Abstract
本发明提供了一种植物复方精油组合物,以及植物复方精油组合物在制备保湿修复化妆品中的应用。通过将奇亚籽油与抗氧化剂复配为复合奇亚籽油,大大提升了奇亚籽油的稳定性,并进一步提升了整个植物复方精油体系的稳定性。同时添加复合微量精油与皮肤调理剂,显著增强了植物复方精油的保湿、修复功效。并且通过合理搭配各成分及其用量,使得产品在少含或不含化学合成辅料的情况下,仍能保持产品整体优异的保质期与稳定性,降低了产品整体的刺激性,更适宜皮肤屏障受损的消费者使用。

技术领域
本发明属于日用化妆品领域,尤其涉及一种含有植物复方精油组合物的保湿修复精华油及其制备方法,以及植物复方精油组合物在制备化妆品中的应用。
背景技术
皮肤屏障是指皮肤表面的一层保护结构,它由皮脂膜、角质层、微生物层等组成,能够防止水分流失,抵御外界刺激和病菌入侵,维持皮肤的健康和稳定。皮肤屏障受损是指这一保护结构被破坏或削弱,导致皮肤出现干燥、敏感、红血丝、痘痘、色斑等问题。
导致皮肤屏障受损的原因有很多,比如过度清洁、去角质、使用不合适的护肤品、暴露在紫外线或污染环境中等。修复皮肤屏障通常通过以下途径:①停止破坏皮肤屏障,避免使用刺激性或功效性强的护肤品,选择温和、低敏的产品;②舒缓敏感、修复屏障,使用含有修复因子的护肤品,为皮肤构建滋润的修复环境;③严格防护、避免二次伤害,做好防晒工作,减少外出时间,避免接触刺激性物质。
皮肤干燥是指皮肤缺乏水分,导致外观和触感粗糙、发痒、呈片状或鳞状的现象。皮肤干燥的原因有很多,比如天气寒冷或干燥、晒伤、刺激性肥皂和洗浴过度、年龄增长、内分泌失调、某些皮肤病或药物等。皮肤干燥可以通过保湿、防晒、适当饮水、使用温和的洗涤剂、改善饮食和睡眠等方式来预防和改善。

②将称取好的西班牙鼠尾草(SALVIA HISPANICA)籽油和生育酚导入乳化锅中搅拌均匀,制得复合奇亚籽油;
③按照权利要求4-7任一项所述的保湿修复化妆品组合物的原料配比,称取剩余原料;
④将称取好的剩余原料与制备好的复合奇亚籽油,导入乳化锅中搅拌均匀;
⑤静置12小时,罐装;

第四方面,本发明涉及前述第一方面任一所述的植物复方精油组合物、或前述第二方面任一所述的保湿修复化妆品组合物、或前述第三方面任一所述的制备方法制得的保湿修复化妆品组合物在制备具有保湿、修复功效的化妆品中的应用。
本发明相对于现有技术,具有如下的优点及有益效果:
本发明通过将奇亚籽油与抗氧化剂复配为复合奇亚籽油,大大提升了奇亚籽油的稳定性,并进一步提升了整个植物复方精油体系的稳定性。同时添加复合微量精油与皮肤调理剂,显著增强了植物复方精油的保湿、修复功效。并且通过合理搭配各成分及其用量,使得产品在少含或不含化学合成辅料的情况下,仍能保持产品整体优异的保质期与稳定性,降低了产品整体的刺激性,更适宜皮肤屏障受损的消费者使用。

实施例一复合奇亚籽油的制备
按照表1所列组成配比称取相应原料,制备复合奇亚籽油MSH-1至MSH-15,各复合奇亚籽油的总重量为3g。

奇亚籽油[西班牙鼠尾草(SALVIA HISPANICA)籽油]中含有高比例的α-亚麻酸,是一种多不饱和脂肪酸,容易受到氧化的影响,导致油脂变质,对保存条件要求较高。本发明通过将奇亚籽油与合适的抗氧化剂复配使用,在不影响奇亚籽油功效与使用感受的情况下,显著提升了奇亚籽油在常规保存条件下的稳定性。
实验例1.稳定性测试
样品测定:将制备得到的MSH-1至MSH-15以及纯SH分别储存在50℃、4℃、室温(25℃)、阳光直射2个月的条件下,观察其外观。
结果表明:SH在50℃、阳光直射及室温条件下液体变浑浊。添加VE-A的MSH-2、MSH-7、MSH-12和添加VC-IP的MSH-4、MSH-9、MSH-14在50℃条件下液体变浑浊。添加VE-P的MSH-3、MSH-8、MSH-13在阳光直射条件下液体变浑浊。添加AST的MSH-5、MSH-10、MSH-15在50℃、阳光直射及室温条件下液体变浑浊。添加VE的MSH-1、MSH-6在50℃、阳光直射及室温条件下液体均澄清、稳定,未发生浑浊、变质,添加VE的MSH-11在阳光直射条件下产生少许沉淀,其他条件下液体澄清、稳定,未发生浑浊、变质。
由上述实验结果可知,SH中添加质量比为0.05和0.1的VE,复配制成MSH,能够有效防止SH发生变质。综合产品成本角度考量,选择0.05VE/99.95SH复配为MSH。
实施例二油性溶剂的制备
按照表2所列组成配比称取相应原料,制备油性溶剂,各油性溶剂的总重量为10g。

实验例2.安全性试验
样品测定:将OS-1至OS-12各样油性溶剂浸湿4层1cm2大小的纱布,置前臂屈侧,其上用较纱布稍大的透明玻璃纸覆盖,四周用橡皮膏固定,经48小时取下,查看诱发局部皮肤出现的反应,于48小时、72小时分别根据局部皮肤表现判读个基质样品的安全性结果。
测试油性溶剂OS-1至OS-12的安全性,斑贴试验皮肤不良反应分级标准如表3所示。

结果显示,经48小时和72小时试验,油性溶剂OS-5的局部皮肤与未测试皮肤相比并未出现淡红斑敏,红肿等现象,油性溶剂OS-5的分级均为0级。油性溶剂OS-4、OS-6的局部皮肤出现轻微淡红斑,分级为1级。油性溶剂OS-1至OS-3、OS-7至OS12的局部皮肤出现红斑、间或出现轻微丘疹,分级为2。
由上述实验结果可知,质量比为33:10.34的SQL/GTCC复配而成的油性溶剂,对皮肤刺激性最小,不会引皮肤过敏,安全性更好,适用于皮肤屏障受损的人群。
实施例三皮肤调理剂的制备
按照表4所列组成配比称取相应原料,制备皮肤调理剂,各皮肤调理剂的总重量为100mg。

实验例3.不同配比的皮肤调理剂对LPS诱导RAW264.7细胞释放NO抑制作用
本实验利用LPS(脂多糖)诱导RAW264.7细胞产生过敏反应,释放NO(一氧化氮)。NO抑制能力强弱可反映样品的抗炎能力大小。
实验方法:
(1)样品制备
制备1μg/mL的LPS PBS溶液作为阳性对照工作液。将上述制备好的皮肤调理剂组合物SC-1至SC-13各称取10mg,溶于100mL阳性对照工作液中,制备为10%的待测样品工作液。
(2)细胞培养
将RAW264.7细胞接种到96孔板中,每一孔为100μl,5×103cells/孔。
在37℃,5%CO2的培养箱中培养24小时。
(3)样品检测
细胞培养24小时后,移除培养基,并PBS洗涤。洗涤后每孔加入90μl培养基+10μl待测样品工作液或阳性对照工作液(保持每孔待测样品终浓度为1%,LPS终浓度为0.1μg/mL)。
样品加入之后,放入37℃,5%CO2的培养箱中培养20小时。20小时后,移取50μl的培养基到新的96孔板上,加入50μl 1%的Griessr试剂,混合后于暗处反应10分钟。酶标仪在540nm处测试样品的吸光光度值,平行重复3次。
NO抑制率%=(ALPS-ABJ)/ALPS×100%
其中:ALPS表示阳性对照样品吸光度,ASC各测试样品吸光度。
实验结果如图1所示。
结论:BSB与VE按0.2:1的比例复配为组合物时,抗炎效果最佳,且优于单种皮肤调理剂与其他皮肤调理剂组合物。
实施例四复合微量精油的制备
按照表5所列组成配比称取相应原料,制备复合微量精油。

实验例4.不同配比的复合微量精油对角质形成细胞的增殖作用
本实验通过评估不同复合微量精油对角质形成细胞的增殖作用,反映样品的修复能力大小。
实验方法:
(1)样品制备
将上述制备好的复合微量精油MEO-1至MEO11各称取4.65mg,溶于996.4mg实施例二制得的OS-5油性溶剂中,制备为精油样品溶液,再各取精油样品溶液1μL,分散于9μL细胞培养级DMSO中,制得待测样品工作液。
(2)细胞培养
将人永生化角质形成(HaCaT)细胞接种到96孔板中,每一孔为100μl,5×103cells/孔。
在37℃,5%CO2的培养箱中培养24小时。
(3)样品检测
1)加药干预:待细胞完全贴壁生长后,更换新鲜培养基,每孔100μL,以待测样品工作液进行加药处理,1μL待测样品工作液/孔,同时以1μL PBS/孔处理作为对照组,设置3个复孔,放回细胞培养箱中继续培养。
2)CCK-8测定:加药干预24小时后吸移原培养基,以含10% CCK-8的培养基进行换液处理,每孔100μL。混匀后继续孵育1小时。
3)酶标仪450nm波长条件下进行吸光度测定。细胞存活率(%)=待测样品组OD值/对照组OD值×100%。
实验结果如图2所示。
结论:MEO-8复合微量精油能够有效促进角质细胞增殖,MEO-1至MEO-7各单独精油基本无促进角质细胞增殖的功效,MEO-9至MEO-11复合微量精油促进角质细胞增殖效果不显著。
实施例五一种保湿修复化妆品的制备
①称取西班牙鼠尾草(SALVIA HISPANICA)籽油2.49875份和生育酚0.00125份;
②将西班牙鼠尾草(SALVIA HISPANICA)籽油和生育酚导入乳化锅中搅拌均匀,制得复合奇亚籽油2.5份;
③称取山茶(CAMELLIA JAPONICA)籽油30份、甜扁桃(PRUNUS AMYGDALUS DULCIS)油10份、深海两节荠(CRAMBE ABYSSINICA)籽油3.6份、亚麻荠(CAMELINA SATIVA)籽油2.5份、玻璃苣(BORAGO OFFICINALIS)籽油2.5份、向日葵(HELIANTHUS ANNUUS)籽油1份、油橄榄(OLEA EUROPAEA)果油1份、猴面包树(ADANSONIA DIGITATA)籽油1份、海檀木(XIMENIAAMERICANA)籽油1份、岩兰草(VETIVERIA ZIZANIOIDES)根油0.1份、檀香(SANTALUM ALBUM)油0.1份、茉莉花(JASMINUM SAMBAC)花油0.1份、香柠檬(CITRUS AURANTIUM BERGAMIA)果油0.1份、玫瑰(ROSA RUGOSA)花油0.03份、母菊(CHAMOMILLA RECUTITA)花油0.01份、沙棘(HIPPOPHAE RHAMNOIDES)果油0.025份、角鲨烷33份、辛酸/癸酸甘油三酯10.34份、红没药醇0.2份和生育酚1份;

⑤静置12小时,罐装。
实验例5.皮肤屏障修复功效测试
测试人群为10名脸部泛红的志愿者,每5人一组,共分为2组测试对象,分别使用本发明实施例五的产品以及某市售复方保湿修复精油。
测试区域:全脸;
使用:每天早晚使用两次。使用德国CK公司TM300测试仪分别测试第0周和持续使用至第4周、第8周后脸颊部位的经皮水份流失,简称TEWL(g/㎡h),即每小时每平方米面积散失多少克水。皮肤水份流失TEWL对评估皮肤水份保护层的功能是非常重要的参数,在国际上已经得到了广泛的认可,可以很好的反应皮肤屏障功能的完整性。标准的TEWL是在皮肤干燥无汗状态下,在环境温度24℃,环境湿度为50%的状态下测量的。
评价标准:皮肤保护层越完好,水份的含量就会越高,皮肤水份流失的TEWL的数值就越低,即皮肤屏障得到修复,结果见图3。
从以上数据可以看出,实施例五的产品与复方保湿修复精油相比,在使用一段时间后均能降低经皮水分散失值(TEWL),说明修复皮肤屏障效果明显。

④将称取好的剩余原料与制备好的复合奇亚籽油,导入乳化锅中搅拌均匀;
⑤静置12小时,罐装。
9.根据权利要求8所述的保湿修复化妆品组合物的制备方法,其特征在于,采用如下方法制备:
①称取西班牙鼠尾草(SALVIA HISPANICA)籽油2.49875份和生育酚0.00125份;
