普通技术人员可以理解,可以使用多个盒。合适的盒以名称PELLICON销售,并且从Millipore公司商业上可获得。
定位在过滤元件20下的是第二衬垫22。优选的,第二衬垫22在结构上与第一衬垫16相同,但当装置处于组装状态时,衬垫22相对于第一衬垫16的位置倒转,如所示的。这允许在其正常定向中端口17A与装置的供给端口连通,而在倒转位置中时与滞留物端口连通。衬垫的端口17B在两种定向中都与渗透物端口连通。
优选的,衬垫16、20的一侧包括多个肋,如图2和3最好的显示。该肋为衬垫提供增加的刚性,且在模制过程中形成。肋定位在衬垫接触保持器板的侧上。在显示的肋构造中,横向肋25A通过衬垫的中心延伸衬垫的宽度。肋25B也延伸通过衬垫的中心,垂直于肋25A。衬垫的中心由圆形肋25C确定界限,且两个相对且对称组的四个隔开的肋25D、25E、25F和25G从圆形肋25C延伸到衬垫的周边,如所示的。本领域中的普通技术人员可以理解,显示的实施例只是示例性的目的;其它肋构造在本发明的范围内。
为了在本发明的过滤装置中正确和有效组装衬垫,保持器板12、13优选包括凹槽26的构造,其相应于邻近它的衬垫中的肋的构造的图案。当正确对准时,每个衬垫中的肋匹配每个相应保持器板中的凹槽,以形成可以在压力下密封的集成的整体的组件。
Claims (1)
1.过滤设备,其包括:
顶部板;
与所述顶部板隔开的底部板;
定位在所述顶部板和所述底部板之间的过滤构件;
定位在所述顶部板和所述过滤构件之间的第一一次性的衬垫,所述第一衬垫具有流体入口、流体出口、多个流体入口孔隙和在其一侧上面向所述顶部板的肋的图案;
定位在所述底部板和所述过滤构件之间的第二一次性的衬垫,所述第二衬垫具有流体入口、流体出口、多个流体出口孔隙和在其一侧上面向所述底部板的多个肋;
其中,所述顶部板和底部板没有流体流道,且每个包括多个凹槽,这些凹槽匹配相应衬垫上的多个肋。
