Claims (17)
- 权 利 要 求1. 一种具有通式 (Xaa)nTpp(Xaa)m的蛋白质, 其中 Tpp选自梅毒 螺旋体外膜蛋白质 Τρρ15、 Τρρ17、 Τρρ47或其具有结合人免疫球蛋 白之免疫活性的片段或衍生物; Xaa代表天然氨基酸残基,其中与 Tpp 每侧最接近的至少一个 Xaa和至少一个通式蛋白的末端 Xaa是选自 Lys、 Arg和 Trp的氨基酸残基; π和 m分别为 0 10的整数, 条件是 n + m≥2。
- 2. 根据权利要求 1的蛋白质, 其中该蛋白质为 MetLysLys-Tpp- LysLysLeuGluLys, 且 Tpp为梅毒螺旋体外膜蛋白质 Tppl5或其具有 结合人免疫球蛋白之免疫活性的片段或衍生物。
- 3. 根据权利要求 1的蛋白质, 其中该蛋白质为 MetLysLys-Tpp- LysLysLeuGluLys, 且 Tpp为梅毒螺旋体外膜蛋白质 Tppl7或其具有 结合人免疫球蛋白之免疫活性的片段或衍生物。
- 4. 根据权利要求 1的蛋白质, 其中该蛋白质为 MetLysLys-Tpp- LysLysLeuGIuLys, 且 Tpp为梅毒螺旋体外膜蛋白质 Tpp47或其具有 结合人免疫球蛋白之免疫活性的片段或衍生物。
- 5. 权利要求 2的蛋白质, 其具有 SEQ ID No. 1的氨基^^列。
- 6. 权利要求 3的蛋白质, 其具有 SEQ ID No. 2的氨基酸序列。
- 7. 权利要求 4的蛋白质, 其具有 SEQ ID No. 3的氨基酸序列。
- 8. 一种免疫检测梅毒螺旋体抗体的方法, 其中包括利用权利要求 1 - 7中任一项的至少一种蛋白质与生物学样本中梅毒螺旋体抗体间 的抗原 -抗体反应。
- 9. 权利要求 8的方法, 其包括以下步骤:1)利用权利要求 1 - 7中任一项的至少一种纯化蛋白质包被一种 固相支持物;2)将待检测的生物学样本加入经包被的固相支持物上, 在适合 形成抗原抗体免疫结合复合物的条件下孵育一段时间;3)适度清洗去除任何未结合的梅毒螺旋体抗体;4)加入经标记的针对人抗体的第二抗体, 在适合形成抗原抗体 免疫结合复合物的条件下孵育一段时间;5)适度清洗去除任何未结合的第二抗体;6)检测孵育混合物中抗原抗体免疫结合复合物的存在。
- 10. 权利要求 9的方法, 其中针对人梅毒螺旋体抗体的第二抗体是 用选自辣根过氧化物酶、 碱性磷酸酶、 β -半乳糖苷酶和乙酰胆碱酯 酶的酶进行标记的。
- 11. 权利要求 8的方法, 其包括以下步骤:1)利用权利要求 1 - 7任一项的至少一种纯化蛋白质包被一种固 相支持物;2)预先制备经标记的与 1)中所用蛋白质相应的蛋白质;3)将待检测个体的生物学样本及由步骤 2)标记的蛋白质加入经 包被的固相支持物上, 在适合形成抗原抗体免疫结合复合物的条件 下孵育一段时间;4)适度清洗去除任何未结合的梅毒螺旋体抗体;5)检测孵育混合物中抗原抗体免疫复合物的存在.
- 12. 权利要求 11的方法, 其中步骤 2)中的蛋白质是用选自辣根过 氧化物獰、 碱性磷酸酶、 β -半乳糖苷醇和乙酰胆碱酯酶的皡进行标 记的。
- 13. 一种用于免疫检测人梅毒螺旋体抗体的检测试剂盒, 其包括 用纯化的权利要求 1 - 7任一项的至少一种蛋白质包被的检测板。
- 14. 权利要求 13的检测试剂盒, 其还包括与包被检测板种类相对 应的经标记的权利要求 1 - 7任一项的蛋白质。
- 15. 权利要求 13的检测试剂盒, 其中蛋白质是用选自辣根过氧化 物酶、 碱性碑酸酶、 半乳糖苷酶和乙跣胆碱酯酵的酶进行标记的。
- 16. 权利要求 13的检测试剂盒, 其还包括经标记的针对人抗体的 第二抗体。
- 17. 权利要求 13的检测试剂盒, 其中针对人抗体的第二抗体是用 选自辣根过氧化物酶、 碱性磷酸酶、 P -半乳糖苷酶和乙酰胆碱酯酶 的酶进行标记的。
