背景技术
新型冠状病毒(SARS-CoV-2)是目前已知的RNA病毒中基因组最大的一种正链RNA病毒。研究发现,新型冠状病毒主要通过人体呼吸道黏膜系统侵染细胞,进入细胞后,病毒基因被蛋白酶切割以开始翻译和复制所需蛋白。3CL蛋白酶识别特异性的酶切位点,将多聚蛋白前体剪切为多个非结构蛋白,对病毒的生命周期至关重要,是极好的抗病毒药物靶点。许多上市的抗病毒药物针对如HIV、HCV的3CL蛋白酶。其中,HIV 3CL蛋白酶的抑制剂洛匹那韦(lopinavir)和利托那韦(ritonavir)在体外与新型冠状病毒的3CL蛋白酶有一定相互作用,但在临床测试中被证明对新型冠状病毒感染的患者没有正向作用。因此,特异性针对新型冠状病毒的3CL蛋白酶的强抑制剂显得尤为重要。

为了简明起见,后文所述“式(I)化合物”或“本申请的化合物”也可以涵盖式(I)化合物的任意同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物。

本申请的药物组合物的剂型可以是液体剂型、固体剂型或半固体剂型。液体剂型可以是溶液剂(包括真溶液和胶体溶液)、乳剂(包括o/w型、w/o型和复乳)、混悬剂、注射剂(包括水针剂、粉针剂和输液)、滴眼剂、滴鼻剂、洗剂和搽剂等;固体剂型可以是片剂(包括普通片、肠溶片、含片、分散片、咀嚼片、泡腾片、口腔崩解片)、胶囊(包括硬胶囊、软胶囊、肠溶胶囊)、颗粒剂、散剂、丸剂、栓剂、膜剂、贴片、气雾剂、喷雾剂等;半固体剂型可以是软膏剂、凝胶剂、糊剂等。本申请的药物组合物可以制成普通制剂、也制成是缓释制剂、控释制剂、靶向制剂及各种微粒给药系统。
在一些实施方式中,所述药物组合物的剂型选自片剂、颗粒剂、散剂、糖浆剂、吸入剂和注射剂。

在第三方面,本申请提供了式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物在制备用于在有此需要的受试者中治疗或预防冠状病毒感染或由冠状病毒引起的疾病或症状的药物中的用途。式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物可以用于在有此需要的受试者中治疗或预防冠状病毒感染或由冠状病毒引起的疾病或症状。
本申请使用的术语“受试者”是指可潜在地从用式(I)化合物进行的治疗中受益的任何人类或非人类生物体。示例性受试者包括任何年龄的人类或哺乳动物。优选地,所述受试者是人。
本文使用的术语“治疗”包括在哺乳动物、尤其人类中治疗疾病或症状且包括:(a)抑制感染、疾病或症状,即遏制或延缓感染、疾病或症状的发展;(b)缓解感染、疾病或症状,即引起疾病或症状的消退,和/或(c)感染、疾病或症状的治愈。

本申请的发明人发现,本申请的化合物能够实现对冠状病毒感染的抑制,例如能够作为可逆的共价小分子抑制剂针对新冠病毒的3CL蛋白酶作用(即3CL蛋白酶抑制剂),从而抑制冠状病毒的病毒复制。因此,本申请的化合物可以用于预防或治疗冠状病毒感染或由冠状病毒引起的疾病或症状。
在一些实施方式中,所述冠状病毒选自严重急性呼吸综合征冠状病毒(SARS-CoV)、新型冠状病毒(SARS-CoV-2)、中东呼吸综合征冠状病毒(MERS-CoV)、冠状病毒OC43(HCoV-OC43)、鼠肝炎冠状病毒(MHV)和与以上任一种冠状病毒的同源性大于85%且具备病毒活性的冠状病毒。在一些实施方式中,所述冠状病毒是新型冠状病毒(SARS-CoV-2)。
在一些实施方式中,由冠状病毒引起的疾病或症状包括由所述病毒引起的呼吸系统感染、急性呼吸综合征(SARS)、肺炎(包括重症肺炎)、肠胃炎(包括急性肠胃炎)、咳嗽、发热、寒战、呕吐、头痛、畏寒、呼吸急促、细胞因子风暴等。
在第四方面,本申请提供了一种治疗或预防冠状病毒感染或由冠状病毒引起的疾病或症状的方法,所述方法包括向有此需要的受试者施用治疗有效量的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物。
在一些实施方式中,本发明化合物可以通过口服、肠胃外、静脉注射、肌肉注射、皮下注射、鼻腔、口腔粘膜、眼、经肺、经呼吸道、经阴道、经直肠、腹膜内、病灶内、病灶周围等途径施用。
“治疗有效量”是指本申请化合物当单独或组合给药时有效治疗或预防冠状病毒感染或由冠状病毒引起的疾病或症状的量。
具体施用剂量将取决于施用途径、疾病的严重程度、患者的年龄和体重,以及主治医师在确定最适合特定患者的个体方案和剂量水平时通常考虑的其他因素。例如,本申请的化合物的日剂量可以特别地为0.001-150mg/kg体重(例如0.1mg/kg体重、1mg/kg体重、10mg/kg体重或100mg/kg体重等)。

本申请式(I)化合物可以用有机合成领域的技术人员所熟悉的多种方法合成。以下具体实施例中给出了一些示例性的式(I)化合物的合成方法,这些方法是合成化学领域所公知的。显然,参照本专利中的示例性方案,本领域技术人员可以适当调整反应物、反应条件和保护基团而容易地设计其他式(I)化合物的合成路线。
下面进一步结合实施例来阐述本发明;但这些实施例并不限制本发明的范围。除非另有声明,各实施例中所用的所有反应物均从商业途径获得;合成实验和产物分析检测中所用仪器设备等均为有机合成中通常使用的常规仪器和设备。
中间体的合成路线
1)化合物5的合成操作步骤
将化合物5_1(50g,353mmol,1eq)溶于NH3/MeOH(7M,250mL)中,在25℃下加入NH4Cl(56.7g,1.06mol,3eq),室温下搅拌反应1小时。然后将TMSCN(70.1g,706mmol,88.4mL,2eq)加入到反应混合物中,室温下搅拌反应16小时,反应完全。向反应液中加入冰水(1L),用EtOAc(300mL*3)萃取,有机相合并,用300mL饱和氯化钠洗涤。有机相用无水硫酸钠干燥,减压旋干,通过柱色谱法(SiO2,PE/EA=20/1→0/1)纯化(TLC,PE:EA=0/1,Rf=0.43)得到目标化合物5(18g,107mmol,30.4%收率)为黄色固体。ESI-MS:[M+H]+,168.0。
2)化合物3的合成操作步骤
将化合物1(40.0g,194mmol,1eq)和化合物2(41.9g,194mmol,1eq)溶于MeCN(720mL)和DMF(80mL)中,冷却到0℃。加入HATU(81.3g,214mmol,1.1eq)和DIEA(75.4g,583mmol,102mL,3eq)。室温下搅拌反应16小时,反应完全。向反应也中加入冰水(1L),用EtOAc(300mL*3)萃取,有机相合并,用300mL饱和氯化钠洗涤。有机相用无水硫酸钠干燥,减压旋干,通过柱色谱法(SiO2,PE/EA=50/1→10/1)纯化(TLC,PE/EA=1/1,Rf=0.25)得到目标化合物3(47.0g,66.0%)为无色油状物。ESI-MS:[M+H]+,367.1。
3)化合物4的合成操作步骤
将化合物3(47.0g,128mmol,1eq)溶于THF(384mL)中,再加入甲醇(128mL),冷却到0℃,搅拌下滴加LiOH·H2O(10.8g,257mmol,2eq)的水溶液(128mL)。然后室温下搅拌反应2小时。反应也中加入H2O(300mL),用盐酸(1M)调节水相pH=3。用EtOAc(100mL*5)萃取水相,有机相合并,用150mL饱和氯化钠洗涤,有机相用无水硫酸镁干燥,过滤并减压浓缩化合物4(39.9g,88.3%)的粗产品为黄色油状物,直接用于下一步。ESI-MS:[M+H]+,353.0。
4)化合物6的合成操作步骤
将化合物4(8.70g,24.7mmol,1eq)和化合物5(4.96g,29.6mmol,1.2eq)溶于MeCN(270mL)和DMF(20mL)中。冷却到0℃,依次加入HAUT(10.3g,27.2mmol,1.1eq)和DIEA(9.57g,74.1mmol,12.9mL,3eq)。室温下搅拌反应过夜。将反应混合物倒入H2O(400mL)中,用EtOAc(60mL*3)提取。在减压下浓缩,通过柱色谱法(SiO2,PE/EA=1/1→1/3)纯化(TLC,PE/EA=1/3,Rf=0.55)得到化合物6(11.2g,90.4%)为黄色油状物。ESI-MS:[M+H]+,502.2。
5)化合物8的合成操作步骤
将化合物6(11.0g,21.9mmol,1eq)和化合物7(5.06g,32.9mmol,5.58mL,1.5eq)以及Na2CO3(6.97g,65.7mmol,3eq)溶于1,4-二氧六环(220mL)中,加入水(44mL)。再加入Pd(OAc)2(492mg,2.19mmol,0.1eq)和RuPhos(1.02g,2.19mmol,0.2eq)。氮气保护下,在80℃下搅拌反应16小时,TLC检测反应完成。将反应混合物倒入40mL H2O中,用EtOAc(100mL*3)提取。减压浓缩,通过柱色谱法(SiO2,PE/EtOAc=1/1至1/3)纯化(PE/EA=1/3,Rf=0.55)得到目标产物8为黄色油状物(6.0g,55.5%收率)。ESI-MS:[M+H]+,494.2。
实施例1:化合物GDI15-532的制备
反应瓶中加入化合物8(0.26g,527μmol,1eq)和Hoveyda-Grubbs II(33.0mg,52.7μmol,0.1eq),在氮气保护下加入除氧溶剂1,2-二氯乙烷(50mL)和Ti(OiPr)4(150mg,527μmol,155μL,1eq)。氮气保护下在80℃下搅拌反应16小时。减压浓缩,得到残留物通过制备HPLC(中性条件)纯化,再经过HPLC手性拆分得到GDI15-532为淡黄色固体(43.0mg,49.4μmol,17.5%收率)。ESI-MS:[M+H]+,466.3。

实施例2:化合物GDI15-566的制备
将化合物GDI15-532(0.3g,644μmol,1eq)溶于DMSO(6mL)中,在0℃下加入K2CO3(178mg,1.29mmol,2eq),在0℃下缓慢滴加H2O2(292mg,2.58mmol,248μL,30%纯度,4eq)。在25℃下搅拌反应16小时。TLC和LC-MS检测反应完成。在0℃下将5mL硫代硫酸钠水溶液缓慢滴加入反应液中搅拌15min,用EtOAc(1mL*3)萃取水相,有机相合并,用10mL饱和氯化钠洗涤,有机相用无水硫酸镁干燥,过滤并减压浓缩得到目标产物22(0.21g,434μmol,67.4%收率)为黄色油状物。ESI-MS:[M+H]+,484.1。
将Pd/C(0.1g,331μmol,10%纯度,1eq)加入MeOH(16mL)中,在25℃下加入化合物22(0.16g,331μmol,1eq)。H2置换三次,在50℃压力15psi下搅拌反应16小时。TLC和LC-MS检测反应完成。硅藻土过滤反应溶液,在减压下浓缩滤液得到目标产物23(0.13g,268μmol,80.9%收率)为黄色油状物。ESI-MS:[M+H]+,486.2。
将化合物23(80mg,165μmol,1eq)溶于DCM(1.6mL),在0℃下依次加入TEA(100mg,988μmol,138μL,6eq)和TFAA(104mg,494μmol,68.8μL,3eq)。在0℃下搅拌反应2小时。LC-MS检测反应完成。将反应混合物减压浓缩,通过高效液相色谱纯化(柱:Waters Xbridge BEHC18 100*30mm*10μm;移动向:[水(NH4HCO3)-ACN];B%:45%-70%,8min)得到GDI15-566(11.3mg,24.3μmol,14.7%收率)为黄色油状物。ESI-MS:[M+H]+,468.2。

将化合物20(300mg,501μmol,1eq,)溶于DCM(6mL),在0℃下加入TFA(0.6mL),在25℃下搅拌反应1小时,LCMS检测反应完成。将反应混合物减压浓缩,分别通过高效液相色谱纯化(柱:Phenomenex C18 80*40mm*3μm;流动相:[水(NH4HCO3)-ACN];B%:15%-45%,8min)和SFC拆分(柱:DAICEL CHIRALPAK AD(250mm*30mm,10μm);流动相:[水(0.1%氨水)-IPA];B%:40%-40%,11min)得到目标产物GDI15-621(33.3mg,66.7μmol,13.3%收率)为白色固体。ESI-MS:[M+H]+,499.2。

实施例24:化合物GDI15-622的制备
将化合物GDI15-575(100mg,273μmol,1eq)和化合物21(48.8mg,301μmol,1.1eq)溶于MeCN(1.8mL)和DMF(0.2mL)中,在0℃下依次加入HATU(114mg,301μmol,1.1eq)和DIEA(106mg,820μmol,143μL,3eq),在25℃下搅拌反应16小时。LCMS检测反应完成。将反应混合物减压浓缩,分别通过高效液相色谱纯化(柱:Waters Xbridge BEH C18 100*30mm*10μm;流动相:[水(NH4HCO3)-ACN];B%:30%-60%,8min)和SFC拆分(柱:DAICEL CHIRALCEL OD(250mm*30mm,10μm);流动相:[Neu-MeOH];B%:10%-35%,16min)得到目标产物GDI15-622(17.1mg,33.5μmol,12.2%收率)为白色固体。ESI-MS:[M+H]+,510.2。

测试例1:化合物的活性测定
SARS-CoV-2/HeLa-ACE2测试
化合物转移到384孔板(Greiner,Part.No.781090-2B),然后加入HeLa-ACE2细胞(细胞密度:250个细胞/μL,培养液:MEM含2% FBS)。植入细胞的板子转移到BSL3实验室。SARS-CoV-2(USA-WA1/2020 Vero E6细胞增殖)稀释到MOI:0.75到1,达到~30-60%细胞感染。测试板在37℃,5%CO2细胞培养箱中孵育48小时后,加入4%多聚甲醛固定细胞。人源多克隆血清抗体作为一抗,山羊抗人H+L缀合Alexa 488(Thermo Fisher ScientificA11013)作为二抗。DAPI(Thermo Fisher Scientific D1306)DNA染色。
未感染细胞毒性测试
化合物转移到1536孔板(Corning No.9006BC),然后加入HeLa-ACE2细胞(细胞密度:120个细胞/μL,培养液:MEM含2% FBS)。测试板在37℃5%CO2细胞培养箱中孵育48h后,进行细胞活力测定。2μL 50% Cell-Titer Glo(Promega No G7573)水中稀释加入细胞测试板,然后用EnVision Plate Reader(Perkin Elmer)读取数值。
酶活性测试
使用200nM重组SARS-CoV-2主蛋白酶和15μM荧光底物(Dabcyl-TSAVL QSGFRK-Glu(EDANS);Genscript)进行抑制试验。测定缓冲液由50mM Tris-HCl、pH 7.3、1mM EDTA组成。SARS-CoV-2 3CLpro溶解在25μL测定缓冲液中,与不同浓度的化合物混合。将混合物在37℃下孵育30分钟。然后加入溶解在25μL测定缓冲液中的底物。使用M5多模式酶标仪在37℃测定荧光(Ex:350nm/Em:490nm,1次/分钟),持续10分钟。与具有最低浓度的反应相比,再与不同浓度的化合物反应的第6分钟的RFU用于生成IC50曲线。对于每种化合物,测定对SARS-CoV-2 3CLpro的IC50值时使用12个化合物浓度。实验数据通过GraphPadPrism软件进行分析。

1.式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,
其中,
P1选自C1-C4亚烷基,其中任选地所述C1-C4亚烷基中的一个或多个氢原子被卤素或C1-C3烷基取代,并且任选地C1-C4亚烷基中的一个或多个碳原子被选自N、O或S的杂原子取代,使得P1与其所连接的两个碳原子形成相应的环烷基或杂环烷基;
P2选自氢或-X-R2,其中X选自-C(O)-、-S(O)2-、-C(O)O-或-OC(O)-;
P3选自C1-C4亚烷基或C2-C4亚烯基,其中任选地所述C1-C4亚烷基或C2-C4亚烯基中的一个或多个氢原子被卤素或C1-C3烷基取代,并且任选地C1-C4亚烷基中的一个或多个碳原子被选自N、O或S的杂原子取代;
R1选自氢、氘、卤素、羟基、氰基、氨基、硝基、C1-C3烷基、C1-C3卤代烷基或C1-C3烷氧基;并且
R2选自C1-C6烷基、C1-C6卤代烷基、C1-C6烷氧基、C1-C6卤代烷氧基、C3-C6环烷基、苯甲基、C6-C10芳基或五元至十元杂芳基,其中任选地所述苯甲基、C6-C10芳基或五元至十元杂芳基中的一个或多个氢原子被卤素、羟基、氰基、氨基、硝基、C1-C3烷基、C1-C3烷氧基、C1-C3卤代烷基、C1-C3卤代烷氧基或C3-C4环烷基取代。
2.根据权利要求1所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,其中,
P1选自C1-C4亚烷基,其中任选地所述C1-C4亚烷基中的1、2或3个氢原子被卤素或C1-C3烷基取代,使得P1与其所连接的两个碳原子形成相应的环烷基;
P2选自氢或-X-R2,其中X选自-C(O)-、-S(O)2-、-C(O)O-或-OC(O)-;
P3选自C1-C4亚烷基或C2-C4亚烯基,其中任选地所述C1-C4亚烷基或C2-C4亚烯基中的1、2或3个氢原子被卤素或C1-C3烷基取代,并且任选地C1-C4亚烷基中的一个碳原子被选自N或O的杂原子取代;
R1选自氢、氘、卤素、氰基、硝基、C1-C3烷基、C1-C3卤代烷基或C1-C3烷氧基;并且
R2选自C1-C6烷基、C1-C6卤代烷基、C1-C6烷氧基、C3-C6环烷基、苯甲基、C6-C10芳基或五元至十元杂芳基,其中任选地所述苯甲基、C6-C10芳基或五元至十元杂芳基中的一个或多个氢原子被卤素、羟基、氨基、C1-C3烷基、C1-C3烷氧基、C1-C3卤代烷基、C1-C3卤代烷氧基或C3-C4环烷基取代。
3.根据权利要求2所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,其中,
P1选自
P2选自氢或-X-R2,其中X选自
P3选自其中Y选自O、NH或NMe;
R1选自氢、氘、卤素、-CN、-NO2、C1-C3烷基、C1-C3卤代烷基或C1-C3烷氧基;并且
R2选自C1-C6烷基、C1-C6卤代烷基、C1-C6烷氧基、C3-C6环烷基、苯甲基、苯基、咪唑基、吡唑基、吡啶基、嘧啶基、吡咯基、吡嗪基、哒嗪基、吲哚基或苯并吡咯基,其中任选地所述苯甲基、C6-C10芳基或五元至十元杂芳基中的一个或多个氢原子被卤素、-OH、-NH2、C1-C3烷基、C1-C3烷氧基、C1-C3卤代烷基、C1-C3卤代烷氧基或C3-C4环烷基取代。
4.根据权利要求1-3中任一项所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,其中,
所述烷基选自甲基、乙基、丙基、异丙基或叔丁基;
所述卤素选自氟、氯或溴;
所述卤代烷基选自一氟甲基、二氟甲基或三氟甲基;
所述烷氧基选自甲氧基、乙氧基、丙氧基或异丙氧基;和/或
所述卤代烷氧基选自三氟甲氧基。
5.根据权利要求1-3中任一项所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,其具有以下结构中的任一种:
6.一种药物组合物,其包含根据权利要求1-5中任一项所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物,以及药学上可接受的载体。
7.根据权利要求6所述的药物组合物,其剂型选自片剂、颗粒剂、散剂、糖浆剂、吸入剂和注射剂。
8.根据权利要求1-5中任一项所述的式(I)化合物或其同位素标记化合物、或其光学异构体、几何异构体、互变异构体或异构体混合物、或其药学上可接受的盐、或其前体药、或其代谢物在制备药物中的用途,所述药物用于在有此需要的受试者中治疗或预防冠状病毒感染或由冠状病毒引起的疾病或症状。
9.根据权利要求8所述的用途,其中所述冠状病毒选自严重急性呼吸综合征冠状病毒(SARS-CoV)、新型冠状病毒(SARS-CoV-2)、中东呼吸综合征冠状病毒(MERS-CoV)、冠状病毒OC43(HCoV-OC43)、鼠肝炎冠状病毒(MHV)和与以上任一种冠状病毒的同源性大于85%且具备病毒活性的冠状病毒。
10.根据权利要求8或9所述的用途,其中所述由冠状病毒引起的疾病或症状选自以下中的一种或更多种:呼吸系统感染、急性呼吸综合征(SARS)、肺炎(包括重症肺炎)、肠胃炎(包括急性肠胃炎)、咳嗽、发热、寒战、呕吐、头痛、畏寒、呼吸急促和细胞因子风暴。
