Abstract

Description
发明的背景
通常可取出的假牙、牙板等包括固定在合适板或基托内的牙。使用假牙稳定剂填充假牙与牙龈或组织之间的间隙。假牙放入口腔之前,将假牙稳定剂放到假牙板表面上,为了装配完好,它应当均匀地接触牙龈和粘液组织。假牙稳定剂不仅配制成具有粘合性,而且要在假牙与牙龈或组织之间提供衬层或衬垫,由此使假牙牢固地定位于口腔内。
数年来,已经作了很大努力开发改进的假牙粘合剂组合物。单独或组合使用了合成和天然聚合物和树胶,而且与各种粘合剂和其他材料结合使用以克服某些缺陷。这些缺陷包括保持力不足、在放入和戴上期间粘合剂从牙板下渗出和从口腔和假牙上脱除残留粘合剂很凌乱和很困难。另外,食物也会留在假牙与戴者的口腔之间。

根据本发明,发现假牙粘合剂组合物含有烷基乙烯基醚-马来酸共聚物和/或含异丁烯的三元共聚物的镁与锌混合盐,所述混合盐还含有规定量的游离酸,可提供优良的假牙保持力。本发明的假牙粘合剂组合物也可以有效地用作伤口敷料、水下粘合剂、生物粘合剂、释放其他活性物质的载体和/或任何其他需要粘合到皮肤或组织的应用,包括美容用的毛孔清洁带。
本发明的目的是提供用于牢固定位的粘合剂组合物用的烷基乙烯基醚-马来酸共聚物和/或三元共聚物的镁与锌的混合盐,还含有规定量的游离酸,它可以与假牙一同使用。这些组合物可有效地将假牙长期固定在原位上。
发明的概述

发明的详细说明
本发明的假牙粘合剂组合物含有烷基乙烯基醚-马来酸共聚物和/或含异丁烯的三元共聚物(优选共聚物)的镁和锌的混合盐,其中所述的混合盐含规定量的游离酸,并含有选自钙、锶及其混合物的阳离子。该粘合剂组合物的形式可以是粉、乳膏、糊、液体、气雾剂和/或薄片。粉洒到假牙上,湿润,接着放入口腔。该组合物也可以与各种常规的释放载体结合,形成可施加到假牙上并放入口腔的液体或糊。这些组合物能够任选含有至少一种非粘合性的自支撑层。具有自支撑层的假牙粘合剂组合物完全湿润,并施加到假牙上。本发明的基本和任选组分的详细说明如下所述。
定义
在本文中使用的“安全和有效的粘合量”或“有效的粘合量”指足以形成与口腔和/或假牙与口腔的颚和牙脊的粘合性,而对使用者无毒、对口腔组织无损害和不改变假牙材料的量。

聚合物
“三元共聚物”或“含异丁烯的三元共聚物”指马来酸酐或酸、烷基乙烯基醚(优选含C1-C5烷基)和异丁烯的三元共聚物,具有结构(A-B)n,其中A是马来酸酐或酸,B是烷基乙烯基醚(优选含C1-C5烷基)和/或异丁烯。三元共聚物的初始酐或酸的比粘度优选至少约为5.5,优选至少约6,优选在25℃以甲基乙基酮的1%重量/体积溶液测得。三元共聚物必须含有至少部分异丁烯。
本发明的假牙粘合剂组合物含有AVE/MA共聚物和/或三元共聚物的混合盐,其中所述混合盐含有阳离子盐官能,含有下述组分(或基本由下述组成):有效粘合量的镁阳离子、约5-50%锌阳离子、0-约2.5%选自铁、硼、铝、钒、铬、锰、镍、铜、钇、钛及其混合物的阳离子、有效粘合量的选自钙、锶及其混合物的阳离子,以反应的初始羧基总量为基准,所述混合盐含约30-60%游离酸成分。AVE/MA共聚物的比粘度有一个范围。例如,比粘度优选1.2-14,优选以共聚物的初始酐或酸在甲基乙基酮中的1%重量/体积溶液于25℃测得。也能够使用其他的方法和溶剂测量比粘度,例如在DMF(二甲基甲酰胺)中的1%重量/体积溶液中于25℃下测量,和在2-丁酮中的1%重量/体积溶液中于25℃下测量。

烷基乙烯基醚马来酸酐共聚物通过烷基乙烯基醚单体例如甲基乙烯基醚、乙基乙烯基醚、二乙烯基醚、丙基乙烯基醚和异丁基乙烯基醚与马来酸酐共聚形成相应的易于水解成酸共聚物的烷基乙烯基醚-马来酸酐共聚物而得到。合适的共聚物可以由现有技术例如US2782182和US2047398中已知的方法制备,这两篇文献在这里全文引入以供参考。酐和酸两种形式也能够从供应商处买到。例如新泽西州Wayne的ISP Corporation就出售分别为“GANTREZS Series”和“GANTREZ AN Series”的商标为“GANTREZ”的聚合物游离酸形式(I)及其相应的酐形式。当酐共聚物溶解于水中时,酐键分裂,形成高极性的聚合物游离酸(I)。因此,比酸形式便宜的酐形式可以用作酸的便利和便宜的前体。宜采用高温提高酐水解成酸的速率。三元共聚物能够由1999年5月4日授予Prosise等的美国专利№5900470、1991年8月6日授予Tazi等的美国专利№5037924和1992年1月21日授予Tazi等的美国专利№5082913所述的方法制成,其全文在这里引入以供参考。
本发明主题聚合物的盐形式可以由AVE/M酐或酸共聚物或三元共聚物与具有至少一种阳离子盐官能在含水介质中的相互作用形成,所述阳离子盐官能例如具有一般为例如羧酸的反应物的官能团的镁、锌、钙、钠、钾、铁或铵化合物,例如氢氧化物、氧化物、乙酸盐、卤化物、乳酸盐等。在一个优选的实施方式中,采用氧化锌、氧化镁,并结合使用碳酸锶和/或氢氧化钙。含有铁阳离子的聚合物混合盐能够由AVE/M酐/酸聚合物与盐形式的铁化合物例如n水合硫酸铁的相互作用形成。
应当避免可形成毒性、刺激或污染副产物的离子,或提供特殊的预防和处理,以保证从最终聚合物盐的产物中排除或不含这种副产物。使用的具体化合物应当基本是纯的,以保证获得基本纯的聚合物盐最终产物。如果所要求的是聚合物的盐形式,那么就将氧化锌粒子的水分散液与氧化镁(与碳酸锶和/或氢氧化钙组合),和任选n水合硫酸铁组合。它与聚合物粉的浆料形式混合,其用量要足以提供最终产物内所要求的阳离子含量。这在环境温度下进行,接着,在连续剧烈混合下缓慢加热至70-95℃,以避免聚合物阳离子盐在局部沉淀,连续混合,以保证所有形成盐的化合物都与共聚物反应。
或者,将聚合物水解,并在一种或多种二价和/或单价金属碱的水性混合物或浆料中通过加热共聚物/碱混合物至约45-100℃范围进行中和。聚合物混合盐与铁阳离子的反应通过向水解和中和的聚合物混合盐中加入铁盐而实现。与铁阳离子的反应的结束由粘度增高至稳定态来指示。或者,在水解和中和反应之前,可以将铁盐与聚合物/金属碱混合物进行混合。
在上述任一种方法中,将形成的浆料或溶液转移到干燥的不锈钢浅盘中,并放入60-70℃强制通风对流的烘箱中,时间要足以蒸发反应介质(水),并从聚合物中脱除水(约18-24小时)。或者,形成的浆料或溶液也能够用热蒸汽于100-200℃转鼓干燥,以蒸发水含量,并回收薄片状聚合物。干燥后,聚合物形成能够容易地从盘或鼓表面上剥下的脆薄片,根据要求研磨成细粉,以提供满意的假牙稳定性能。这些AVE/MA聚合物混合盐的制备方法进一步在1991年12月17日授予Holeva等的美国专利№5073604、1999年2月16日授予Liang等的5872161和1998年11月3日授予Synodis等的5830933中有描述,其全文在这里引入以供参考。
聚合物混合盐与水或唾液接触时具有特别好的粘合质量,这使它们极其适用于假牙组合物中的假牙粘合剂材料。本发明的组合物含有安全和有效量的聚合物混合盐粘合剂量,优选占组合物的至少20%(重量),更优选至少30%,(重量),作为唯一的粘合剂组分或作为辅助粘合剂与其他粘合剂组分结合使用。
任选的非粘合剂自承性层

非粘合剂自承性层可以是适于为本发明粘合剂组合物提供强度和/或完整性的任何物理形态。这些物理形态包括非织造织物、织造织物、连续的、切断的及其组合。另外,非粘合剂自承性层可以由本行业内已知的任何方法形成。这些方法包括非粘合、喷粘、纺粘、针刺、梳理、热粘合水刺、熔喷、孔印制粘合、针缝、湿法成网、干法成网及其组合。
其他的粘合剂组分
本发明的组合物也可以包含其他的粘合剂组分。这些粘合剂组分如果存在,是以安全和有效的粘合量使用。总的来说,其他的粘合剂组分的含量可以约为0-90%,优选约为10-70%,最优选约为20-50%,以组合物重量为基准。
合适的粘合剂组分包括水溶性亲水性胶体或暴露于水份就溶胀形成粘性物料的聚合物。这些粘合剂材料包括天然胶、合成的聚合物胶、AVE/MA共聚物酸、AVE/MA共聚物酐、AVE/MA/IB、合成的聚合物、粘康粘合剂(mucoadhesive)聚合物、亲水性聚合物、糖衍生物和通常用于假牙稳定组合物并与本发明聚合物相容的粘合剂材料、纤维素衍生物及其混合物。这些材料的例子包括刺梧桐树胶、爪耳胶、白明胶、褐藻胶、藻酸钠、黄蓍胶、脱乙酰壳多糖、聚乙二醇、丙烯酰胺聚合物、丙烯酸聚合物、聚乙烯醇、聚胺、聚季铵化合物、聚丁烯、硅氧烷、聚环氧乙烷、聚乙烯基吡咯烷酮、聚丙烯酰胺阳离子聚合物。
优选纤维素衍生物、聚乙二醇、聚环氧乙烷、刺梧桐树胶、藻酸钠、脱乙酰壳多糖、聚乙烯醇及其混合物。最优选纤维素衍生物,例如甲基纤维素、羧甲基纤维素、羟乙基纤维素、羟丙基纤维素、羟基-丙基甲基纤维素及其混合物。
其他成分
本发明也可以含有还原剂。还原剂有助于在向假牙施加本发明的聚合物混合盐后从口腔取出假牙。本文中使用的优选还原剂是抗坏血酸及其水溶性盐。还原剂也可以与鳌合剂组合使用。优选的鳌合剂包括柠檬酸盐、酒石酸盐、乳酸盐等。还原剂和/或鳌合剂也可以由组合物中的本行业内已知的载体释放,所述载体对口腔施用安全(即用于人无毒并获得批准)。这些载体包括表面活性剂、溶剂等。
还原剂和/或鳌合剂以安全和有效的量使用。本文中使用的“安全和有效的量”术语指足以有助于松开在口腔内固定的假牙而对使用者无毒、对口腔组织无损害和替换的假牙材料的量。由此,假牙戴者可将聚合物混合盐粘合剂组合物施加到假牙或口腔组织上,并将假牙放入口腔内。当要求取下时,戴者在口中沙沙摇动,假牙就释放含有还原剂和/或鳌合剂的组合物和合适的溶剂(一种或多种),它们有助于使假牙活动。

本发明的还含有非粘合剂自承性层的假牙粘合剂组合物,也可以包含粘到干假牙上的涂层,如果它存在,它就放到假牙粘合剂组合物的一面上。适用作该类粘合层的组合物包括聚丁烯、硅氧烷、橡胶、凡士林、天然聚合物、合成聚合物及其混合物。粘合层的含量可以约为0-70%,优选约为0.5-20%,以组合物的重量为基准。
其他的合适组分包括着色剂、防腐剂例如对羟苯甲酸甲酯和对羟苯甲酸丙酯、增稠剂例如二氧化硅和聚乙二醇和载体例如液态凡士林、凡士林、矿物油和甘油。优选聚乙二醇、二氧化硅和凡士林。着色剂、防腐剂、增稠剂和载体的含量可以约为0-20%,以组合物重量为基准。
本发明的组合物还可以包含一种或多种提供香味、香气和/或感知性优点(加热剂或冷却剂)的组分。合适的组分包括天然或人工的甜味剂、薄荷醇、乳酸薄荷酯、冬青油、薄荷油、留兰香油、叶醇、丁子香花油、茴香脑、水杨酸甲酯、桉叶油素、桂皮、乙酸1-薄荷酯、一串红、丁子香酚、欧芹油、噁烷酮(oxanone)、α-紫罗兰酮、甘牛至草、柠檬、橙子、丙烯基乙氧愈创木酚、肉桂、香草醛、麝香草酚、沉香醇、称为CGA的肉桂醛甘油醛缩醇,及其混合物,还有冷却剂。冷却剂能够是多种材料中的任一种。其中包括这样的材料:氨甲酰、薄荷醇、酮缩醇、二醇及其混合物。本发明组合物中优选的冷却剂是对薄荷烷氨甲酰剂(paramenthan carboxyamide agent)例如商品名为“WS-3”的N-乙基-对-薄荷烷(menthan)-3-氨甲酰、称为“WS-23”的N,2,3-三甲基-2-异丙基丁酰胺及其混合物。其他的优选冷却剂选自薄荷醇、由Takasago制备的称为TK-10的3-1-薄荷氧基丙烷-1,2-二醇、由Haarmann和Reimer制备的称为MGA的薄荷酮甘油醛缩醇和由Haarmann和Reimer制备的称为Frescolat的乳酸薄荷酯。本文中使用的薄荷醇和薄荷基术语包括这些化合物的右旋和左旋异构体和其外消旋混合物。TK-10在7/10/84授予Amano等的美国专利№4459425中有描述。WS-3和其他试剂在1979年1月23日授予Watson等的美国专利№4136163中有描述,它们的全文在这里引入以供参考。这些试剂的含量可以约为0-50%,以组合物的重量为基准。
本发明的组合物可以用作假牙粘合剂和/或用作湿组织例如粘膜组织、伤口、口腔粘膜等上的生物粘合剂。本发明的粘合剂组合物能够用来释放一种或多种适于局部施加到粘膜或湿组织上的治疗活性成分。本文使用的“治疗活性成分”术语指透过身体的湿组织或粘膜表面例如口腔、伤口被吸收时或施加到皮肤表面上时具有药理活性的试剂。治疗活性成分的含量可以约为0-70%,以组合物重量为基准。
用于本发明组合物的治疗活性成分包括抗菌剂例如碘、磺酰胺、双缩二胍或酚,抗生素例如四环素、新霉素、卡那霉素、灭滴灵或氯林可霉素,抗炎剂例如阿斯匹林、扑热息痛、消痛灵(甲氧萘丙酸)及其盐、布洛芬、痛主消、氟比洛芬、吲哚类辛、丁子香酚、或氢化可的松;牙用脱敏剂如硝酸钾、氯化锶或氟化钠;麻醉剂如利多卡因、苯佐卡因;抗霉剂;芳香族化合物如樟脑、桉叶油、食用香料、日用香料、或感知剂(加热剂或冷却剂)、和醛衍生物如苯甲醛;胰岛素;类固醇;和抗肿瘤药。认为在某些治疗方式中,为得到最佳疗效,在同一释放体系中组合这些试剂是有用的。因此,例如在单一释放体系中可混合抗菌剂和抗炎剂以得到综合效果。
组合物的制备方法

聚合物的优选溶剂是水、甲醇、丙醇、异丙醇、四氢呋喃、乙酸甲酯、苯甲醛、甲醛溶液、甲基乙基酮、双丙酮醇、N-甲基-2-吡咯烷酮、N-乙烯基-2-吡咯烷酮、二甲基甲酰胺及其混合物。通常用作增塑剂的化合物也能够用作聚合物的溶剂。这些增塑剂包括邻苯二甲酸二甲酯、邻苯二甲酸二乙酯、邻苯二甲酸二辛酯、甘油(glycercin)、二甘醇、三甘醇、IgepalC0-630、GafacRE-610、山梨糖醇、磷酸三甲苯酯、癸二酸二甲酯、羟基乙酸乙酯、羟基乙酸乙基邻苯二甲酰乙酯和对-甲苯乙基磺酰胺。

Claims (10)
1.一种假牙粘合剂组合物,它含有烷基乙烯基醚与马来酸或酐的共聚物的混合盐,所述的混合盐含阳离子盐官能,含有:
(a)5-50%锌阳离子,优选15-40%;
(b)有效粘合量的镁阳离子;
(c)有效粘合量的选自钙、锶及其混合物
的阳离子;
(d)0-2.5%,优选0.01-2%选自铁、硼、铝、钒、铬、锰、镍、铜、钇、钛及其混合物的阳离子;
(e)30-60%游离酸成分;
其中镁、钙和锶阳离子的总量为5-65%,优选15-55%。
2.如权利要求1所述的假牙粘合剂组合物,其中所述的共聚物的比粘度大于5.5。
3.一种假牙粘合剂组合物,它含有烷基乙烯基醚、马来酸或酐与异丁烯的三元共聚物的混合盐,所述的混合盐含阳离子盐官能,含有:
(a)5-50%锌阳离子,优选15-40%;
(b)有效粘合量的镁阳离子;

5.如权利要求4所述的假牙粘合剂组合物,其中所述的其他粘合剂组分选自天然胶、合成聚合物、粘康粘合剂聚合物、亲水性聚合物、天然聚合物、糖衍生物、纤维素衍生物,优选选自羟乙基纤维素、羟丙基纤维素、羟丙基甲基纤维素、羧甲基纤维素钠及其混合物、AVE/MA共聚物的混合盐、AVE/MA共聚物酸、烷基乙烯基醚-马来酸酐共聚物及其混合物。
6.如权利要求4所述的假牙粘合剂组合物,其中所述的载体选自液态凡士林、凡士林、矿物油、甘油及其混合物。
7.如权利要求1或3所述的假牙粘合剂组合物,其中所述的组合物还含有至少一种非粘合剂自承性层。
8.如权利要求7所述的假牙粘合剂组合物,其中所述的非粘合剂自承性层选自聚酯、聚丙烯、尼龙、人造丝、纤维素乙酸酯、非粘合剂纤维素衍生物、布、羊毛纤维、纸、塑料、皮革、微晶蜡、合成纤维、天然纤维及其混合物。
9.如权利要求8所述的假牙粘合剂组合物,其中所述的非粘合剂自承性层的物理形式选自非织造织物、织造织物、连续的、切断的及其组合。
10.一种提高假牙与口腔粘合性的方法,它是将权利要求1或3所述的组合物施加到假牙、口腔或两者上,然后使假牙固定到口腔内。
