本发明涉及微生物发酵生产领域,特别是指一种高酰基结冷胶的后提取方法。包括发酵液预处理、絮凝沉降,固液分离、干燥、粉碎等工艺步骤,本发明解决了高酰基结冷胶目前只有美国CPkeclo公司生产,价格昂贵的技术问题,具有整体工艺简单,整体方法中能耗低、原材料消耗少、生产成本大大降低,产品质量可以达到美国FccIV标准等优点。

Description

高酰基结冷胶的后提取方法
技术领域
本发明涉及微生物发酵生产领域,特别是指一种高酰基结冷胶的后提取方法。
背景技术
结冷胶是一种由伊乐藻假单胞杆菌对碳水化合物进行纯种发酵后,经提取精制而得的多糖胶质。它是一种全透明的凝胶剂。结冷胶有两种存在形式:一种是天然的,结冷胶主链上接有酰基,即高酰基结冷胶;一种是低酰基的,结冷胶主链上酰基被脱除或部分脱除。
高酰基结冷胶柔软、富有弹性、粘着力强,不易失水,持水性能好,具有用量低,弹性可调节,良好的配伍性,优异的热稳定性、耐酸、抗酶解等性能,广泛用于食品、医药、日化等行业,是卡拉胶、海藻酸胶等凝胶剂的理想替代品。
高酰基结冷胶,目前只有美国CPkeclo公司生产,价格昂贵,在国内专利文献中未见报道。
发明内容
本发明的目的在于通过优化工艺、降低生产成本等手段,提供一种高酰基结冷胶的后提取方法以克服现有技术存在的不足。
本发明的整体技术构思是:
高酰基结冷胶的后提取方法,包括发酵液预处理、固液分离;提取方法中各工艺步骤和工艺条件是:
(1)发酵液预处理:
将发酵液调PH值4-4.5,升温90±2℃,灭酶杀死菌体,维持10-30分钟后降至室温;
(2)絮凝沉降,固液分离:
往预处理后的发酵液加入占发酵液体积2-3%的电解质溶液,反应30-40分钟,电解质溶液选用碱金属氯化物的水溶液,加入发酵液体积的25-30%、浓度为90%以上的异丙醇后搅拌均匀,固液分离;
(3)干燥、粉碎:
将步骤(2)中制成的产物于80-85℃条件下真空干燥,粉碎得高酰基结冷胶粉末产品。
本发明中各工艺步骤中的工艺条件是:
步骤(2)中碱金属氯化物的水溶液浓度为10%。
步骤(2)中的碱金属氯化物的水溶液选用Cacl2、Nacl、Alcl3水溶液中的一种。
步骤(2)中碱金属氯化物的水溶液优选Cacl2水溶液。
步骤(2)中的固液分离设备选用卧式螺旋卸料离心机。
步骤(3)中真空干燥的真空度为-0.08mpa。
步骤(3)中真空干燥的时间为2小时。
本发明所取得的实质性特点和显著技术进步在于:
1、本发明的整体工艺简单,只包括有发酵后预处理,絮凝沉降,固液分离,干燥粉碎。
2、整体方法中能耗低、原材料消耗少、生产成本大大降低,表现在发酵液预处理维持高温时间短,不使用酶制剂,絮凝沉降,固液分离时不用助滤剂、吸附剂。
3、产品质量可以达到美国FccIV标准。
具体实施方式
以下对本发明的实施例做进一步描述:
实施例
高酰基结冷胶的后提取方法,包括发酵液预处理、固液分离;提取方法中各工艺步骤和工艺条件是:
(1)发酵液预处理:
将发酵液调PH值4.2,升温至92℃,灭酶杀死菌体,维持20分钟后降至室温;
(2)絮凝沉降,固液分离:
往预处理后的发酵液加入占发酵液体积2.5%的电解质溶液,反应36分钟,电解质溶液选用碱金属氯化物的水溶液,加入发酵液体积的2 8%、浓度为90%以上的异丙醇后搅拌均匀,固液分离;
(3)干燥、粉碎:
将步骤(2)中制成的产物于83℃条件下真空干燥,粉碎得高酰基结冷胶粉末产品。
本实施例中各工艺步骤中的工艺条件是:
步骤(2)中碱金属氯化物的水溶液浓度为10%。
步骤(2)中碱金属氯化物的水溶液选用Cacl2水溶液。
步骤(2)中的固液分离设备选用卧式螺旋卸料离心机。
步骤(3)中真空干燥的真空度为-0.08mpa。
步骤(3)中真空干燥的时间为2小时。

Claims (7)

1、高酰基结冷胶的后提取方法,包括发酵液预处理、固液分离;其特征在于提取中各工艺步骤和工艺条件是:
(1)发酵液预处理:
将发酵液调PH值4-4.5,升温90±2℃,灭酶杀死菌体,维持10-30分钟后降至室温;
(2)絮凝沉降,固液分离:
往预处理后的发酵液加入占发酵液体积2-3%的电解质溶液,反应30-40分钟,电解质溶液选用碱金属氯化物的水溶液,加入发酵液体积的25-30%、浓度为90%以上的异丙醇后搅拌均匀,固液分离;
(3)干燥、粉碎:
将步骤(2)中制成的产物于80-85℃条件下真空干燥,粉碎得高酰基结冷胶粉末产品。
2、根据权利要求1所述的高酰基结冷胶的后提取方法,其特征在于所述的步骤(2)中碱金属氯化物的水溶液浓度为10%。
3、根据权利要求1或2所述的高酰基结冷胶的后提取方法,其特征在于步骤(2)中的碱金属氯化物的水溶液选用Cacl2、Nacl、Alcl3水溶液中的一种。
4、根据权利要求3所述的高酰基结冷胶的后提取方法,其特征在于所述的碱金属氯化物的水溶液优选Cacl2水溶液。
5、根据权利要求1所述的高酰基结冷胶的后提取方法,其特征在于所述的步骤(2)中的固液分离设备选用卧式螺旋卸料离心机。
6、根据权利要求1所述的高酰基结冷胶的后提取方法,其特征在于所述的步骤(3)中真空干燥的真空度为-0.08mpa。
7、根据权利要求1所述的高酰基结冷胶的后提取方法,其特征在于所述的步骤(3)中真空干燥的时间为2小时。
