本发明公开了一种靶向视网膜神经节细胞并过表达神经生长因子的腺相关病毒载体及应用,属于生物医药技术领域。本发明提供了一种靶向视网膜神经节细胞过表达神经生长因子的腺相关病毒载体,所述腺相关病毒载体以神经元特异性启动子启动神经生长因子的过表达。本发明还基于所述腺相关病毒载体制备了治疗青光眼的药物,通过将携带神经生长因子基因序列的腺相关病毒载体直接进行玻璃体腔注射,可使视网膜神经节细胞长期表达神经生长因子,从而起到神经保护的作用。

技术领域
本发明属于生物医药技术领域,具体涉及一种靶向视网膜神经节细胞并过表达神经生长因子的腺相关病毒载体及应用。
背景技术
青光眼是一种常见的眼部神经退行性疾病,以视网膜神经节细胞(RGCs)死亡和轴突变性为特征。主要的治疗策略是通过药物和手术等方法来减少高眼压对视网膜神经的损伤。然而,降低眼压并不总能预防RGCs的进展性死亡和视力损害。由于失去的RGCs难以再生,保护RGCs免于青光眼导致的死亡仍然是首要关注的问题。
神经生长因子(Nerve Growth Factor,NGF)是临床治疗神经损伤常用的神经营养因子,广泛应用于角膜神经修复、视神经损伤修复等领域。研究表明,神经营养因子的缺乏可导致轴突运输受阻,最终导致细胞凋亡,这也是青光眼的重要发病机制。然而,神经营养因子的双重作用影响了其对青光眼RGCs的保护作用。研究表明,包括NGF在内的神经营养因子可与RGCs上的原肌球蛋白相关激酶(Trk)受体结合介导神经元存活,也可通过与Müller细胞上的p75NT受体(p75NTR)结合间接介导RGCs的凋亡,从而导致治疗效果欠佳。
发明内容
本发明的目的在于提供一种靶向视网膜神经节细胞并过表达神经生长因子的腺相关病毒载体及应用,所述腺相关病毒载体可靶向视网膜神经节细胞,使神经生长因子的表达集中于视网膜神经节细胞,对青光眼具有很好的神经保护作用。
本发明提供了一种靶向视网膜神经节细胞过表达神经生长因子的腺相关病毒载体,所述腺相关病毒载体以神经元特异性启动子启动神经生长因子的过表达。
优选的,所述腺相关病毒载体的载体骨架包括AAV。
优选的,所述AAV包括AAV2。
优选的,所述神经元特异性启动子包括Syn启动子。
优选的,所述过表达的神经生长因子的基因ID:18049。
本发明还提供了上述腺相关病毒载体在制备治疗青光眼的药物中的应用。
本发明还提供了上述腺相关病毒载体在制备对青光眼患者的视网膜神经节细胞有神经保护作用的药物中的应用。
优选的,所述药物的剂型包括注射剂。
本发明还提供了一种治疗青光眼的药物,以上述腺相关病毒载体为有效成分,还包括药学上可接受的辅料。

有益效果:本发明提供了一种靶向视网膜神经节细胞过表达神经生长因子的腺相关病毒载体,所述腺相关病毒载体以神经元特异性启动子启动神经生长因子的过表达。在本发明实施例中,使用可特异性结合RGCs的神经元特异性启动子hSyn,使得神经生长因子直接作用于RGCs,尽可能减少p75NTR介导的RGCs凋亡。通过AAV载体靶向转染RGCs,可使RGCs长期过表达NGF,从而对青光眼患者的视网膜起到神经保护作用。
本发明还基于所述腺相关病毒载体制备了治疗青光眼的药物,通过将腺相关病毒载体直接进行玻璃体腔注射,可使视网膜神经节细胞长期表达神经生长因子,从而起到神经保护的作用。

具体实施方式
本发明提供了一种靶向视网膜神经节细胞过表达神经生长因子的腺相关病毒载体,所述腺相关病毒载体以神经元特异性启动子启动神经生长因子的过表达。
本发明所述腺相关病毒载体的载体骨架优选包括AAV,所述AAV优选包括AAV2,所述AAV2对视网膜的转染效率高,效果好,如在实施例中向小鼠玻璃体腔内注射该载体,可见对视网膜神经节细胞具有较高的转染效率(图1)。本发明所述神经元特异性启动子优选包括Syn启动子,实施例中以来源于人的hSyn启动子为例进行说明,但是不能仅将其认定为本发明得全部保护范围,所述Syn启动子为本领域所熟知的神经元特异性启动子。本发明所述过表达的神经生长因子的基因ID:18049。
在本发明中,Syn启动子是一种神经元特异性启动子,相较于CMV等广谱启动子,Syn启动子可使腺相关病毒在玻璃体腔注射后靶向视网膜神经节细胞,从而使所携带的神经生长因子在视网膜神经节细胞内特异性表达,从而在青光眼中起到神经保护作用。
本发明实施例中,以AAV2为载体骨架,构建得到一个腺相关病毒载体AAV2-hSyn-NGF,在所述AAV2-hSyn-NGF中优选还可以添加flag或荧光标签,如mNeonGreen、WPRE等。
本发明还提供了上述腺相关病毒载体在制备治疗青光眼的药物中的应用。
本发明实施例通过视神经钳夹模型(ONC),证实眼内注射AAV-hSyn-NGF载体后可明显保护视网膜神经节细胞,其数量较单纯视神经钳夹模型高;眼内注射AAV-hSyn-NGF载体组神经节细胞数量较单纯微球模型(MB)多,起到保护作用;利用模拟人遗传性青光眼的小鼠模型DBA2/J小鼠,证实注射AAV-hSyn-NGF载体组视网膜神经节细胞更多,说明起到了保护作用。
本发明还提供了上述腺相关病毒载体在制备对青光眼患者的视网膜有神经保护作用的药物中的应用。
本发明所述药物的剂型优选包括注射剂,用于玻璃体腔注射。
本发明还提供了一种治疗青光眼的药物,以上述腺相关病毒载体为有效成分,还包括药学上可接受的辅料。
本发明所述药物用量以所述腺相关病毒载体计,为1×108-1×1011载体颗粒/眼。
为了进一步说明本发明,下面结合实施例对本发明提供的一种靶向视网膜神经节细胞并过表达神经生长因子的腺相关病毒载体及应用进行详细地描述,但不能将它们理解为对本发明保护范围的限定。
实施例1
1、AAV2-Syn-NGF的构建
委托和元生物技术(上海)股份有限公司合成载体结构:AAV2-hSyn-NGF的腺相关病毒载体。
2、AAV2-hSyn-NGF注射液的制备
以PBS为溶剂,制备后注射液滴度为1×108-1×1011v.g./μL。
3、视神经钳夹模型(ONC)中的应用
方法:从角膜缘后1mm向小鼠眼内注射AAV-hSyn-NGF载体,每组3只,随机分组。3周后,使用0.6%的戊巴比妥钠麻醉小鼠后,打开结膜囊,分离眶内组织后暴露视神经,使用反向显微自闭颞在眼球后1~2mm处钳夹视神经10秒,构建视神经钳夹模型。
结果如图2所示,与单纯视神经钳夹模型相比,眼内注射AAV-hSyn-NGF载体后可明显保护视网膜神经节细胞,其数量较单纯视神经钳夹模型高。
4、微球模型(MB)中的应用
方法:从角膜缘后1mm向小鼠小鼠眼内注射AAV-hSyn-NGF载体,随机分组,每组3只。3周后,向前房注入2μL 2.5%的聚苯乙烯微球建造青光眼模型。
结果如图3所示,眼内注射AAV-hSyn-NGF载体组神经节细胞数量较单纯MB模型多,起到保护作用。
5、DBA/2J小鼠模型
方法:DBA2/J小鼠是一种模拟人遗传性青光眼的小鼠(北京维通利华实验动物技术有限公司,分为AAV-Syn-NGF载体注射组和非注射组,每组3只)。在5.5月龄时向小鼠眼内注射AAV-hSyn-NGF载体,12月龄时进行观察。
结果如图4所示,注射AAV-hSyn-NGF载体组视网膜神经节细胞更多,说明起到了保护作用。
尽管上述实施例对本发明做出了详尽的描述,但它仅仅是本发明一部分实施例,而不是全部实施例,人们还可以根据本实施例在不经创造性前提下获得其他实施例,这些实施例都属于本发明保护范围。

1.一种靶向视网膜神经节细胞过表达神经生长因子的腺相关病毒载体,其特征在于,所述腺相关病毒载体以神经元特异性启动子启动神经生长因子的过表达。
2.根据权利要求1所述腺相关病毒载体,其特征在于,所述腺相关病毒载体的载体骨架包括AAV。
3.根据权利要求2所述腺相关病毒载体,其特征在于,所述AAV包括AAV2。
4.根据权利要求1所述腺相关病毒载体,其特征在于,所述神经元特异性启动子包括Syn启动子。
5.根据权利要求1所述腺相关病毒载体,其特征在于,所述过表达的神经生长因子的基因ID:18049。
6.权利要求1~5任一项所述腺相关病毒载体在制备治疗青光眼的药物中的应用。
7.权利要求1~5任一项所述腺相关病毒载体在制备对青光眼患者的视网膜神经节细胞有神经保护作用的药物中的应用。
8.根据权利要求6或7所述应用,其特征在于,所述药物的剂型包括注射剂。
9.一种治疗青光眼的药物,其特征在于,以权利要求1~5任一项所述腺相关病毒载体为有效成分,还包括药学上可接受的辅料。
10.根据权利要求9所述药物,其特征在于,所述药物用量以所述腺相关病毒载体计,为1×108-1×1011载体颗粒/眼。
