Abstract

Description
本发明涉及呈粉末形式的食用油——其优选包含多不饱和脂肪酸(PUFA)、更具体地长链多不饱和脂肪酸(LC-PUFA),其制备以及在食物产品、更特别地在婴儿配方奶粉中的用途。
含有通常呈甘油酯形式的不饱和脂肪酸并且尤其是多不饱和脂肪酸(PUFA)的食用油已示出具有有益的健康效果。这些健康效果包括降低胆固醇水平、预防冠心病以及抑制血小板聚集。例如,含有ω-3和ω-6脂肪酸二十二碳六烯酸(DHA)和二十碳五烯酸(EPA)的鱼油因其健康益处已被用于食物产品和营养产品中。
此外已知的是,长链多不饱和脂肪酸(LC-PUFA)、花生四烯酸(ARA)、二十二碳六烯酸(DHA)改善婴幼儿的大脑和眼睛发育。因此大多数婴儿配方奶粉和较大婴儿配方奶粉用这些LC-PUFA强化。
由于其不饱和键,PUFA具有发生氧化的倾向,尤其是在储存期间。氧化反应产物具有令人不愉快的味道和/或气味;异味,可能是金属味、鱼腥味或腐臭味。因此,这些油的保质期和储存稳定性是相对短的。
已知的是通过将此类油掺入基质中来改善此类油的保质期。基质使油稳定防止氧化。
一种此类稳定方法是包封在含有乳化剂和碳水化合物源的基质中。
乳化剂的实例是蛋白质,如酪蛋白酸盐、大豆蛋白和乳清蛋白。
碳水化合物源的实例是糖醇如甘露醇、葡萄糖浆、乳糖和麦芽糊精,其中葡萄糖浆是所使用的最常规的碳水化合物源。甘露醇的缺点是它具有轻泻影响并且对味道产生负面影响。另一方面,麦芽糊精在加工期间对粘度产生不希望的影响。乳糖容易结晶,因而在包封物生产期间将油从形成的液滴中排出。并且包括葡萄糖在内的这些物质都不存在于人类母乳中或除此以外不是婴儿配方奶粉中所希望的。因此,这些碳水化合物源优选不被引入婴儿配方奶粉中。
此外,目前已知的经包封的食用油的储存稳定性仍然有限并且持续希望进一步改善经包封的食用油的储存稳定性/保质期。这种氧化程度以及因此储存稳定性可以通过异味和/或气味来检测。另外地或可替代地,其可以通过测量过氧化值和/或茴香胺值来检测。
因此,本发明的目的是提供一种经包封的食用油、优选经包封的PUFA、更优选经包封的LC-PUFA,其包含有利地添加到婴儿配方奶粉中并且提供改善的储存稳定性以及因此更好的感官特性的碳水化合物源。
现已发现,使用低聚半乳糖(GOS)和/或人乳低聚糖(HMO)作为碳水化合物源解决了这些问题。
GOS是不可消化的低聚糖,由于其与人类母乳中存在的低聚糖极为相似以及其经证实的在(生命早期)肠道微生物群中的以下益处而有利地添加到婴儿配方奶粉中:促进有益细菌、特别是双歧杆菌和乳酸杆菌的生长,抑制病原体粘附,以及改善肠道屏障功能。
此外,已经发现,与常规的基于葡萄糖的包封物或基于乳糖的包封物相比,含有GOS和/或HMO的包封物具有更高的储存稳定性。
人乳含有多种不可消化的低聚糖。这些低聚糖是人乳提供的先天免疫系统中的主要要素。它们通过促进有益微生物群(如双歧杆菌和乳酸杆菌)的生长在健康肠道微生物群系的发展中发挥着重要作用,并且在防止病原体和毒素的粘附中发挥着重要作用。
人乳中存在的低聚糖与家畜(如奶牛)的乳中存在的低聚糖不同。人乳中存在超过100种不同的低聚糖,其中存在最丰富的低聚糖是岩藻糖基化乳糖如2’-岩藻糖基乳糖(2’-FL)和3-岩藻糖基乳糖(3-FL)、唾液酸化乳糖如3’-唾液乳糖(3’-SL)和6’-唾液乳糖(6’-SL)、以及四糖像乳糖-N-四糖(LNT)和乳糖-N-新四糖(LNnT)。存在最丰富的人乳低聚糖(HMO)是2’-FL。
因此,本发明涉及一种粉末组合物,其包含基于粉末的总重量:
-10-80wt%的氧化敏感的食用油,
-5-30wt%的至少一种选自蛋白质和淀粉的乳化剂,以及
-5-65wt%的低聚半乳糖(GOS)或人乳低聚糖(HMO)。
应当注意,GOS在海藻油DHA包封物中的用途已经描述于CN 108576820中。除了油和GOS之外,这些包封物还含有蛋黄卵磷脂作为乳化剂。然而,如在本发明中使用的蛋白质和淀粉是比蛋黄卵磷脂更好的乳化剂,并且因此本发明提供了改善的产品。
本发明的粉末组合物优选地包含基于粉末组合物的总重量约10wt%至约80wt%、更优选约15wt%至约60wt%、甚至更优选约20wt%至约55wt%的量的氧化敏感的食用油。
食用油优选地包含至少10wt%、更优选至少25wt%、并且最优选至少28wt%的一种或多种含有至少18个、更优选18-30个、最优选18-24个碳原子和至少2个、更优选2-6个碳-碳双键的羧酸或其酯。
这些羧酸可以呈游离酸、酯、或游离酸(包括其盐,如钠盐)和酯的混合物的形式。典型地,该一种或多种羧酸将呈如存在于或衍生自天然来源的复杂混合物的形式。合适的羧酸的实例是二十二碳六烯酸(DHA)、二十碳五烯酸(EPA)、共轭亚油酸(CLA)(包括顺式-9、反式-11和反式-10、顺式-12异构体及其混合物)、十八碳四烯酸、亚油酸、α-亚麻酸(ALA)、γ-亚麻酸(GLA)、花生四烯酸(ARA)及其混合物。优选地,该一种或多种羧酸选自DHA、ARA、EPA及其混合物。在DHA和EPA的混合物中,组分可以以任何比率存在,但对于某些应用,0.4至8.0的DHA/EPA重量比可能是优选的。
所述羧酸的合适的酯包括与含有一至六个碳原子的脂族醇的酯,如乙酯。其他合适的酯包括与食物产品中可接受的醇和多元醇的酯。其他酯的实例是单甘油酯、二甘油酯和三甘油酯及其混合物。三甘油酯是特别优选的,典型地作为与单甘油酯和/或二甘油酯一起的主要组分(即,大于按重量计50%),例如最高达35wt%的二甘油酯和最高达5wt%的单甘油酯。
食用油的其他组分可以包括例如具有12至30个碳原子的饱和羧酸、具有12至30个碳原子的单不饱和羧酸及其混合物,典型地作为它们的酯、如甘油酯(例如三甘油酯)。
术语食用油涵盖无毒并且可以作为正常饮食的一部分食用的油。食用油典型地在25℃和大气压下是液体并且优选地在0℃-25℃的温度范围内和大气压下是液体。油通常是疏水性的,意指其在25℃下与水以1:1的重量比基本上不混溶。油优选地是从天然来源获得的或可获得的,如植物油、动物油(包括鱼油)或动物脂肪、或微生物油,但也可以是合成的。油可以是来自不同来源的油的混合物或是合成油与一种或多种来自天然来源的油的混合物。
特别优选的是,食用油是鱼油。鱼油可以直接或间接地从鱼中获得并且可以是例如鱼油浓缩物、分馏鱼油或改性鱼油。鱼油包括来自鲭鱼、鳟鱼、青鱼(herring)、金枪鱼、鲑鱼、鳕鱼、鲱鱼、鲣鱼以及沙丁鱼的油。鱼油典型地含有ω-3和ω-6PUFA(作为其三甘油酯)的混合物以及其他组分。在特别优选的实施例中,油是包含至少30wt%的DHA的鱼油浓缩物。可替代地,还可以使用从含有大量LCPUFA的真菌和海藻中可获得的油。
在30℃下与空气接触储存12周至16周之后,食用油优选具有小于10meq/kg、优选小于9meq/kg、更优选小于8meq/kg、甚至更优选小于7meq/kg、如小于6或小于5meq/kg的过氧化值。
根据本发明的粉末组合物包含至少一种选自蛋白质和淀粉的乳化剂。还可以使用这些乳化剂的组合。此乳化剂以5-30wt%、优选7-25wt%、并且最优选10-20wt%的量存在于粉末组合物中。
合适的蛋白质的实例是乳蛋白像酪蛋白酸盐和乳清蛋白,以及植物蛋白如大豆、豌豆和马铃薯蛋白。
淀粉的实例是乳化改性淀粉,如通过与正辛烯基琥珀酸酐(NOSA)反应而改性的淀粉。
此外,除蛋白质或淀粉之外,粉末组合物可以进一步包含消泡剂或乳化剂;如单甘油酯、二甘油酯。
粉末组合物包含5-75wt%、优选10-65wt%、并且最优选20-60wt%的低聚半乳糖(GOS)和/或人乳低聚糖(HMO)。
低聚半乳糖(GOS)是具有不同链长、键类型和支化度的碳水化合物的复杂混合物。GOS优选地包含基于干重40-100wt%、更优选50-90wt%、并且最优选55-80wt%的低聚糖(DP≥3)。GOS可以进一步含有单糖像葡萄糖和半乳糖,以及二糖如乳糖、乳果糖和异乳糖。基于干重,乳糖含量通常在0-60wt%、优选0-40wt%、并且最优选0-30wt%的范围内;单糖含量通常在0-20wt%的范围内。此外,GOS可以含有0-5wt%的量的蛋白质。
GOS的合成典型地涉及许多通过β-半乳糖苷酶(β-D-半乳糖水解酶;EC 3.2.1.23)催化的半乳糖基转移过程,其使用乳糖作为半乳糖基供体并且使用乳糖或中间体GOS物质作为半乳糖基受体。此类β-半乳糖苷酶的实例是环状芽孢杆菌(Bacillus circulans)、米曲霉(Aspergillus oryzae)、黑曲霉(Aspergillus niger)、马克斯克鲁维酵母(Kluyveromyces marxianus)、脆壁克鲁维酵母(Kluyveromyces fragilis)、独特掷孢酵母(Sporobolomyces singularis)、发酵乳杆菌(Lactobacillus fermentum)和土生蝶孢银耳(Papiliotrema terrestris)。混合物的组成尤其取决于所使用的酶。例如,衍生自曲霉属(Aspergillus)的真菌β-半乳糖苷酶主要产生β1-6键(从而产生主要包含β1-6键的GOS制剂,其可以称为“6’-GOS”),而衍生自芽孢杆菌属(Bacillus)的细菌β-半乳糖苷酶主要产生β1-4键(产生主要包含β1-4键的GOS制剂,其也可以称为“4’-GOS”)。在本发明中,可以使用任何类型的GOS混合物以及因此从任何类型的酶获得的GOS。
除了GOS之外,还可以添加来自单独来源、即除GOS之外的来源的乳糖。出于经济原因,本发明的粉末组合物中的乳糖与总的二糖加低聚糖含量(因此,包括乳糖)的重量比优选地尽可能地高。然而,过高的乳糖含量增加乳糖结晶的风险,因而在包封物生产期间将油从乳液的液滴中排出。
人乳低聚糖优选地选自由以下组成的组:岩藻糖基化乳糖、唾液酸化乳糖、四糖及其组合,优选地选自由以下组成的组:2’-岩藻糖基乳糖(2’-FL)、3-岩藻糖基乳糖(3-FL)、3’-唾液乳糖(3’-SL)、6’-唾液乳糖(6’-SL)、乳糖-N-四糖(LNT)、乳糖-N-新四糖(LNnT)及其组合,更优选地为2’-岩藻糖基乳糖(2’-FL)或2’-FL与一种或多种额外的HMO的组合,最优选地为2’-FL。
根据本发明的粉末组合物可以包含5-65wt%的一种单一HMO、5-65wt%的一种或多种HMO的组合、或5-65wt%的GOS与一种或多种HMO的组合。
除了HMO之外,还可以添加乳糖。出于经济原因,本发明的粉末组合物中的乳糖与HMO的重量比优选地尽可能地高。然而,过高的乳糖含量增加乳糖结晶的风险,因而在包封物生产期间将油从乳液的液滴中排出。
此外,根据本发明的粉末组合物可以包含一种或多种抗氧化剂。在一个实施例中,组合物可以包含至少一种在油相周围的基质中的抗氧化剂;此类抗氧化剂的优选实例是抗坏血酸盐,如抗坏血酸钠。可替代地或另外地,根据本发明的组合物可以包含至少一种在油相中的抗氧化剂,如柠檬酸、迷迭香、生育酚、抗坏血酸棕榈酸酯及其混合物。
根据本发明的组合物可以进一步包含稳定剂(如磷酸二钾)和/或螯合剂和/或缓冲剂如柠檬酸盐和/或正磷酸盐。如果需要的话,组合物还可以包含自由流动助剂,如磷酸三钙。
在优选的实施例中,根据本发明的粉末组合物是自由流动的粉末。术语“自由流动的粉末”是指可以倾倒(例如,从一个具有约10cm2至50cm2的开口的容器至另一个具有相似尺寸的容器)而颗粒基本上不结块的微粒材料。详细地,术语“自由流动的”用于没有粘性并且因此没有或几乎没有任何附聚或粘附至接触表面倾向的粉末状材料。所谓的休止角θr有时被用作粉末流动特性的量度。休止角是当粉末倾倒在平坦表面上时,粉末锥体与该表面之间形成的角度。典型地,对于自由流动的粉末,θr小,例如小于60°或小于45°,如40°或更小。
自由流动的粉末有时被称为“干燥粉末”,但是在此上下文中词语“干燥”不一定暗指组合物中不存在水。
典型地,粉末具有约10μm至约1000μm、优选约50μm至约800μm、更优选约90至400μm、并且最优选100μm至约300μm的平均粒度。粒度可以使用技术人员已知的技术来确定,例如通过使用众所周知的库尔特计数器(Coulter Counter)。例如,超过95wt%的颗粒可以具有小于800μm的尺寸和/或超过85wt%的颗粒可以具有小于500μm的尺寸和/或超过90wt%的颗粒可以具有大于20μm的尺寸和/或65wt%的颗粒可以具有大于200μm的尺寸。
本发明组合物的粉末形式优选地具有200至600g/l、如300至500g/l的堆积密度。
根据本发明的粉末组合物可以通过制备成分的水性乳液,然后将该乳液喷雾干燥来制备。在特别优选的实施例中,使用喷雾干燥设备进行喷雾干燥。此类喷雾干燥器在顺流塔中使用喷嘴雾化,该塔将半干燥的颗粒向下引导到传动带上。然后,将材料作为多孔层平缓地输送到后干燥和冷却区域,形成脆饼状结构(其在带的末端破裂)。
在优选的实施例中,通过将油添加到所有其他成分的水溶液中,然后进行均质化,将油和所有其他成分乳化至通常50至70wt%的干物质含量。

干燥优选地通过喷雾干燥进行。喷雾干燥的条件是本领域技术人员已知的或者可以由本领域技术人员容易地确定。喷雾干燥优选在使得所得粉末具有20μm至800μm的平均粒度的条件下进行。当使用本领域技术人员已知的雾化喷嘴系统,通过在50至200巴的压力下操作的高压泵将制备的乳液进料到喷雾干燥器中时,获得合适的结果。喷雾干燥器典型地以150℃-200℃的入口温度和60℃-90℃的出口温度操作。
更优选地,为了有效地喷雾干燥根据本发明的配制品以便以良好产率获得可自由流动的粉末,使用特定的喷雾干燥设备。因此,最优选地,使用Deck或喷雾干燥设备以使上述配制品成为具有所述粉末形态的自由流动的粉末。此类设备总体上类似并且以其温和且通用的干燥能力而闻名。此类型的喷雾干燥器设备可以最好被描述为由以下部分构成的干燥器,其中在第一阶段中,使粉末半干燥(<15%水分),并且在第二阶段中在该半干燥粉末被收集在多孔传动带上之后被干燥至所希望的最终水分含量(典型地<5%)。可以通过将上述组分合并来制备混合物,例如通过混合以及可选地搅拌以形成乳液。然后优选地将乳液喷雾干燥至水含量小于按重量计5%(优选小于按重量计4%)。然后收集如此形成的微粒材料(即粉末)。组合物优选地在惰性气体(例如,氮气)的存在下包装并且在使用之前有利地储存在低于室温的温度下,例如约5℃至约10℃。
取决于食物产品的性质,根据本发明的粉末组合物可以优选地以按重量计0.01%至20%、优选按重量计0.01%至10%、更优选按重量计0.01%至5%的量掺入食物产品中。例如,烘焙产品中可以耐受相对大量的组合物,而某些饮料则需要较少的量。合适的食物产品包括例如食品补充剂、健康零食、糖果或食品棒、运动营养产品、医疗营养产品、谷类食品、烘焙产品(例如面包、饼干或曲奇、零食棒)、基于油的产品(例如,涂抹酱、沙拉调味品)、乳制品(例如,奶、可复原的奶制品、酸奶、冰淇淋)、配方奶粉(喂养婴幼儿的液体或可复原的粉末)以及(非乳)饮料如果汁。
在优选的实施例中,将根据本发明的粉末组合物掺入配方奶粉中。配方奶粉选自下组:婴儿配方奶粉、较大婴儿配方奶粉和成长型配方奶粉。可以通过将根据本发明的粉末组合物与蛋白质源(例如乳清蛋白)、另外的脂质源、另外的碳水化合物源、维生素、和矿物质和/或牛乳组合来制备营养组合物。
所述额外的脂质源可以是适合在配方奶粉中使用的任何脂质或脂肪。优选的脂肪源包括(无水)乳脂、菜籽油、椰子油、棕榈仁油、大豆油、棕榈油精、棕榈油、(高油酸)向日葵油、(高油酸)红花油及其组合。
优选地存在于配方奶粉中的维生素和矿物质的实例为维生素A、维生素B1、维生素B2、维生素B6、维生素B12、维生素E、维生素K、维生素C、维生素D、叶酸、肌醇、烟酸、生物素、泛酸、胆碱、钙、磷、碘、铁、镁、铜、锌、锰、氯化物、钾、钠、硒、铬、钼、牛磺酸和L-肉碱。矿物质通常以盐的形式添加。
优选地存在于配方奶粉中的碳水化合物的实例为乳糖、不可消化的低聚糖(如低聚半乳糖(GOS)和/或低聚果糖(FOS))和人乳低聚糖(HMO)。通过本发明的粉末组合物添加部分GOS和/或HMO以及可选地乳糖;所需的任何另外的GOS、HMO和乳糖将单独添加到配方奶粉中。
如有必要,配方奶粉可以含有乳化剂和稳定剂,如大豆卵磷脂、单甘油酯和二甘油酯的柠檬酸酯等。配方奶粉还可以含有可能具有有益效果的其他物质,如乳铁蛋白、核苷酸、核苷等。
实例
实例1
将23.8kgGOS(前菲仕兰坎皮纳公司(FrieslandCampina))、8.6kg乳糖、5.3kg酪蛋白酸钠、2.1kg抗坏血酸钠、1.3kg乳清蛋白浓缩物以及0.7kg脂肪酸的单甘油酯和二甘油酯(E471)溶解在43.9kg水中。
随后将28kg DHA鱼油混合在此溶液中并均质化,形成乳液。将乳液巴氏灭菌(75℃,15秒)并且随后在设备上喷雾干燥。
通过干共混以每100g基础粉末460mg DHA的浓度将所得产物掺入婴儿配方奶粉基础粉末(前菲仕兰坎皮纳公司)中。将所得产物在20℃的温度下在氮气下储存4个月。
对比实例A
重复实例1,不同之处在于由32.4kg葡萄糖浆代替GOS和乳糖。
实例2
将实例1和对比实例A的婴儿配方奶粉基础粉末提交给6人测试小组。每个样品是通过将13g搅拌到87ml 40℃的水中制备的。小组成员对粉末给出范围为1(极度异味)至5(无异味)的分数。
对比实例A的粉末的平均分数为2.5;实例1的粉末的平均分数为3.8,说明与对比粉末相比,根据本发明的粉末在4个月的储存时间期间氧化更低。
实例3
将21.0kg2′-FL粉末(前菲仕兰坎皮纳公司)、5.5kg酪蛋白酸钠、1.7kg抗坏血酸钠、1.4kg乳清蛋白浓缩物以及0.6kg脂肪酸的单甘油酯和二甘油酯(E471)溶解在41.6kg水中。
随后将27.9kg DHA鱼油混合在此溶液中并均质化,形成乳液。将乳液巴氏灭菌(76℃,15秒)并且随后在设备上喷雾干燥。
将所得粉末在20℃下在氮气下储存15个月,并由菲仕兰坎皮纳公司内部味道小组在保质期开始和结束时进行测试。将0.5g粉末搅拌入50ml的40℃的半脱脂乳中并在0.5小时内品尝。小组成员对粉末给出范围为1(极度异味)至5(无异味)的分数。
在保质期开始时,平均分数为5.0;在15个月保质期结束时,平均分数为4.0。这些分数比含葡萄糖浆的对比产品(下文的对比实例B)的分数更佳或与其相当。
对比实例B
将31.0kg葡萄糖浆、8.0kg酪蛋白酸钠、2.6kg抗坏血酸钠、2.0kg乳清蛋白浓缩物以及0.9kg脂肪酸的单甘油酯和二甘油酯(E471)溶解在62.5kg水中。
随后将42.7kg DHA鱼油混合在此溶液中并均质化,形成乳液。将乳液巴氏灭菌(75℃,15秒)并且随后在设备上喷雾干燥。
在保质期开始时,平均分数为4.1;在15个月保质期结束时,平均分数为3.9。
Claims (14)
1.一种粉末组合物,其包含基于该粉末的总重量:
-10-80wt%的氧化敏感的食用油,
-5-30wt%的至少一种选自蛋白质和淀粉的乳化剂,以及
-5-65wt%的低聚半乳糖(GOS)和/或人乳低聚糖(HMO)。
2.根据权利要求1所述的粉末组合物,其中,该氧化敏感的食用油包含多不饱和脂肪酸、优选长链多不饱和脂肪酸、最优选花生四烯酸(ARA)和/或二十二碳六烯酸(DHA)。
3.根据权利要求1或2所述的粉末组合物,其中,该乳化剂是选自酪蛋白酸盐、乳清蛋白及其组合物的蛋白质。
4.根据前述权利要求中任一项所述的粉末组合物,其包含10-65wt%、优选20-60wt%的低聚半乳糖(GOS)。
5.根据前述权利要求中任一项所述的粉末组合物,其中,该低聚半乳糖(GOS)包含基于干重40-100wt%、更优选50-90wt%、并且最优选60-80wt%的低聚糖(DP≥3)。
6.根据前述权利要求中任一项所述的粉末组合物,其中,该低聚半乳糖(GOS)包含基于干重0-60wt%、优选0-40wt%、并且最优选0-30wt%的乳糖。
7.根据前述权利要求中任一项所述的粉末组合物,其包含10-65wt%、优选20-60wt%的一种或多种人乳低聚糖(HMO)。
8.根据前述权利要求中任一项所述的粉末组合物,其包含10-65wt%、优选20-60wt%的低聚半乳糖(GOS)与一种或多种人乳低聚糖(HMO)的组合。
9.根据前述权利要求中任一项所述的粉末组合物,其中,该HMO选自由以下组成的组:岩藻糖基化乳糖、唾液酸化乳糖、四糖及其组合,优选地选自由以下组成的组:2’-岩藻糖基乳糖(2’-FL)、3-岩藻糖基乳糖(3-FL)、3’-唾液乳糖(3’-SL)、6’-唾液乳糖(6’-SL)、乳糖-N-四糖(LNT)、乳糖-N-新四糖(LNnT)及其组合,更优选地选自2’-岩藻糖基乳糖(2’-FL)或2’-FL与一种或多种额外的HMO的组合,最优选地为2’-FL。
10.一种用于制备根据前述权利要求中任一项所述的粉末组合物的方法,该方法包括步骤(i):制备包含所述氧化敏感的食用油、所述乳化剂和所述碳水化合物源的水性乳液,以及(ii)将该乳液喷雾干燥。
11.根据权利要求1-9中任一项所述的粉末组合物在制造食物产品中的用途。
