Abstract

一种用于检测呼吸道感染的卡他摩拉氏菌选择性培养基,选用新鲜牛肉浸液琼脂、葡萄糖、中性红、可溶性淀粉为基础培养基,加入乙酰唑胺抑制奈瑟氏菌生长,加入万古霉素抑制其他杂菌生长。其特点是卡他摩拉氏菌检出率高,原材料来源广,取材方便,制作简单,成本低,适合于国内外大中小型实验室应用。

Description

卡他摩拉氏菌选择性培养基
本发明涉及到检测人类呼吸道感染常见致病菌之一的卡他摩拉氏菌的一种培养基。
当前国内通常采用血液琼脂培养基检测卡他摩拉氏菌。卡他摩拉氏菌与呼吸道正常寄生的奈瑟氏菌特征很相似,在血液琼脂培养基上不易辨认,检出率很低。据国外报道,Soto Hernandez JL利用卡他摩拉氏菌DNA酶阳性的生化特性,用DNA酶试验琼脂、万古霉素、TMP、两性霉素B、甲苯胺兰O配方制成选择性培养基,获得较好的分离卡他摩拉氏菌效果,但是标本接种到该培养基后需要在5%CO2环境下培养48小时以后再加甲苯胺兰O,15分钟内观察菌落颜色,选择出DNA酶阳性的卡他摩拉氏菌,这种培养基检测时间长,方法较繁杂,检出率高。Vaneechoutte--M以组织浸出液琼脂加绵羊血为基础琼脂,加万古霉素,TMP、两性霉素B、乙酰唑胺,这种培养基能抑制大部分奈瑟氏菌和酵母杆菌及其他杂菌生长,但未被抑制的奈瑟氏菌与卡他摩拉氏菌没有明显区别特征,而且组织浸出液制备较繁,国内仅科研单位实验室应用,在全国中小实验室难以推广。
本发明的目的在于研制出一种取材方便,配制简单,成本低,适合国内大中小实验室应用,卡他摩拉氏菌检出率达到国外先进水平的培养基。
本发明通过以下方式实现:卡他摩拉氏菌选择性培养基,利用不同细菌对各种化学物质的敏感性不同的特性,选用多种有利于卡他摩拉氏菌生长的材料,加入能抑制其他细菌生长,特别是抑制奈瑟氏菌生长的材料,经过严格控制配方优化组合而成。选用新鲜牛肉浸液琼脂为基础营养,加入碳酸酐酶抑制剂----乙酰唑胺,抑制大部分奈瑟氏菌生长,加入葡萄糖,中性红指示剂,使未被抑制的奈瑟氏菌菌落呈现红色,以便与卡他摩拉氏菌落相区别,加入可溶性淀粉,降低培养基的毒性,加入万古霉素,抑制其他杂菌生长。
本发明的技术内容是:各种成份的含量分别为:琼脂1.5g~2.5g,新鲜牛肉浸液80ml~100ml,葡萄糖0.8g~1g,中性红0.5ml~0.8ml,可溶性淀粉0.5g~0.7g,乙酰唑胺0.8mg~1mg,万古霉素1mg~1.3mg。
本发明的优点在于:a.新鲜牛肉浸液取材容易,制备简单;b.未被抑制的奈瑟氏菌与卡他摩拉氏菌有明显的区别特征;c.卡他摩拉氏菌生长良好,检出率高。
实施例:首先放入新鲜牛肉浸液100ml,加琼脂2g,再依次加入葡萄糖1g,中性红0.6ml,可溶性淀粉0.5g,经过充分搅拌匀之后组成基础培养基,然后再加入乙酰唑胺0.8mg,万古霉素1.2mg搅拌匀,将PH值调至7.2,这样卡他摩拉氏菌选择性培养基就制成了。

Claims (2)

1.一种主要由琼脂、乙酰唑胺、万古霉素组成的卡他摩拉氏菌选择性培养基,其特征在于:在其中加入了作为基础营养的新鲜牛肉浸液,还加入了使奈瑟氏菌菌落呈红色的葡萄糖和中性红指示剂以及降低培养基毒性的可溶性淀粉,各种成份的含量分别为琼酯1.5g~2.5g,新鲜牛肉浸液80ml~100ml,葡萄糖0.8g~1g,中性红0.5ml~0.8ml,可溶性淀粉0.5g~0.7g,乙酰唑胺0.8mg~1mg,万古霉素1mg~1.3mg。
2.根据权利要求1所述的培养基,其特征在于:各种成份的优化含量分别为琼脂2g,新鲜牛肉浸液100ml,葡萄糖1g,中性红指示剂0.6ml,可溶性淀粉0.5g,乙酰唑胺0.8mg,万古霉素1.2mg。
