The preparation method of this example is the same as that of example 1, and the preparation method of example 1 is followed to prepare the Chinese medicinal preparation of this example.
Example 3
The traditional Chinese medicine composition for dispelling the effects of alcohol in the embodiment comprises the following components in parts by mass: 60g of hovenia dulcis thunb, 40g of mung bean, 20g of sea buckthorn, 20g of dried orange peel, 30g of mulberry, 5g of turmeric and 10g of hawthorn.
The preparation method of this example is the same as that of example 1, and the preparation method of example 1 is followed to prepare the Chinese medicinal preparation of this example.
Test case
1. Experimental materials
1.1 Laboratory animals
SPF KM mice, male, 18.0 g-22.0 g in weight, supplied by the university of Shenyang medical science animal center, qualification number SCXK (Liao) 2020-0001, were fed water freely, and were fed adaptively for three days before the experiment.
1.2 Experimental herbs and reagents
The fruits of hovenia dulcis thunb, mung bean, sea buckthorn, dried orange peel, mulberry, turmeric and hawthorn used in the experiments were identified as the fruits of hovenia dulcis thunb (Hovenia acerba lindl.) of the genus Rhamnaceae, the seeds of mung bean (VIGNA RADIATA (lin.) Wilczek) of the genus vigna of the family Leguminosae, the dried ripe fruits of sea buckthorn (Hippophae rhamnoides l.) of the genus Elaeagnaceae, the dried ripe fruit peel of citrus (Citrus reticulata Blanco) of the family Rutaceae and its cultivated variety, the ripe ears of mulberry (Morus alba linn.) of the family Moraceae, the dried rhizome of turmeric (Curcuma Longa l.) of the genus Curcuma of the family Zingiberaceae and the dried ripe fruits of hawthorn (Crataegus pinnatifida bge.) of the genus Rosaceae, respectively.
Sea Wang Jinzun pieces (Shenzhen sea Wang Jiankang Co., ltd.); red star Erguotou white spirit (56%vol, beijing red star Co., ltd.).
2. Experimental part
2.1 Sobering up experiment
Healthy male mice were divided into 50 randomly into 5 groups: model group, positive control group, example 1 group, example 2 group, example 3 group, 10 each. After three days of adaptive feeding, the patients are fasted for 12 hours before the experiment, the model group is filled with gastric physiological saline according to the dosage of 10ml/kg, the positive control group is filled with gastric sea Wang Jinzun oral liquid according to the dosage of 10ml/kg, and the group of the invention is used for changing the dosage of gastric mixed solution according to the group of the invention, namely 10ml/kg of the mixed solution according to the group of the embodiment 1, the embodiment 2 and the embodiment 3. After 30min, 15ml/kg of a commercially available 56 DEG Erguotou white wine is filled. The mice were recorded for alcohol latency and alcohol time.
The drunk latency period is the time from the time of filling the stomach and the erguotous ends to the time of completely eliminating the regular reflection; the sober-up time is the time from complete disappearance of the specular reflection to complete recovery of the specular reflection. The eversion and the reflection are that the mouse keeps the supine position for more than 30s, namely the eversion and the reflection disappear, otherwise the eversion and the reflection recover.
Before the experiment, the mice are weighed and marked, and the stomach-filling administration dosage and the wine administration dosage of the mice are set in combination with the pre-experiment.
2.2 Effects of the invention on serum ethanol concentration in mice
Healthy male mice were randomized into 6 groups: blank, model, positive control, example 1, example 2, example 3, 10 per group. After the three days of adaptive feeding, the blank group and the model group are fasted without water forbidding for 12 hours before the experiment, the CMC-Na solution with the concentration of 0.5% is filled into the stomach according to the dosage of 10mL/kg, the positive control group is filled with the CMC-Na solution with the concentration of 10mL/kg (1 g/mL), the mixed gastric solution is respectively filled into the groups of examples 1-3 according to the dosage of 10mL/kg (1.5, 3.0 and 3.7g/mL according to the traditional Chinese medicine composition), after 30 minutes, the two pot heads of the stomach are filled into the groups except the normal saline of the blank group, after the two pot heads are filled into the stomach for 2 hours, the eyeballs of mice are picked up, the blood is taken into a centrifuge tube with the concentration of 1.5mL, serum is separated, and the serum is stored in a refrigerator at the temperature of minus 80 ℃ for refrigeration. The ethanol concentration in serum was determined by gas chromatography as follows:
a) Preparation of standard serum samples: taking 50 mu L of blank serum, placing the blank serum into 7 centrifuge tubes (numbered 0, 1,2,3, 4, 5 and 6), taking a blank (without ethanol) sample tube as a tube number 0, and adding 10 mu L of acetonitrile; the number 1-6 tube is a standard sample tube, and 10 mu L of series acetonitrile solution with ethanol solubility is added to prepare series standard serum samples with the ethanol concentration of 0.2, 0.5, 1,2, 4 and 10mg/mL respectively.
B) Preparation of internal standard solution: accurately weighing 1g of acetone, placing the acetone into a 100mL volumetric flask, adding acetonitrile to fix the volume, and preparing an internal standard stock solution containing 10mg/mL of acetone; precisely sucking 2mL of internal standard stock solution, placing the internal standard stock solution in a 10mL volumetric flask, and fixing the volume by acetonitrile to prepare 2mg/mL of internal standard solution.
C) Sample pretreatment: adding 10 mu L of internal standard solution into each sample tube, mixing for 1min by vortex, adding 150 mu L of acetonitrile, mixing by vortex, centrifuging for 5min at the rotating speed of 12000 r.min -1, taking supernatant into a clean centrifuge tube, adding 180 mu L of acetonitrile to dilute the supernatant, mixing by vortex for 10s, filtering by a filter membrane, and taking 1 mu L of sample.
D) Gas chromatography conditions: agilent DB-624 chromatographic column (30 m×250 μm×1.4 μm), sample inlet temperature 230 ℃, flame ionization detector (flame ionization detector, FID) temperature 250 ℃, programmed temperature, initial temperature 50 ℃, holding 5min,60 ℃/min to 220 ℃, holding 1min. Column flow 1mL/min, split sample introduction, split ratio 50:1, sample injection amount is 1 mu L.
E) Drawing a standard curve: taking a standard serum sample, operating according to a sample pretreatment method, carrying out sample injection measurement according to a gas chromatography condition, recording the peak area of ethanol and an internal standard, taking the ratio of the peak area of ethanol and the internal standard as an ordinate and the concentration of ethanol as an abscissa, drawing a standard curve, and calculating a regression equation as y= 2.0905x-0.0777 (r= 0.9988). The concentration of the serum ethanol has good linear relationship within the range of 0.2 mg/mL-10 mg/mL.
F) Determination of serum samples: taking a serum sample of an experimental mouse, adding 10 mu L of acetonitrile, operating according to a sample pretreatment method, carrying out sample injection measurement according to a gas chromatography condition, recording the peak area of ethanol and an internal standard, calculating the concentration of ethanol in the serum sample by a standard curve method according to the ratio of the peak area of ethanol to the internal standard, carrying out statistical treatment, and comparing the differences among groups.
2.4 Statistical treatment
The SPSS21.0 statistical software is adopted for analysis and treatment, and the data are all obtainedThe comparison between groups adopts single-factor analysis of variance, and the difference is significant and statistically significant as represented by P < 0.05.
3. Experimental results
3.1 Effects of the invention on the drunk latency and sober-up time of mice, and experimental results are shown in Table 1.

As can be seen from the results in Table 1, compared with the model group, the high, medium and low dose groups of the traditional Chinese medicine composition can remarkably shorten the drunk time (p < 0.01) of the mice with acute alcoholism and can prolong the drunk latency (p < 0.01) of the mice. The effects of the high, medium and low dose groups of the traditional Chinese medicine composition are superior to those of a sea Wang Jinzun control group in the aspects of prolonging the drunk latency period and shortening the drunk time.
3.2 Effects of the invention on ethanol concentration in mouse blood, serum ethanol concentration measurement results are shown in Table 2.

After the ethanol enters the human body, most of the ethanol is absorbed by the intestinal tract except for a small part of the ethanol is absorbed by the stomach, the ethanol is completely absorbed into the blood within 30-90 min, the drunk appearance has a direct relation with the concentration of the ethanol in the blood, and the higher the concentration of the ethanol, the more serious the drunk. Therefore, examining the concentration of ethanol in blood is an important index for evaluating the anti-hangover effect. The effect of the application on the ethanol concentration of the mouse serum is known by measuring the ethanol concentration in the mouse serum. As can be seen from the results in table 2, the ethanol concentration of the model group was significantly increased (P < 0.01) compared to the blank group, indicating that the model replication was successful. Compared with a model group, in the aspect of reducing the ethanol concentration, the high, medium and low dosage groups and the positive group of the traditional Chinese medicine composition can obviously reduce the ethanol concentration, the difference has significance (P is less than 0.01), and the effects of the high, medium and low dosage groups of the traditional Chinese medicine composition are better than those of a sea Wang Jinzun control group.
4. The weight proportion of the traditional Chinese medicine composition and the research of the modified formula of the traditional Chinese medicine composition
The test example carries out weight proportion research and addition and subtraction formula research on the traditional Chinese medicine compound. The formulation of the test drug is shown in Table 3 below.

The experimental method is as follows: healthy male mice were randomly divided into 10 groups of 100: model group, positive control group and formula 1-8, 10 in each group. After three days of adaptive feeding, the patients are fasted for 12 hours before the experiment, the model group is irrigated with gastric physiological saline according to the dosage of 10ml/kg, the positive control group is irrigated with gastric sea Wang Jinzun solution according to the dosage of 10ml/kg, and the formula of the invention is 10ml/kg of 1-8 groups of gastric irrigation mixed solution. After 30min, 15ml/kg of a commercially available 56 DEG Erguotou white wine is filled. The mice were recorded for alcohol latency and alcohol time.
The mice were weighed and labeled prior to the experiment, and the mice were set with the stomach-lavage dose and the tank wine dose in combination with the pre-experiment, and the experimental results are shown in table 4.

As can be seen from the results in table 3, the formula after subtracting certain medicinal flavors on the basis of the Chinese herbal compound of the present application, such as the formula 6 minus mulberries, the formula 7 minus mung beans and the formula 8 minus turmeric, although the drunk latency can be prolonged and the drunk time shortened to some extent (p < 0.05 compared with the model group), is not as effective as the original formula of the present application, in particular, unlike the formulas 1 to 3 (p is less than 0.01 compared with the model group) in the original formula. This illustrates that there may be some synergy between the various flavors of the present application. Particularly, under the weight proportion of the compositions 1-3, the traditional Chinese medicine composition has the best effect in the aspects of prolonging the drunk latency period and shortening the drunk time, and has extremely obvious difference (p is less than 0.01) compared with a model group.
In conclusion, the invention combines with practical experiments, preferably selects the specific composition and the proportion of the traditional Chinese medicine composition for dispelling the effects of alcohol, and uses seven medicinal materials with homology of medicine and food for combination and compatibility, so that the traditional Chinese medicine composition for dispelling the effects of alcohol can promote in vivo ethanol decomposition through various ways, and lighten the damage of alcohol to human bodies, in particular to liver, cardiovascular and cerebrovascular diseases and spleen and stomach. The animal experiment result shows that the traditional Chinese medicine composition for dispelling the effects of alcohol can increase the tolerance time of mice to alcohol, can obviously shorten the drunk time of experimental mice, has excellent anti-alcoholic effect on drunk mice, shows that the traditional Chinese medicine composition for dispelling the effects of alcohol can effectively avoid toxic action of acetaldehyde on human bodies, realizes the effects of preventing alcohol, rapidly dispelling the effects of alcohol, protecting heart and cerebral vessels, strengthening spleen and stomach and protecting liver, realizes the strengthening of two functions of dispelling the effects of alcohol and protecting viscera, has the advantages of less number of medicine tastes, low cost, convenient administration, no obvious toxic or side effect, convenient daily carrying, suitability for timely administration of people with the requirements of dispelling the effects of alcohol, and the like, and has extremely broad market prospect.

3. The traditional Chinese medicine composition for dispelling the effects of alcohol according to claim 1, which is characterized by comprising the following components in mass:
30g of the hovenia dulcis thunb, 20g of the mung bean, 20g of the sea buckthorn, 20g of the dried orange peel, 30g of the mulberry, 10g of the turmeric and 20g of the hawthorn.

Decocting semen Phaseoli Radiati, fructus Hippophae, pericarpium Citri Tangerinae, mori fructus, curcuma rhizome and fructus crataegi with water, filtering the extractive solution, and concentrating to obtain water decoction fluid extract;
Reflux-extracting semen Hoveniae with ethanol, filtering the extractive solution, recovering ethanol, and concentrating to obtain ethanol extract;
Mixing the water decoction fluid extract and the alcohol extract fluid extract to obtain the traditional Chinese medicine composition for dispelling the effects of alcohol.
