Abstract
一种输送极少液量的装置及其制造方法,其载体包含微结构空腔系统。本发明在低成本进行大批量生产的情况下,也能满足对微结构的形状和横截面提出的高的精度要求并保证光学分析测量技术的改进的条件。整体结构的载体的制法是,在板状的载体部分之间设置了一层已处于溶解状态的待粘接层,该层的材料成分与载体部分的材料成分类同,且其层厚比微结构空腔的宽度和深度尺寸小得多。该装置作为分析仪器主要用于医学、生物工程和药物学。
Description
本发明涉及输送极少液量的一种装置及其制造方法,其中载体包含一个微结构空腔系统。
这类空腔可应用在例如生物工程中涉及微量液体的各种场合中,但精确知道该空腔中所包含的微微升含量的液体体积是很重要的。如果形成的空腔呈微通道,则液体可借助动电作用或加压而连续地被输送。密封的空腔用来封闭精确确定的试样容积并阻止少量液体试样的蒸发。在微液体显示器的领域也需要这样的类似结构,在这种显示器中,着色的液体必须通过微通道输送到一个精确确定的位置。
多数已知的技术解决办法强调了主要用于医学、生物工程和药物学的分析仪器的微结构化近年来已取得了重大的进展。
原来在玻璃毛细管中进行的分析现已越来越多地在本身带有分支通道结构的板状微液体结构中进行。虽然这种通道结构最初是象在半导体技术中制造集成电路那样通过蚀刻制作在硅晶片上的,但现在已转向采用塑料。采用塑料的动机不仅是由于塑料的制造成本低廉,而且也由于塑料具有有利的性能,例如光学透明度、生物相容性和一定波长范围内的低的荧光。
以液体引入通道中的并在通道分支中相互反应的物质在一个连续的通道中可利用光学的方法进行分析。由于为此目的确定的通道横截面尺寸迄今为止为10微米至100微米的范围,所以对这类产品的制造提出了很高的要求。
例如根据美国专利US 5 376 252提出的一种微液体构件,在两层平板状稳定的底层之间放置一层弹性的中间层,该中间层包含一个通过模塑成型制成的微结构通道系统。
这种结构的缺点是,弹性中间层的变形会影响整个构件。计量和泄漏问题会对适用性产生不利影响。
美国专利US 5 770 029描述了用于电泳目的的组合装置形式的另一种已知的解决办法,这个主要由两部分组成的装置在一块底板内设有微通道形式的微结构,微通道用一块盖板层密封。这些微通道包含一个富集通道和一个电泳主流通道,这些通道是这样布置的:废物不可能进入主流通道中,而是通过一个单独的出口排出该装置。作为一次性使用的经济方案,建议全部组成部分都用塑料制成。在这种情况下,当这些通道的宽度和深度尺寸要求为10微米和小于此尺寸时,特别是这些尺寸的波动范围小于5%时,可用该专利所述的制造技术改变这些通道的形状和横截面。
在微液体显示器的应用范围内,对图像显示装置的制造技术也提出了很高的要求,在这种显示装置中与图像内容一致的至少两种对比度不同的液体通过微型泵在一个曲折形状的微通道结构中进行泵送。在关掉泵后,形成一种包含液体段的稳定的图案,该图案可再现显示内容。与液晶显示比较,对使用者来说,一个决定性的优点是,用很低的电能消耗提供了清晰的字母数字显示,因为这种显示是在无电流的静态中进行的。如果需要改变图像内容,则用新的图像内容置换旧的图像内容,并将旧的图像内容移入一个分离器中,在该分离器的输出端将分离的液体重新供给相应的微型泵。
这种原理要求使用光学透明的聚合物材料,微通道的尺寸小于10微米。为了达到用两种液体正确表示符号,对这些微通道的制作精度和重复性提出了特殊的要求。特别是,为了精确地确定液体段的位置,对重复性和通道横截面的恒定性要求很高,以及为了微通道中的压力降保持尽可能小,要求有极高的表面光洁度。
本发明的目的在于,在低成本进行大批量生产的情况下也能满足对微结构的形状和横截面提出的高的精度要求。此外,还可保证光学分析技术的改进的条件。
根据本发明,本发明目的是通过在一种载体中包含用于输送极少量液体的微结构空腔系统的装置来实现的,该载体除了具有微结构空腔外,基本上是一种整体式结构。
该载体用热塑性材料制成,所以它与试样物质不发生相互作用。
载体的相当于一个整体的单一结构是这样形成的:即该载体是由板状的载体部分与一层待粘接层制成的,该层材料成份在溶剂中与板状载体部分的成分类同。其中,特别重要的是,该层的厚度比微结构空腔的宽度和深度尺寸小得多。因此,通过例如在真空条件下经热模冲压制成的微结构空腔具有精度高的参数。可避免出现边缘变圆、材料增厚、弯曲和类似变化等现象,因而能对装入空腔中的液体的流动特性产生正面的影响。
由于该粘接层是以液态导入两板状载体部分之间的,所以在溶剂挥发以后,该层便与两个板状部分的表面形成反应性粘接。即使用电子显微镜在连接面上也看不出有明显的边缘结构,所以这样制成的载体除微结构空腔外呈一个整体结构。因为载体不含杂质,材料性能不存在突变,这对封闭的试样的测量精度产生正面的影响。避免了由一些物理参数例如折射指数、吸收系数和导热系数的空间变化所引起的影响。
此外,本发明还涉及一种制造输送极少液量的装置的方法,该装置在一种载体中含有一个微结构空腔系统。为了形成载体的一个相当于一个整体的结构,在板状载体部分之间放一层处于溶解状态的待粘接的层,该层的材料成分与板状载体部分的材料成分类同,且其层厚比微结构空腔的宽度和深度尺寸小得多。
下面结合示意附图来详细说明本发明。
附图表示在制作一种试样分析装置时箭头方向下的三个步骤。图示只表示原理,所以不符合实际的尺寸比例。
根据附图,制作本发明装置从第一步骤开始,首先将第一聚合物即板状载体部分1用模具2例如在真空条件下通过热模冲压形成微结构3。载体部分1在这里用聚甲基丙烯酸甲酯(PMMA)制成。但也可用别的材料,特别是热塑性塑料例如聚碳酸酯(PC)或聚乙烯(PE)来制备。
首先用一种很硬的材料一般为金属或硅用显微技术的方法制作所要求的微结构3的结构负片图案。制作金属模具例如可用众所周知的称为LIGA技术的一种方法步骤(详见LIGA方法,微电子(LIGA Process,Microelectron),Eng.4(1986)35-56页)。抗蚀层用X射线光刻方法通过一个掩模用同步加速器辐射进行曝光。为此,例如可用德国专利DE 44 18 779 C1和DE 44 24 274 C1所述的装置。抗蚀材料经显影后产生的形状用上述材料以电镀方式进行填充,所以去掉抗蚀层后,就显示出微结构3的结构负片图案作为模具2。为了用硅制作结构负片图案,例如可用湿化学蚀刻硅的众所周知的方法或用反应性离子蚀刻进行表面处理的方法。
在采用微系统结构的成形装置的情况下,例如按德国专利DE 19648 844 C1将模具2与聚合物载体部分1一起加热到超过聚合物材料的玻璃化转变温度的温度。将模具2中所显示的结构形态转移到载体部分1上的步骤最好在真空条件下进行,即将这两部分在高压作用下相互挤压。由于载体部分1和模具2仍处在紧密接触的状态下冷却到低于聚合物材料的玻璃化转变温度的温度,所以载体部分1的结构成为固定的。这样,在脱模后,微结构3就保持下来,当然,也可采用别的复制技术例如浇注、紫外线反应浇注、注塑或别的冲压方法。
用与载体部分1相同的聚合物材料制成的第二载体部分4作为微结构3的盖板用。这样就形成了微结构空腔5。这些空腔5的至少一部分通过通道6与外界连通,这样就可保证腔内注满和排空。如图所示,通道6可形成在载体部分4中或用其它方式例如可侧向引到外界。
为了连接载体部分1和4,按步骤2在这两个载体部分之间引入一层7,这层7在一种溶剂例如乙酰乙酸甲酯中只含有与两个载体部分1和4的相同材料。在一定条件下,也可用一种其分子结构与两个载体部分1和4的分子结构十分相似的聚合物。该层7的厚度特别重要,因为它对微结构空腔5的功能起决定性的影响。所以,必须避免该层材料进入微结构3中,特别是避免它的边缘区被溶剂腐蚀。在通道横截面为10×10微米至40×40微米的情况下,层厚必须小于0.6微米。此外,在两载体部分1和4连接时必须避免毛细管效应的负面影响。最后,层7在整个待连接的表面上具有均匀的分布也是很重要的。如果该层材料在通道6的区域聚积的话,则该材料通常有可能进入微结构3中。所以,在待涂敷的载体部分4中的通道6最好在涂敷期间用适当加工的触针封闭,从而形成充分封闭的表面。经过也包括载体部分1在内的清洁处理以后,进行层7的涂敷。应通过清洁处理将附着在表面上的会破坏该层7的均匀性和妨碍参与工件的完好连接的微粒,充分地去除掉。
首先例如用吸管吸取少量的大约几微升已溶解的聚合物材料置于载体部分4的表面中心,然后通过固定在一个转台上的载体部分4的快速旋转调节需要的层厚。确定厚度的主要因素是已溶解的聚合物材料的粘度和旋转速度。例如如果在PMMA载体中的微结构空腔5具有10微米范围的宽度和高度在10微米范围,则在PMMA与溶剂乙酰乙酸甲酯的比例为1∶7~1∶11的情况下,所需的转数为每分钟几千转(4000~6000转/分钟)。在这些条件下达到的层7的厚度可达到上述范围并比微结构空腔5的宽度和高度尺寸小得多。在层7涂敷后,立即将两载体部分1和4通过机械接触进行相互粘接,微结构3便通过载体部分4的粘接而成封闭状态。其中,应避免倾斜误差和横向偏移并保证均匀的压力条件。这项工作例如可在一种适当的装置内进行,即把两载体部分1和4精确配合和相互间没有横向滑动并用一个模头施加均匀的力。也可通过辊压工序实现粘接。为了避免溶剂的过早挥发,在20-60秒内进行快速粘接对达到永久的和均匀的粘接来说,是特别重要的。否则,势必或多或少地会损害两个工件的确定的和长期稳定的连接。然后,溶剂在室温下慢慢蒸发。当然,可通过提高温度或通过使用带有或不带真空设备的干燥炉来缩短加工时间。
在第三步骤中形成一个整体8的最终结构,即在溶剂蒸发时由于溶剂在表面内形成的断开部分实施重新连接,其中每个表面上边界层区域内的分子与层7的分子相结合。
在溶剂挥发后,就形成了一个单一的载体,这个单一的载体只由一种材料所组成并且在原先边界层上也不存在材料性能不连续性的现象。
连接层的涂敷不限于上述的方法,虽然这种方法达到了特别好的结果。特别是作为另一选择辊涂也是一种很适用的方法。
Claims (6)
1.一种在载体内包含一个微结构空腔系统、用来输送极少液量的装置,其特征为,该载体除了微结构空腔(5)外,基本上是一个整体结构。
2.按权利要求1的装置,其特征为,该载体由一种热塑性材料构成。
3.按权利要求2的装置,其特征为,该载体用板状载体部分(1、4)与一待粘接的层(7)制成,在一种溶剂中,该层的材料成分和板状载体部分(1、4)的材料成分类同,且其层厚比微结构空腔(5)的宽度和深度尺寸小得多。
4.按权利要求3的装置,其特征为,微结构空腔(5)通过在真空条件下经热模冲压至少在一个板状载体部分(1)内而形成。
5.按权利要求4的装置,其特征为,微结构空腔(5)呈通道形,这些部分通道从该载体中引出。
6.一种在载体中包含一个微结构空腔系统、用来输送极少液量的装置的制造方法,其特征为,为了形成载体的相应的整体结构,在板状载体部分(1、4)之间设置了一层已处于溶解状态的待粘接的层(7),该层的材料成分与板状载体部分(1、4)的材料成分类同,且其层厚比微结构空腔(5)的宽度和深度尺寸小得多。
